    Notes: Summary Basophil-rich infiltrates in regional lymph nodes of guinea-pigs were demonstrated by electron microscopy after the intradermal injection of T cell mitogens (PHA and Con A). Basophils infiltrated the stroma of the lymph node via the postcapillary venules (PCV) and migrated to the paracortex. Prior to infiltration of the lymph nodes a cutaneous basophil hypersensitivity reaction was seen in the mitogen-injected skin. B cell mitogen (LPS) injection did not induce this response.

    Notes: Abstract Parathyroid crisis occurring in primary hyperparathyroidism is characteized by extremely high circulating levels of parathyroid hormone and acute onset of severe hypercalcemia. We describe a 62-year-old woman with parathyroid crisis probably due to an intraturnoral hemorrhage. Renal dysfunction reduced the effectiveness of preoperative management and continued to deteriorate for 5 days after parathyroidectomy. The normalization of serum calcium after parathyroidectomy delayed and it took 6 days. Maintenance of renal function is important for pre- and postoperative courses of the present case. The rapid decrease in serum parathyroid hormone after parathyroidectomy was followed by a rapid and transient (about fivefold) increase in serum alkaline phosphatase with peak value on the 10th postoperative day. This indicated that reversal phase from bone resorption (accelerated by parathyroid hormone) to bone formation lasted about 10 days under the conditions of the present case.

Kinase assay in myelin basic protein (MBP) containing polyacrylamide gels revealed that endothelin-1 (ET-1) and ET-3 increased MBP kinase activities in glomerular mesangial cells (MC) from Wistar-Kyoto (WKY) rats and spontaneously hypertensive rat (SHRSP). ET-1 stimulated MBP kinase activities more potently than ET-3.2. Immunoprecipitation with anti-41-kDa MAPK antiserum showed that the MBP kinases activated by ET-1 correspond to 43- and 41-kDa MAPK.3. Since Phorbol 12-myristate 13-acetate, a direct activator of protein kinase C, also activated MAPK, protein kinase C was suggested to mediate ET-induced activation of MAPK.4. These results suggest that MAPK may mediate the ET actions in glomerular mesangial cells from normotensive rats as well as spontaneously hypertensive rats. Since ET is produced by vascular endothelial cells of the kidney and glomerular mesangial cells, the ET signalling pathway may have some physiological and pathophysiological significance in wivo glomerulus.

    Notes: Abstract The relationship between parathyroid hormone (PTH) secretion and extracellular calcium (Ca) level is reciprocal causality. The equilibrium operating point determines basal PTH secretion rate and basal extracellular Ca level. We studied how this equilibrium was achieved in the subjects with decreased PTH secretion or decreased parathyroid glands number. Basal/maximum ratio of serum PTH, which reflects the basal secretory state of parathyroid glands, was increased in 9 hypoparathyroid patients treated with vitamin D3 (VD3) [7 patients with idiopathic hypopar-athyroidism (IHP), and two patients with postsurgical hypoparathyroidism] and in seven of nine parathyroid adenectomized patients. There was a negative correlation between the ratio and basal serum Ca level in the patients with IHP after VD3 treatment (r=0.7167, P〈0.05) and in the patients after parathyroid adenectomy (r=0.7760, P〈0.05). The regression curves in these two groups coincided regardless of the difference in maximum PTH secretion rate, which suggested that the basal secretory state of parathyroid glands was determined by extracellular Ca level in a similar manner in these subjects. There was a sigmoidal relationship between basal/maximum ratio of serum PTH and basal serum Ca level, when the data were collected from 15 hypoparathyroid patients before or after VD3 treatment, 9 parathyroid adenectomized patients, and 10 normal subjects (r=0.9057, P〈0.001). This sigmoidal curve is thought to represent the fundamental relationship between the basal secretory state of parathyroid glands and extracellular Ca level.

    Notes: Summary Acid phosphatase (ACPase) was ultracytochemically demonstrated in the lymph-node sinus reticular cells and macrophages of rats. After the uptake of horseradish peroxidase (HRP), marked ACPase activities were seen in both reticular cells and macrophages, although only sparse ACPase activity was detected in the reticular cells of the control. After the injection of HRP into the footpad, the mast cells in the regional lymph node became degranulated, and the released granules were taken up by reticular cells and macrophages. In macrophages, these taken-up mast-cell granules exhibited ACPase reaction products, whereas none of the granules taken up by reticular cells showed ACPase activity. The heparin-protamine complex was also engulfed by reticular cells and macrophages, and ACPase activity was demonstrable in the complex taken up by both types of cell. It is probable that, as is the case in macro-phages, reticular cells in the lymph-node sinuses take up and digest foreign substances through the formation of phagolysosomes, but they do not digest granules originating from the mast cells in the lymph node of the same animal.

    Notes: Abstract  In order to evaluate the roles of the RET proto-oncogene in normal human tissues and tumors derived from the neural crest cells, we examined the expression of RET in a variety of adult human tissues, pheochromocytomas, medullary thyroid carcinomas (MTCs), ganglioneuromas, and a neurinoma. Northern blot analysis demonstrated that RET is normally expressed in the adrenal medulla and cerebellum among adult human tissues. RET transcripts were detected in all of 11 sporadic and one familial pheochromocytomas. The levels of RET mRNA were higher in 8 of 12 pheochromocytomas than in the normal adrenal medulla, indicating that RET is overexpressed in the majority of sporadic pheochromocytomas. There was a considerable difference in the level of RET expression in each pheochromocytoma ranging from 0.2 to 10 times the transcripts found in the normal adrenal medulla. The sizes of the transcripts of 7.0, 6.0, 4.5, and 3.9 kb were the same as those found in the adrenal medulla, suggesting no rearrangements of the RET gene in pheochromocytomas. Southern blot analysis revealed neither amplification nor gross structural changes in the RET gene. RET was also expressed at high levels in four MTCs examined, whereas its transcripts were detected at low abundance in only one of three ganglioneuromas. RET was not expressed in a neurinoma. These results suggest that RET may play some roles in a limited range of adult human tissues, and that its high levels of expression may have relevance to development or growth of pheochromocytomas and MTCs.

MRI showed periventricular lesions, high intensity on T2-weighted and low intensity on T1-weighted images at trigones of the lateral ventricles. 1H-MRS of these lesions revealed high lipid and low N-acetyl aspartate peaks. We presume such lipids in periventricular lesions with high T2 signal may be pathognomonic of SLS.

    Notes: Abstract The paper presents a microcomputer-aided pyroelectric thermal imaging system developed for medical use. The processing procedures of this system can be divided into two stages: data acquisition and noise reduction. In the first stage, the video signal produced by the pyroelectric vidicon camera is digitised in synchronism with a chopper. In the second stage, averaging, immage-difference processing and local smoothing are applied to the obtained digital image. As a result, temperature resolution of less than 0·5°C was achieved on a four-bit digital image. The system was applied to pain management, and demonstrated a good performance in the evaluation of effects of nerve-block operations.
