    Notes: Abstract The polymerase chain reaction was used to amplify six small variable number of tandem repeat loci in two reactions (D19S20 co-amplifying with D17S5 and D1S80; D17S766 co-amplifying with D16S83 and D17S24). When coupled with fluorescent detection of the products, this provides a rapid, highly discriminating automated test. Preferential amplification of small alleles, leading to ‘allelic dropout’ was found to occur in D19S20 and D16S83. Population databases are presented for Caucasians and Afro-Caribbeans at loci D19S20, D16S83 and D17S24, and for Asians at D19S20.

    Notes: Abstract. The centromere/kinetochore complex is a chromosomal assembly that mediates chromosome motility and mitotic regulation by interacting with microtubules of the mitotic spindle apparatus. Centromere protein A (CENP-A) is a histone H3 homolog that is concentrated in the chromatin of the inner kinetochore plate of human chromosomes. To identify DNA sequences associated with the inner kinetochore plate, we used anticentromere autoantibodies to immunoprecipitate CENP-A associated chromatin selectively from Indian muntjac fibroblasts. DNA was cloned from immunoprecipitated CENP-A- associated chromatin and characterized by DNA sequence and hybridization analyses. A novel centromeric satellite DNA sequence was identified and shown by fluorescence in situ hybridization analysis to be present at all centromeres of the Indian muntjac. This satellite DNA constitutes a 972 bp monomer repeat and shows partial homology with satellite II DNA of the white-tailed deer. Southern blot analysis of muntjac genomic DNA suggests that this satellite DNA is present in repetitive tandem arrays and contains complex internal arrangements. In conjunction with previous work showing the association of CENP-A with human α-satellite DNA, we conclude that the mammalian inner kinetochore plate contains a unique form of chromatin that contains CENP-A in association with complex satellite DNA.

    Notes: The seepage meter and the mini-piezometer were utilized in an attempt to evaluate ground-water reservoir interactions over a 12-month period at Echo Bay in Lake Mead. In conjunction with these techniques three standard piezometers, refraction seismic data, and water chemistry data were utilized to interpret seepage device results. During a four-month period, from December 1979 to March 1980, an 8 ft (2.5 m) rise in reservoir stage, the reservoir contributed water to Echo Wash bank storage at rates of up to 0.29 gpd/ft2 (12 lpd/m2). Ground-water discharge occurred for the remainder of the project, during a stage decline from April 1980 to May 1980, a rise in June 1980, and leveling off and slight decline for the remainder of the year, July 1980 to December 1980. The maximum seepage meter ground-water discharge rate of 3.0 gpd/ft2 (122 lpd/m2) was recorded in December 1980. Seepage meter water chemistry data for June were similar to Lake Mead water chemistry and were interpreted to be previously recharged Lake Mead water. September water chemistry data showed two possible components of ground-water discharge, a high SpC calcium sulfate Echo Wash ground water and a lower SpC Lake Mead recharged bank storage water. December ground-water chemistry data showed discharge to be a high SpC calcium sulfate water similar to Echo Wash ground-water quality which was apparently unaffected by Lake Mead inflow. Mini-piezometer data were collected at each seepage meter site. However, these data usually did not provide correlative results with seepage meter data probably because of suspended sediment in the piezometer water column and plugging of the perforated tip. Seepage meters were successfully utilized to characterize reservoir ground-water interaction in Echo Bay.

    Notes: Abstract: Tubulin, isolated from brain tissue of chicks at different stages during late embryonic and early posthatched development by ion-exchange chromatography and by in vitro microtubule reassembly, was analyzed by high-resolution isoelectric focusing and by two-dimensional polyacrylamide gel electrophoresis. Similar results were obtained with tubulins purified by the two methods. Sixteen isoelectric species of tubulin that differ in apparent net charge under denaturing conditions were detected by isoelectric focusing. By two-dimensional polyacrylamide gel electrophoresis, the chick brain tubulins were resolved into at least seven forms of α and 10 forms of β tubulin. The number and relative proportions of the maltiple brain tubulins were modulated during development. Since there are only four α tubulin and four β tubulin genes in chickens, posttranslational modification of the tubulins must play a prominent role in the heterogeneity. Analysis of isotubulin distributions through cycles of microtubule assembly and disassembly indicated that the tubulins differ very little, if at all, in their capacity to assemble into microtubules. Therefore, the chemical differences that distinguish the multiple tubulins have very little structural impact on the protein surface areas involved in microtubule formation. Partial fractionation of the multiple tubulins during ion-exchange chromatography was observed, suggesting that it may be possible to isolate individual native tubulin variants for biochemical studies.

    Notes: Radiofrequency Ablation in Pediatric Ebstein's Anomaly. Introduction: Abnormal anatomy and complex electrophysiology in patients with Ebstein's anomaly of the tricuspid valve may confound attempts at radiofrequency ablation (RFA). Methods and Results: Data for 65 pediatric Ebstein's patients (9.8 ± 5.4 years, 4 months to 20 years; 39 ± 25 kg, 5.1 to 108 kg) were obtained from the Pediatric Radiofrequency Ablation Registry. The degree of tricuspid regurgitation (DOTR) and the degree of Ebstein's anomaly were assessed with echocardiography/Doppler. Leading indications were drug refractoriness (24 [37%] of 65 patients) and life-threatening arrhythmia (14 [22%] of 65 patients). For the 65 patients, 82 typical (nondecremental) accessory pathways (APs) (62% right free wall, 34% right septal, and 4% left sided), 17 other supraventricular tachycardias (1 ectopic atrial, 7 AV reentry, 5 Mahaim, and 4 intra-atrial reentry tachycardias), and 1 ventricular mechanism were mapped. Thirty-four (52%) of 65 patients had a single AP (21 right free wall, 10 septal, and 3 left); 19 (29%) of 65 patients multiple APs; 6 (9%) of 65 patients a single AP plus a non-AP mechanism; and 6 (9%) of 65 patients non-AP mechanism(s) only. REA acute success rates and recurrence rates for right free wall, right septal, and other mechanisms were 79%/32%, 89%/29%, and 75%/27%. Mild DOTR and a body surface area (BSA) ≤ 1.7 m2 independently predicted a better acute success rate. BSA ≤ 1.7 m2 also predicted long-term success. Conclusion: In this patient subset, life-threatening arrhythmias and multiple electrophysiologic mechanisms are commonly encountered during REA. Mild DOTR and a BSA ≥ 1.7 m2 predict a higher acute success rate. While acute success rates are relatively high, recurrence is frequent.
