    Notes: Summary A choline-requiring strain of Torulopsis pintolopesii when growing on choline-methyl-14C or choline-methyl-3H excretes the radioactivity incorporated in the first 24–48 h of incubation up to ca. 85–95% of the radioactivity added at the beginning of the incubation. The addition of non-radioactive methionine did not interfere with the uptake and excretion of radioactivity from choline-Me-14C. Radioactive methyl group of methionine previously incorporated by growing cells of T. pintolopesii on varying concentrations of choline was not excreted in appreciable amounts. No evidence was obtained for the oxidation of choline to betaine, degradation to trimethylamine, or net incorporation of labelled choline into lecithin. The occurrence of a new pathway for the utilization of choline in yeasts is suggested. The requirement of choline by T. pintolopesii is explained tentatively by the formation and excretion of a compound containing the carbon and hydrogen atoms from the methyl groups of choline and whose chemical structure still under study, may comprise a heteroside containing mannitol as the polyhydroxylated moiety.

    Notes: Summary The chemical composition of cell walls from choline-less Torulopsis pintolopesii grown with choline or with methionine was studied. Methioninegrown cells synthesized a weakened cell wall compared to normal choline-grown yeast. The ethylenediamine fractionation procedure yielded three fractions—A, B, and C—with different solubilities. Glucose and mannose were detected in hydrolysed unfractionated cell walls from yeasts grown under both conditions as well as in all fractions. Glucose content was greater in fractions B and C from methioninegrown cells; the mannose content was about the same. Walls from choline-grown cells (W c ) had 25% more protein than walls from methionine-grown cells (W m ). The amino acid composition of the proteins of W c and W m was not qualitatively altered. Seventeen amino acids were identified; glutamic and aspartic acids and valine predominated. W c had 3.5 times more lipid than W m . The amount of phosphorus was the same. Yeasts grown on methionine synthesized more ergosterol than choline-grown cells. The rate of formation of spheroplasts was higher in methionine-grown cells. Rates of incorporation of adenine, glutamic acid, and uracil were similar in cells grown on methionine or choline; incorporation of phenylalanine and tyrosine was depressed in methionine-grown cells.

    Notes: . Cross-reactivity between fungal and Trypanosoma cruzi polysaccharides, owing to common residues of β-D-galactofuranose, β-D-galactopyranose, and α-D-mannopyranose, was demonstrated by using a) rabbit immune sera against T. cruzi epimastigotes and b) sera from patients with Chagas’disease. Several chagasic (Ch) sera precipitated partly purified galactomannans from Aspergillus fumigatus and from T. cruzi epimastigotes and also the galactoglucomannan from Dactylium dendroides. Reaction of one Ch serum with T. cruzi galactomannan (GM) was completely inhibited by synthetic β-D-Galf-(1 → 3)-Me α-D-Manp, and that of another Ch serum with a purified D. dendroides galactoglucomannan (GGM) was partly inhibited by (1 → 6)-linked (81%) or by (1 - 3)-linked (33%) β-D-Galf-Me α-D-Manp. The β-D-Galf-(1 → 3)-α-D-Manp epitope was present in both T. cruzi and D. dendroides polysaccharides. Rabbit anti-T. cruzi antisera precipitated A. fumigatus GM, T. cruzi antigenic extracts containing the lipopeptidophosphoglycan (LPPG), T. cruzi alkali-extracted GM, a synthetic GM, and D. dendroides GGM. Weak reactivities were obtained for a Torulopsis lactis-condensi GM containing β-D-Galp terminal residues and for baker's yeast mannan with α-D-Manp-(1 - 3)-α-D-Manp-(1- → 2)-α-D-Manp-(1 → 2) side chains. An anti-LPPG rabbit serum precipitated D. dendroides GGM—a reaction inhibited (82%) by β-D-Galf-(1 → 3)-Me α-D-Manp and, less efficiently, by a (1 → 5)-linked β-D-Galf-tetrasaccharide. Sera from mice immunized with D. dendroides whole cells reacted with CL-strain trypomastigotes as shown a) by indirect immunofluorescence, b) by a Staphylococcus adherence test, but were not lytic. Mice immunized with D. dendroides were not protected against a challenge with virulent T. cruzi trypomastigotes.

    Notes: SYNOPSIS. Cells of the insect flagellate Crithidia fasciculata contained mannan and arabinogalactan components, whose porportion varied with culture age, the former predominating during early stages, and the latter during the later stages of exponential growth and the deceleration phase. The mannan was a β-D-(1→2)-linked D-mannopyranan. The arabinogalactan had a complex structure containing, in part, a β-D-(1→-3)-linked galactopyranose main-chain substituted in the 2 positions by single-unit D-arabinopyranose side-chains and with some unsubstituted units.

    Notes: The 43 kDa glycoprotein (gp43) of Paracoccidioides brasiliensis is the major diagnostic antigen of paracoccidioidomycosis (PCM), a prevalent fungal infection in South America. A 15-mer sequence from gp43, denominated P10, induced T-CD4+ T helper 1 cellular immune responses in mice of three different haplotypes and protected against intratracheal challenge by a virulent isolate of P. brasiliensis. In an attempt to improve delivery of P10, a promiscuous antigen also presented by human leucocyte antigen-DR alleles, aiming at immunotherapy, we synthesized a multiple antigen peptide with the protective T-cell epitope expressed in a tetravalent 13-mer analog of P10 (M10). M10 induced specific lymph node cell proliferation in mice preimmunized with peptides in complete Freund's adjuvant (CFA). In addition, M10 immunization without CFA significantly protected intratracheally infected mice. We conclude that M10 is a candidate for an anti-PCM vaccine. In this report we describe: (1) the synthesis of M10; (2) the induction of M10-elicited T-cell response and (3) in vivo protection of mice immunized with M10 and challenged by a virulent strain of P. brasiliensis.

    Notes: Abstract Vitamin requirements, determined by two different methods, and other nutritional characteristics of 46 psychrophobic and 30 non-psychrophobic yeasts were investigated. Vitamin requirements and induced nutritional imbalances emerged as criteria for definingCandida slooffii. Strains of this species behaved uniformly. Variations in vitamin requirements of strains ofSaccharomyces telluris, Torulopsis bovina andTorulopsis pintolopesii were confirmed. The choline requirement exhibited by several strains (by as many as 86% inS. telluris) was evident on a simple defined medium without methionine, but not on Wickerham's vitamin-free basal medium. The group of choline-requiring strains comprised strains responding similarly to choline or methionine, and some strains growing with choline but not with methionine on short incubation. Other frequently required vitamins were biotin, pantothenate, thiamine, nicotinic acid, pyridoxine, and inositol.
