    Notes: Abstract. Objective and Design: We investigated the effect of a novel N-benzoyl-L-phenylalanine derivative compound (JTE-607) on production of various cytokines and other immune responses in vitro and on endotoxin shock in vivo.¶Materials and Methods: Human, monkey, rabbit, mouse and rat peripheral blood mononuclear cells (PBMCs), and human fibroblasts, umbilical vein endothelial cells (HUVEC), mesangial cells and T cells were used in vitro. Endotoxin shock was induced by lipopolysaccharide (LPS) in Corynebacterium parvum (C. parvum) sensitized male C57BL/6 mice in vivo.¶Results: JTE-607 inhibited inflammatory cytokine production, including tumor necrosis factor (TNF)-α, interleukin (IL)-1β, IL-6, IL-8 and IL-10, from LPS-stimulated human PBMCs, with IC50 values of 11, 5.9, 8.8, 7.3 and 9.1 nM, respectively. The inhibitory effects of JTE-607 were also seen in mRNA expression of those cytokines. The potency of JTE-607 on cytokine production from PBMCs of other species, and from other human cells were much lower than that on human PBMCs. JTE-607 did not affect either LPS-stimulated microbead phagocytosis or reactive oxygen species production at 1 μM in human PBMCs but slightly suppressed expression of major histocompatibility complex class II antigen at 1 μM, although it was 100-fold less active than it was as a cytokine inhibitor. JTE-607 (0.3-10 mg/kg, i.v.) showed dose dependent inhibition of mortality after LPS challenge in C. parvum sensitized mice in accordance with a decrease of plasma TNF-α.¶Conclusions: These results suggest that JTE-607 is a multiple cytokine inhibitor specific for human PBMCs. This compound may be useful for the treatment of various cytokine mediated diseases such as septic shock without causing immunosuppression.

    Notes: Abstract Oral FPL-52694 [5-(2-hydroxypropoxy)-8-propyl-4-oxo-4H-1-benzopyran-2-carboxylic acid Na], a new mast cell stabilizer, dose-dependently inhibited gastric acid secretion but increased the volume and pepsin output in pylorus-ligated rats. Intraduodenal FPL-52694 significantly inhibited all of the volume, acidity, acid output and pepsin output. Concerning the acidity, oral administration of the agent showed much more potent inhibition than intraduodenal administration. Oral FPL-52694 markedly inhibited the development of pylorus-ligated ulcers, water-immersion stress- and aspirin-induced gastric erosions and moderately inhibited the formation of reserpine-induced gastric erosions in rats. Intraduodenal FPL-52694 also inhibited pylorus-ligated ulcers whereas it had no effect on aspirin-induced gastric erosions. Histamine-induced gastric erosions were not affected by oral FPL-52694. These effects of FPL-52694 were almost the same as those of cimetidine, except that cimetidine tended to inhibit histamine-induced gastric erosions. Although the precise mechanism of action of FPL-52694 remains unknown, oral FPL-52694 appears to be a promising agent for the treatment of peptide ulcers.

    Notes: Abstract.  It was demonstrated experimentally for the first time that particle growth of photoproduced UF5 was enhanced by ionizing radiation from an α-emitting 241Am source. A gas mixture of UF6, CH4, and Ar in a reaction cell was irradiated with a single shot of a KrF laser at 248 nm to form UF5 particles. Transmission electron microscope images of the samples indicated that the average size of UF5 particles produced without the α-ray source was independent of the laser energy. UF5 particles produced with the α-ray source were found to be much larger in size than those produced without the α-ray source, and the enhancement of particle growth was marked when the laser energy was low. An explanation was attempted for the anomalous behaviors of particle size distribution from the viewpoint of competition between the second-order coagulation rate and the rate of diffusion from the reaction zone.

    Notes: Abstract An improved analytical method is presented in which the reaction yield in the IR multiple-photon dissociation by a focused uniform beam is expressed in a generalized form as a function of fluence. The analytical solution is derived for a real focusing geometry by assuming the cumulative log-normal distribution (CLND) for the functional form of the dissociation probability vs. fluence. Also presented is a shortcut analytical method with simple and convenient algebraic expressions which approximate the exact analytical solution, thereby speeding up the analysis of experimental data.

    Notes: Abstract A 287 base pair insertion/deletion polymorphism in intron 16 of the angiotensin I converting enzyme (ACE) gene was examined by polymerase chain reaction in 36 Kawasaki disease patients (16 without coronary aneurysm, 20 with coronary aneurysm). A polymorphism in the ACE gene was characterized by three genotypes: two D alleles (genotype DD), two I alleles (genotype II), and heterozygous allele (genotype DI). Genotype II was found in 65% of the patients with aneurysm and 12.5% of those without aneurysm (P 〈 0.01, odds ratio 13.0, 95% confidence intervals). Conclusion Patients with Kawasaki disease and coronary aneurysm more often than expected had the genotype II suggesting that reactions induced by the type of ACE polymorphism predispose to coronary aneurysm formation.

    Notes: Summary A study was made of intrathecal infusion as a route for administration of drugs for the CNS. We recognize the presence of blood brain and CSF brain barriers. CDP-choline was used as the brain activator, and the effect of the intrathecally infused drug was investigated clinically. Fifteen patients with disturbances of consciousness due to primary cerebral lesions received a total of 39 intrathecal infusions of CDP-choline, and were examined neurologically and electroencephalographically before and after treatment.

    Notes: Summary A rare case of dural haemangioma with extracranial component is described. A subcutaneous frontal tumour was thought before operation to have originated in the skull. Angiography showed drainage into the superior sagittal sinus via a cortical vein. Dural haemangioma is thought to be a clinical rarity. Also, an arteriovenous malformation draining into the superior sagittal sinus is rarely encountered.

    Notes: Summary Both the Moyamoya Phenomenon and occlusion of the internal carotid fork are essential radiological findings in true Moyamoya Disease of unknown aetiology. However, the Moyamoya Phenomenon is often observed in occlusive diseases of the internal carotid bifurcation of known aetiology. The authors recently observed acute development of the unilateral Moyamoya Phenomenon following severe vasospasm of the anterior and middle cerebral arteries due to rupture of an anterior communicating aneurysm. The following four factors have been suspected of contributing to development of the Moyamoya Phenomenon: 1. The chronology of arterial occlusion. 2. Extent and location of occlusion. 3. The cause of occlusion. 4. Anatomical and functional disposition of the basal circulation. As regards the chronology, chronic or slowly progressive arterial stenosis has been thought to be a mandatory factor in development of a Moyamoya network, which plays an important role in the form of collateral channels. However, based on the findings outlined in this paper, the congenital factor may be the most important of the four factors.

    Notes: Summary Statistical evaluation of 71 cases (adults) of histologically confirmed supratentorial surgically treated glioblastoma multiforme seen during the period 1958 to 1973 revealed that the state of calcification as determined by roentgenography and the histological appearance of the neoplasm appeared to have a relationship with the postoperative period of survival. Postoperative adjuvant treatment, location of the neoplasm, duration of preoperative symptoms, patient's age at onset, preoperative condition, and cystic formation in the neoplasm also appeared to have a relationship, although it was slight.
