    Notes: Summary The presence of calcitonin in the cat thyroid was studied immunohistochemically in a series of gland development. the first positive cells are to be found on the 38th day of gestation, i.e. 1–2 days after level nine of ontogenetic development has been reached. The cytoplasm of these cells form only a narrow border round the nucleus. With advancing development the bumber of calcitonin-positive and its amount increases. From approximately the 50th day of prenatal development, the initially diffusely scattered, solitary calcitonin-positive cells are gradually replaced by groups of cells, which begin to occupy a characteristic position in relation to the follicular epithelium. The largest quantity of calcitonin-positive cells is found in foetuses about to be born. In non-pregnant adult cats, the incidence of immunohistochemically calcitonin-reactive cell is more sporadic and their distribution in the lobes of the thyroid is uneven.

    Notes: Zusammenfassung 1. Das Verhältnis der A- und B-Zellen in den Langerhansschen Inseln der erwachsenen Ratte zeigt Unterschiede nach dem Geschlechte. Die Weibchen haben in den durchschnittlichen Inseln eine höhere Prozentzahl an B-Zellen als die Männchen. In unserem Material hatten die Weibchen annähernd 75%, die Männchen 68% B-Zellen in den durchschnittlichen Inseln. 2. Das prozentuale Verhältnis der A- und B-Zellen ist vom Alter abhängig. Im Gegensatz zu der Relation bei den erwachsenen Tieren entspricht es bei den 50 g schweren Männchen und Weibchen annähernd dem Verhältnis 1∶1. Weibchen haben 53,3% B-Zellen, Männchen 53,7% B-Zellen. 3. Die Änderung des Verhältnisses der A- und B-Zellen zugunsten der B-Zellen ist an die Geschlechtsreifung gebunden und wird bei den Weibchen schneller durchlaufen, bei den Männchen langsamer. Die ersten Unterschiede zwischen den Inseln beider Geschlechter wurden in unserem Material beim Stadium von 75 g Körpergewicht festgestellt; die Weibchen hatten 63,5% und die Männchen 61% B-Zellen. Der größte Unterschied ist beim 100 g-Stadium vorhanden: Die Weibchen hatten 72,6%, die Männchen 64,0% B-Zellen. Von diesem Stadium an bleibt der Prozentsatz der B-Zellen bei den Weibchen im Grunde unverändert, bei den Männchen steigt er langsam weiter an. 4. Bei den kastrierten Ratten verschwinden diese Geschlechtsunterschiede in der prozentualen Verteilung der A- und B-Zellen sehr schnell. Die Kastraten haben eine ähnliche prozentuale Verteilung der A- und B-Zellen wie die unreifen Tiere. 5. Mitosen der A-Zellen findet man bei der normalen Ratte praktisch nur bei den unreifen Stadien. Ihr Vorkommen ist aber nicht, wie die Literatur angibt, nur auf die fetale Zeit und die Periode knapp nach dem Wurfe beschränkt. Mitosen der B-Zellen kann man in jedem Alter finden. Nach der Kastration kann man sowohl A-Zellmitosen als auch B-Zellmitosen in allen Altersstadien feststellen.

    Description / Table of Contents: Zusammenfassung 1. Die Hauptmasse der Langerhansschen Inseln von Natrix natrix L. und Natrix tessellata Laurentii liegt im milznahen Teil des Pankreas. Sie bestehen aus verzweigten Bändern von Inselzellen die an das exokrine Gewebe meist unmittelbar angrenzen. Dünne Ausführungsgänge können in die Inseln eingebaut sein. 2. Lichtmikroskopisch lassen sich mit Hilfe verschiedener, am gleichen Schnitt angewandter Färbemethoden 3 Zelltypen unterscheiden: A-, B- und D-Zellen. 3. Auch elektronenmikroskopisch werden aufgrund der Granulation 3 Zelltypen differenziert. Die A-Zellen enthalten große, kugelförmige Granula (640±140 nm) mit homogenem, stark osmiophilen Inhalt. Die Form der β-Granula (500±95 nm) wechselt je nach angewandter Fixationsmethode; bei Doppelfixation in Glutaraldehyd-OsO4 ist der Innenkörper meist wenig elektronendicht, nach Fixation mit OsO4 allein fällt der Granuluminhalt vielgestaltig aus. Einige Granula besitzen eine Substruktur mit periodischer Querstreifung. 4. Von den Granula des dritten Zelltyps, der D-Zellen, wird bei OsO4-Fixerung die Hüllmembran, bei Doppelfixation auch der feingekörnte Inhalt erhalten. Nur die ϑ-Granula zeigen unter dem Elektronenmikroskop deutliche Argyrophilie; die α-Granula sind gewöhnlich nur schwach imprägniert.
    Notes: Summary 1. In two species of snakes, Natrix natrix L. and Natrix tessellata Laurentii, the bulk of islet tissue is concentrated in the splenic part of the pancreas. The islets are composed of tortuous bands of cells and in close contact with the exocrine tissue without any connective tissue capsule. 2. By light microscopy 3 different cell types can be distinguished (A-, B- and D-cells) after staining the same section with different procedures. 3. Under the electron microscope these 3 cell types can be identified by shape and size of their secretory granules: the contents of the large spherical α-granules (average diameter 640±140 nm) appears very electron dense and homogenous. The shape of the β-granules changes according to the fixatives employed: after the double fixation with glutaraldehyde OsO4 the core of the granule is slightly electron dense, after fixation with OsO4 alone its shape is polymorphic. The substructure of some granules shows a periodical striation. 4. In the granules of the third cell type, the D-cells, after OsO4-fixation, only the limiting membrane is preserved. After double fixation the interior of the granule contains a pale, granular material. After silver impregnation of frozen sections only the δ-granules are intensively argyrophilic under the electron microscope.

