    Notes: Summary Fas ligand (FasL) is highly expressed in testicular tissues and thought to be responsible for protection from allograft rejection by inducing apoptosis of anti-graft activated T cells. FasL-expressing islets have been shown to induce a granulocyte-mediated inflammatory reaction. We investigated whether a graft can be protected from alloimmune responses by manipulating the Fas/FasL-system. We transplanted allogeneic islets under the kidney capsule of streptozotocin-induced diabetic mice together with testicular tissue. Significant prolongation of survival of C3H islet allograft was observed in C57BL/6 (B6) recipients transplanted with C3H testicular tissue, but not in those transplanted with C3H-gld testicular tissue expressing non-functional FasL. No significant prolongation was observed in B6-lpr recipients expressing non-functional Fas. Immunohistochemical staining of C3H testicular tissue in the composite graft showed a high expression of FasL, but not that of the C3H-gld testicular tissue. In situ terminal deoxynucleotidyl transferase-mediated dUDP-biotin catalysed DNA nick-end labelling (TUNEL) staining of a composite graft of C3H islet and testicular tissue in B6 recipients demonstrated extensive apoptosis of infiltrating mononuclear cells around the graft. The protective effect of C3H testicular tissue was abrogated when anti-FasL monoclonal antibody was administered i. p. postoperatively. Our results suggest that FasL-positive testicular allografts protect composite islet allografts and indicate that manipulation of Fas/FasL mediated apoptosis is a suitable strategy for controlling rejection of islet allografts.     Notes: Summary Abnormal granular structures, which stained positively with periodic acid-Schiff (PAS-positive granular structures; PGS), were observed in the brain of senescence accelerated mouse (SAM). They were small, round to ovoid, homogenous structures measuring up to 5 μm in diameter and usually grouped in clusters. PGS were localized in the hippocampus, piriform cortices, olfactory tubercle, nucleus of the trapezoid body, and cerebellar cortices. Quantitative analysis revealed that PGS remarkably increased in the hippocampus of SAM-P/8, a substrain of SAM, with advancing age, although a few PGS also appeared in the aged control mice, SAM-R/1 and DDD. Their histochemical nature, morphological features and distribution pattern were different from those of corpora amylacea and other similar bodies. A close anatomical relationship between PGS and glial fibrillary acidic protein-positive astrocytes was inferred from immunohistochemical studies. PGS is considered to be one of the morphological manifestations of senescence in mice brains, and are found to occur more numerously in the brains of learning or memory deficit mice, SAM-P/8.

    Notes: Summary Serum coenzyme Q10 (CoQ10) levels were measured at rest and during incremental exercise in 21 patients with chronic obstructive pulmonary disease (COPD) and 9 patients with idiopathic pulmonary fibrosis (IPF). The mean serum CoQ10 levels at rest in patients with COPD and IPF were 0.56 ± 0.20 and 0.45 ± 0.16 μg/ml, respectively. In both groups these levels were decreased compared with those of healthy subjects. In the patients with COPD, CoQ10 levels were significantly correlated with body weight, however, there was no correlation between CoQ10 levels and ventilatory function, PaO2, VO2/kg at rest, or maximal VO2. In eight of nine patients whose PaO2 at rest was lower than 75 torr, serum CoQ10 levels were lower than 0.5 μg/ml. We studied the effects of the oral administration of CoQ10 at 90 mg/day for 8 weeks on pulmonary function and exercise performance in eight patients with COPD. Serum CoQ10 levels were significantly elevated in association with an improvement in hypoxemia at rest, whereas pulmonary function was unaltered. Oxygen consumption during exercise was not changed, whereas PaO2 was significantly improved, and heart rate was significantly decreased compared with the results obtained at an identical workload at baseline. Furthermore, lactate production was suppressed during the anaerobic exercise stage after CoQ10 administration, and exercise performance tended to increase. These data suggested that CoQ10 has favorable effects on musclar energy metabolism in patients with chronic lung diseases who have hypoxemia at rest and/or during exercise.

    Notes: Summary There are two pathways known to mediate optic signals to the rabbit flocculus through the dorsal cap of the inferior olive; one (I) projects from the contralateral and the other (II) from the ipsilateral eye, both to the contralateral flocculus. The present study on anesthetized rabbits revealed that the two pathways are relayed at different loci of the dorsal cap, I at its caudal and II its rostral half. The ventrolateral outgrowth of the inferior olive also projects to the flocculus, but a prominent field potential could not be produced by optic signals. The present study further demonstrated that transmission in pathway I accompanies an inhibitory action upon that in II. Since direct stimulation of these pathways at preolivary levels revealed similar inhibition, and since this was reflected in the depression of antidromic invasion of dorsal cap neurons, it is concluded that inhibition occurs postsynaptically in dorsal cap neurons. Inhibition is unidirectional, as activities in pathway II did not influence those in I.

