    Notes: Abstract During the life cycle of Physarum polycephalum, uninucleate amoebae develop into multinucleate syncytial plasmodia. These two cell types differ greatly in cellular organisation, behaviour and gene expression. Classical genetic analysis has identified the mating-type gene, matA, as the key gene controlling the initiation of plasmodium development, but nothing is known about the molecular events controlled by matA. In order to identify genes involved in regulating plasmodium formation, we constructed a subtracted cDNA library from cells undergoing development. Three genes that have their highest levels of expression during plasmodium development were identified: redA, redB (regulated in development) and mynD (myosin). Both redA and redB are single-copy genes and are not members of gene families. Although redA has no significant sequence similarities to known genes, redB has sequence similarity to invertebrate sarcoplasmic calcium-binding proteins. The mynD gene is closely related to type II myosin heavy-chain genes from many organisms and is one of a family of type II myosin genes in P. polycephalum. Our results indicate that many more red genes remain to be identified, some of which may play key roles in controlling plasmodium formation.

    Notes: The use of information channels by individuals in organizations is substantially discretionary. Although much studied, variation in patterns of discretionary use has yet to be explained adequately in terms of an accepted integrative theory validated across varieties of channel types and organizational settings. In a step toward the development of such a theory, the present paper proposes a basic channel-disposition model that relates an individual's attitude or disposition toward a channel to his/her actual use of the channel in the context of information and channel supply-and-demand structures in the organization. An exploratory study of the dispositions of 186 users of ten management information system (MIS) reports in four organizational settings provides limited evidence in support of the model proposed.

    Notes: We have demonstrated wavelength conversion in the 1.5 μm regime using the near band gap nonlinearity of semiconductor quantum wells. In a 7.5 mm long passive InGaAsP/InP single quantum well waveguide, a wavelength shift of 20 nm has been obtained with −11 dB conversion efficiency using picosecond pulses. We confirmed the ultrafast nature of the nonlinearity by measuring the conversion efficiency as a function of the frequency shift. An order of magnitude increase in the value of n2 close to the band gap is observed compared with the off-resonance value. © 1996 American Institute of Physics.

    Notes: Abstract Microspores of several genotypes of Brassica campestris ssp. parachinensis have been cultured in vitro and induced to undergo embryogenesis and plant formation. Conditions favourable for embryogenesis in this species include a bud size of 2–2.9 mm, NLN-13 culture medium (Nitsch and Nitsch 1967; Lichter 1981, 1982; Swanson 1990), and an induction through exposure to 32°C for a period of 48 h. Longer periods of an elevated temperature for induction of embryogenesis resulted in embryo abortion at early developmental stages. With the protocol developed here, microspores of 60–80% of donor plants could be induced to produce embryos, although embryo yields were low, i.e. 2–5 embryos per 10 buds. Some genotypes responded to culture conditions with high numbers of embryo formation (100–150 embryos per 10 buds) but most of these subsequently failed to mature. The pattern of cell division and morphological changes of the microspores in culture were studied using various microscopic techniques.

    Notes: Summary Microspore-derived embryos of Brassica napus were transformed using the disarmed octopine-producing LBA4404 strain of Agrobacterium tumefaciens containing the binary vector pBin19. Octopine-producing strains have previously been reported to be ineffective in transforming Brassica. Four actively growing yellow/ green sectors were selected from the embryos on 50 mg/l kanamycin and plants regenerated. Analysis for NPT-II activity in these young plants initially indicated no expression of the bacterial NPT-II gene. The plants were nevertheless grown to maturity, selfed and S1 seed was collected. Three of the S1 plants produced microspores which were from 4 to 20 times more tolerant to kanamycin than the original parent. Southern analysis revealed that one plant (EC-1) had a single site of insertion and the other two plants (EC-2 and EC-6) had two sites of insertion with sequence homology to the bacterial NPT-II gene. Microspores from the EC-2 and EC-6 transgenics produced embryos on approximately five times the level of kanamycin tolerated by microspores from untransformed plants, while the EC-1 transgenic produced microspores with more than 20 times the tolerance to kanamycin. Analysis of S1 progeny of the EC-1 transgenic indicated that 100% of the progeny exhibited the trait through both Southern analysis and by expressing tolerance to kanamycin in microspore-derived embryos.

    Notes: Summary In vitro microspore mutagenesis and selection was used to produce five fertile double-haploid imidazolinone-tolerant canola plants. The S2 plants of three of the mutants were resistant to at least the field-recommended levels of Assert and Pursuit. One mutant was tolerant to between five and ten times the field-recommended rates of Pursuit and Scepter. Two semi-dominant mutants, representing two unlinked genes, were combined to produce an F1 hybrid which was superior in imidazolinone tolerance to either of the heterozygous mutants alone. Evaluation of the mutants under field conditions indicated that this hybrid and the original homozygous mutants could tolerate at least two times the field-recommended rates of Assert. The field results indicated the mutants were unaffected in seed yield, maturity, quality and disease tolerance. These genes represent a potentially valuable new herbicide resistance system for canola, which has little effect on yield, quality or maturity. The mutants could be used to provide tolerance to several imidazolinones including Scepter, Pursuit and Assert.

    Notes: Abstract A homogenization and plating technique is described which increases the number of shoots produced and decreases the time required for plant regeneration from callus cultures of birdsfoot trefoil. A 2- to 15-fold increase in the number of plants recovered per gram of callus is observed depending on the genotype. Characterization of a sample of the regenerated plants indicated no differences between plants from homogenized versus nonhomogenized callus for traits such as time of first flower, number of branches per plant, pollen stainability, stomate length, and whole plant yield. The technique has proven useful for efficient recovery of plants from long-term cultures and cultures selected for herbicide tolerance where a 15-fold increase in plant regeneration was obtained.

    Notes: Abstract Microspore cultures were initiated from the North American sweet corn hybrid, ‘Seneca 60’. Donor plants were grown under two environments. One treatment comprised plants that matured completely in the greenhouse (GH) (28°C/23°C: day/night), while in a second treatment donor plants were isolated and divided into two treatment sets: cultured directly at 25°C, or given a heat treatment of 32°C for 10 days. Greenhouse-grown plants produced fewer embryo-like structures (ELS) than growth chambertreated plants regardless of the culture temperature treatment. If the microspores isolated from GC plants were subsequently provided with the initial high culture temperature, the number of ELS that could be recovered was more than doubled compared to the cultures incubated at 25°C continuously. The high culture temperature treatment also resulted in a higher quality of ELS (more compact), which led to a higher frequency of ELS that survived and were subsequently transferred to regeneration medium. However, while plant regeneration and subsequent selfed seeds were obtained, the frequency was very low and further research is required in this area.