    Description / Table of Contents: Zusammenfassung 1. Das Pankreas von Testudo graeca und Emys orbicularis, das licht- und elektronenmikroskopisch untersucht wurde, enthält zwei Inseltypen. Einmal die soliden Inseln, die von größeren Zusammenballungen endokriner Zellen gebildet werden, zum anderen verstreute endokrine Zellen, teilweise in der Nähe von größeren Ausführungsgängen, hauptsächlich aber im exokrinen Gewebe an der Basalmembran der kleineren Gänge. 2. Durch spezifische Färbung kann man in den soliden Inseln lichtmikroskopisch bis zu drei Zelltypen differenzieren, die färberisch den Zelltypen der Langerhansschen Inseln anderer Wirbeltiere entsprechen. Die A-Zellen enthalten gröbere Granula, die sich nach Voroxydation mit Azokarmin und mit Malloryschem sauren Phosphorwolframhämatoxylin anfärben, bei der Nachfärbung mit Trichrom ponceaurot. Die B-Zellen enthalten feinere Granula, die sich mit Aldehydfuchsin anfärben, eine positive Zink-Reaktion (Dithizon-Reaktion) geben und positiv metachromatisch mit Pseudoisocyanin reagieren. Die D-Zellen färben sich mit Aldehydfuchsin-Ponceau-Lichtgrün schmutzig grün. 3. Auch elektronenmikroskopisch lassen sich eindeutig drei Typen von endokrinen Zellen differenzieren, die in ihren Grundzügen den Zelltypen bei anderen Wirbeltieren entsprechen. Am charakteristischsten für die Differenzierung sind die Granula. Die α-Granula sind kugelförmig oder leicht ovoid, ihre osmiophile Substanz ist gekörnt und füllt fast das ganze Granulum aus. Die β-Granula fallen durch die bedeutende Polymorphie ihrer osmiophilen Substanz auf. Die δ-Granula sind überwiegend Gebilde mit einer fein gekörnten, geringer osmiophilen Substanz oder kleinen Vakuolen. 4. Innerhalb der soliden Inseln beobachteten wir zahlreiche feine und feinste Ausführungsgänge. 5. Die „Inseln“ begleiten die Ausführungsgänge. Die Balken der endokrinen Zellen hängen auf diese Weise untereinander auf verhältnismäßig große Entfernung zusammen. 6. Der Bau des endokrinen Gewebes des Pankreas der Schildkröten ist verhältnismäßig primitiv. Er bildet den Übergang zwischen dem Inselapparat der Selachier und der höheren Wirbeltiere.

Light- and electron-microscopic studies of the pancreas of Testudo graeca and Emys orbicularis L. show, that the organ contains two types of islets: (a) solid islets, which are formed by extensive accumulations of endocrine cells; and (b) scattered endocrine cells, some of which are seen in the region of larger ducts. The majority of these cells occur, however, in the exocrine tissue near the basement membrane of the small ducts. 2. With the help of specific staining methods it is possible to distinguish light-microscopically up to three different cell types in the solid islets, which — as judged by their appearance — correspond to the cells observed in the islets of Langerhans of other vertebrates. The A-cells contain coarse granules which stain with azo-carmine and Mallory's phosphotungstic hematoxylin (after oxidation); they also stain with trichromic Ponceau red. B-cells contain fine granules, which stain with aldehyde fuchsin; they give positive results with the dithizone test for the determination of Zn; and they react metachromatically with pseudo-isocyanine. D-cells have a dull-green appearance after treatment with aldehydefuchsin-Ponceau-light green. 3. Also electron-microscopically three types of endocrine cells are distinguishable which in their characteristics correspond to the cell types of other vertebrates. The most characteristic feature is the granules. The α-granules are spherical or slightly ovoid, their osmiophil substance is granular and practically fills the whole granule. The β-granules impress by the polymorphism of their osmiophil substance. δ-granules are structures which exhibit a finely granular but less osmiophil substance or small vacuoles. 4. Within the solid islets numerous small ducts are observed. 5. The islets “accompany” the excretory ducts. Thus the cords of endocrine cells are connected with each other over comperatively long distances. 6. The structure of the endocrine part of the pancreas of the tortoise is comparatively primitive. It is to be regarded as a transistory stage between the islets apparatus of the selachians and that of the higher vertebrates.