    Notes: Abstract Fluctuations in the amount of vanillylmandelic acid (VMA) and homovanillic acid (HVA) were studied in random urine samples from 13 infants with neuroblastoma. In patients with a small tumour, many samples contained amounts below the cut off values, suggesting that detection of a patient with neuroblastoma depended on mathematical probability. Using high performance liquid chromatography a patient with a tumour of about 10 g may well be overlooked, whereas a patient whose tumour weighs over 30 g would probably be detected. With qualitative screening, although more likely to miss a patient with low VMA excretion, a patient with a tumour weighing over 50 g would be detected. In a thorough hospital examination care should be exercised in interpreting borderline values of VMA and HVA; excretion information suggesting the existence of a tumour may aid localization by radiological imaging procedures.

    Notes: Abstract Sixty-eight cases of neuroblastoma detected during mass screening being performed by 11 local self-governing bodies of Japan were studied concerning age, clinical stage, and survival rate. Their average ages at the first screening, at the beginning of thorough examination, and at the start of therapy were 215.7, 245.6, and 264.7 days, respectively. This screening, which is aimed at the 6-month-old infants is considered to be acceptable, on the grounds that the majority of the cases (95.6%) were asymptomatic at the time of thorough examination, that for most of the cases (98.5%) the initiation of therapy was at under 1 year of age, and that their 60-month survival rate was 87.5%. However, there seems to be room for discussion of the best age at which subjects should be screened, because the 68 cases included some patients already with advanced stages at the time of thorough examination, and because “false negative cases” were identified besides the 68 positive cases.

    Notes: Summary Carvedilol, a novel β-blocker with a vasodilating action, was given either alone (monotherapy) or with diuretics (combination therapy) to 42 patients with renal hypertension. The hypotensive effect, safety, and optimal dose were investigated. In all, 23 untreated patients (16 men and 7 women; average age, 56.4 ± 2.5 years) made up the monotherapy group and 19 diuretic-treated patients (11 men and 8 women; average age, 56.4 ± 2.5 years) comprised the combined therapy group. All subjects had an initial blood pressure (BP) of 〉 160/95 mmHg and were started on 5 mg/day oral carvedilol. The dose was gradually increased to a maximum of 20 mg/day, or until either the BP was reduced to 〈 149/89 mmHg or the reduction in mean BP was 〉 13 mmHg compared with baseline levels. The total study period was 8 weeks. With monotherapy, the BP and heart rate decreased significantly from 167/102 to 150/94 mmHg and from 81 to 74 beats/min, respectively. With combined therapy, the BP and heart rate fell significantly from 176/103 to 142/85 mmHg and from 81 to 70 beats/min, respectively. Responders were defined as subjects with a BP of ⩽ 149/89 mm Hg or those showing a fall of ⩽ 13 mmHg in mean BE Responders accounted for 52.2% of the monotherapy group and 73.6% of the combination therapy group. Orthostatic hypotension was not seen in either group. Serum creatinine and blood urea nitrogen (BUN) levels were not altered by administration of carvedilol. Dizziness was noted by 1 of the 23 subjects in the monotherapy group. These results suggest that carvedilol is an effective and well-tolerated antihypertensive agent for the management of renal hypertension. We considered the optimal dose to be from 10 to 20 mg once daily, which is similar to that recommended for essential hypertension.

MReffect was found out in the thin films obtained by post-annealing of the following two kinds of sputterdeposited iron nitride related films. (1) α-Fe particles dispersed in AlN granular film was obtained byan annealing of Al0.31Fe0.69N sputter deposited film in hydrogen. The MR=0.82% was found out inthis nitride system. (2) Fe3O4 thin films were prepared by thermal decomposition of sputter depositediron nitride films in low oxygen partial pressure.     Notes: The real part, f' of the anomalous dispersion correction to atomic structure factors was measured by using Pendellösung fringes for Si 220 and {\bar 22}0 reflections in the wavelength range from Cu Kα1 to Rh Kα1. In order to avoid ambiguity in the Thomson-scattering term due to the nature of the bonding, the quantity (f'X - f'Ag) was compared with the available theoretical calculations. In this expression, f'X and f'Ag are the corrections for radiation X and Ag Kα1, which is used as the standard, respectively. The present experimental results fit best the values calculated by Cromer & Liberman [J. Chem. Phys. (1970), 53, 1891-1898].