Die Langerhansschen Inseln liegen vorwiegend in mittleren Abschnitten des Pankreas. Die Achse der Insel bildet eine Kapillare, um welche die endokrinen Zellen radiär angeordnet sind. 3. Lichtmikroskopisch unterscheidet man mit Sicherheit aufgrund der Färbbarkeit der Zellen bzw. ihrer Granula A- und B-Zellen. Die A-Zellen pflegen zumeist höher zu sein; am Kapillarpol sind sie oftmals verschmälert. Zwischen die A-Zellen sind niedrigere und breitere B-Zellen eingefügt. Im Gesamtbild herrschen Zellen mit färberischen Eigenschaften der A-Zellen vor. Die Inselzellen unterscheiden sich eindeutig von den zymogenen Zellen durch die Größe der Nukleolen. In den Inseln fanden wir weder kleine Ausführungsgänge noch andere Lumina. 4. Elektronenmikroskopisch fanden wir drei Zelltypen, die sich in ihrem Aussehen und in der Größe der Granula unterscheiden. Zum Teil hängt die Struktur der Zellen von der Art der Fixation ab. Die A-Zellen enthalten die kleinsten und dichtesten Granula (Durchmesser der α-Granula 193±37 nm, der β-Granula 250±56 nm). Nach OsO4Fixation wird die „osmiophile “ Substanz der β-Granula durch einen sternförmigen Komplex von zarten Fäserchen repräsentiert. Regelmäßig enthält das Cytoplasma der B-Zellen eine kleine Menge von Glykogen. Die Fixation beeinflußt am stärksten das Bild der D-Zellen. Während nach OsO4 Fixierung von den meisten Granula nur die Hüllmembranen erhalten bleiben, sind die Körnchen nach Glutaraldehyd-OsO4-Fixation gut erhalten. Die Granula der D-Zellen sind am größten (Durchmesser 277±53 nm), weniger dicht und unregelmäßiger geformt als die der A-Zellen. Das Cytoplasma der D-Zellen enthält ebenfalls eine geringe Menge von Glykogen.
    Notes: Summary 1. The islets of Langerhans in two species of lizards — Lacerta agilis and Lacerta viridis — were studied light- and electron microscopically. 2. The islets prefer in their distribution the middle part of the gland, the endocrine cells are arranged as a epithelial layer around capillaries. 3. By light microscopy two different cell types — A- and B-cells — could be distinguished according to their staining reactions. A-cells are elongated elements; one pole extends to the capillary. The B-cells are cuboidal and less frequent. Islet cells are easy to differentiate from acinar cells by the size of their nucleolus. In lizards no combination of smaller ducts with the islets like in other reptiles was observed. 4. With the electron microscope 3 different cell types were detected according to the size, form and electron density of their granules. A cells contain the smallest granules with highest electron density, their mean diameter is 193±37 nm. The osmiophilic content of the β-granules appears as a starlike complex of small fibers, the diameter of the β-granules is 250±56 nm. The cytoplasma of B-cells contains frequently small quantities of glycogen. The appearance of the third cell type — the D-cells — varies considerably with the fixation method applied: osmium-tetroxid preserves only the membrane of the granules, which appear as empty vacuoles. Glutaraldehydfixation reveals a faintly osmiophilic content in the granules, which is less electron dense than that of the α-granules. δ-granules are the largest of all 3 cell types (mean diameter 277±53 nm), their cytoplasm contains small amounts of glycogen.

    Notes: Summary The ultrastructure of crystalline beta granules of the islets of Langerhans in the alligator has been investigated. From optical diffraction analysis and serial sectioning, the existence of four distinct types of crystalline inclusions was established in ultrathin sections. The first type is the most frequent and is interpreted as a rhombohedroni with a base, the ortho-hexagonal unit-cell edges being a=18.9 nm, c=23.0 nm. The second type of crystal (not observed in serial sections) is found compatible with a rhomb-dodecahedron which indexes on a cubic cell with a=9.6 nm. The third type of crystal was assigned to dipyramids. Dipyramids are extremely rare, and only two diffraction patterns were obtained; their crystal system could not be determined. Prisms, which are second in abundance, represent the fourth type of crystal. Spacings as well as the symmetry differ from those of the above three crystal types and indicate a tetragonal cell with a=4.2 nm, c=14.2 nm. The data for the prismatic crystals are strikingly similar to those of proinsulin and may represent the first case of agreement between crystals (i) formed in vitro and studied by X-ray diffraction and (ii) those investigated in situ by electron microscopy.

    Notes: Summary Optical diffraction analysis was carried out on crystalline inclusions in the rough endoplasmic reticulum of the insulin and somatostatin cells in the islet organ of the hagfish. A striking difference in crystalline arrangement was observed between the inclusions of the insulin and somatostatin cells. The crystallographic arrangement of the inclusions observed in situ in the insulin cells differed from that previously found by means of X-ray diffraction analyses of hagfish insulin crystals formed in vitro.
