    Notes: Abstract Development of an echinothurid sea urchin is described for the first time; eggs of Asthenosoma ijimai (Yoshiwara) have been fertilized in the laboratory, and development has been followed light-microscopically up to the early juvenile stage. The eggs, which are orange and float, are 1.2 mm in diameter, by far the largest echinoid eggs on record. The embryos, larvae and early juveniles are lecithotrophic, and no exogenous food is needed for development. The embryonic stages include a remarkable biscuit-shaped late blastula, which has never been described for any echinoid before. The larva, with its para-arms unsupported by skeletal ossicles, is unlike the echinoid prism or pluteus and more resembles the early bipinnaria or early auricularia of asteroids and holothurians, respectively. All stages through late larvae float just beneath the surface until settlement occurs during the third and fourth weeks at 20°C. Metamorphosis from late larva to juvenile is gradual and no part of the larval body appears to be cast off.

    Notes: Abstract. Three lines of evidence indicate that arginine-46 (R46) and arginine-275 (R275) are essential to the function of UhpT, the Pi-linked antiport protein of Escherichia coli. A role for arginine was initially suggested by the sensitivity of UhpT to inhibition by 2,3-butanedione, an arginine-directed probe. Since the presence of substrate protected against this inhibition, this work further suggested that arginine(s) may lie at or near the UhpT active site. In other work, each UhpT arginine was examined individually by using site-directed mutagenesis to generate a cysteine or a lysine derivative. With two exceptions (R46, R275), all arginines could be replaced by either cysteine (10 of 14 residues) or lysine (12 of 14) without loss of function, implicating R46 and R275 as essential to UhpT function. This idea was strengthened by examining a multiple alignment of the eleven known UhpT-related proteins (≥30% identity). That alignment showed R46 and R275 were two of the only three arginines strongly conserved in this group of proteins. Considered together, these different approaches lead us to conclude that UhpT and its relatives have only two arginine residues (R46, R275) whose presence is essential to function. Prior biochemical work had placed R275 at the external entrance to the translocation pathway, and a symmetry argument emerging from the multiple alignment suggests a similar position for R46. Accordingly, by virtue of their locations at the entrance to this pathway, we speculate that R46 and R275 function in establishing substrate specificity.

    Notes: Abstract We performed molecular genetic analyses of the family of a boy suffering from chronic granulomatous disease (CGD) after immunocytochemically confirming him and his mother to be an X-linked CGD patient and a mosaic carrier, respectively. Southern blot hybridization using cDNA for the cytochrome b558 heavy chain gene (CYBB) as a probe showed that the patient had a deletion in the 5′ region of the CYBB and his phenotypically normal mother was heterozygous for this deletion. Polymerase chain reaction analyses of all 13 exons of the patient’s CYBB gene demonstrated that the deletion extends from exon 7 or neighboring introns to 5′ upstream. The length of the deletion was determined by pulsed-field gel electrophoresis and Southern blotting of genomic DNA using CYBB cDNA and the genetic marker pERT55-5, centromeric to CYBB, as probes. Both probes recognized common SfiI-NotI fragments of 120 kb and 95 kb in normal individuals and the patient, respectively. These results revealed that the patient has a 25-kb deletion spanning from the middle of CYBB to 5′ upstream. This is the only report of a large 5′ deletion in CYBB and also the first observation that CYBB and pERT55-5 are within 120 kb in Xp21.

    Notes: Abstract In the progeny of a trisomic type for chromosome 6, Purple, a 16-chromosome type was obtained, which had a pair of new metacentric chromosome 6 in excess. The new metacentric chromosome 6 was shorter than any of the 14 chromosomes of normal barley complement and showed a heteropycnotic nature at late prophase in somatic mitosis. At metaphase I in the plants with 14+one metacentric chromosome 6 (2n=15) the chromosome configuration was exclusively 7II+1I indicating that the extra metacentric chromosome 6 could not associate with the normal chromosome 6. At diakinesis and metaphase I in the new 16-chromosome plants most of the sporocytes showed 8IIor 7II+2I. Neither tetravalents nor trivalents were observed at meiosis. The chromosome behaviour at anaphase I and later stages of meiosis was regular in general, resulted in a fairly high pollen fertility of about 61 per cent. Seed fertility however, was very low. The transmission rate of the new metacentric chromosome 6 through the pollen was extremely low in 16-chromosome plants. Possible origin of new basic number and B-chromosome in diploid level through trisomic condition was suggested (Summary see p. 138).

    Notes: Summary Microelectrodes were inserted into IIIrd cranial nucleus of anaesthetized rabbit. IIIrd nucleus was identified by observing the field potentials evoked antidromically by stimulation of IIIrd cranial nerve. After stimulation of VIIIth nerve extracellular field potentials, spike potentials in secondary vestibular fibers, and postsynaptic potentials in IIIrd nucleus neurones were recorded. VIIIth nerve impulses either excite or inhibit IIIrd nucleus neurones postsynaptically with disynaptic latencies around 1.7 msec. By local stimulation of the medulla, it was found that the secondary vestibular impulses inhibiting IIIrd nucleus neurones are mediated by the superior nucleus. The excitatory impulses are relayed by the rostral half of the medial nucleus as well as a certain structure(s) relevant to the brachium conjunctivum. Preliminary pharmacological investigations on the inhibition of IIIrd nucleus neurones are reported.

    Notes: Abstract Stretch induced activation and release induced deactivation of single glycerol-extracted insect flight muscle fibres were investigated. The results are interpreted to indicate that the muscle length controls the number of acting cross bridges, whereas their attachment-detachment kinetics in mainly determined by the state of strain of the cross bridges. It is concluded that the net detachment rate of the cross bridges is enhanced if the muscle is released thereby “unloading” the cross bridges. This behaviour of the unloaded cross bridge is a basic postulation of most of the molecular muscle contraction models. 1. The delayed tension rise induced by stretches of different amplitudes could be restored to the level before the stretch by a release to the initial length. 2. The delayed tension decrease induced by a release of moderate (up to δL=1.5% L i)amplitude is quantitatively restored within the delayed increase induced by the restretch to the initial length. 3. Stiffness, which decreased during the delayed tension drop after release, is restored during a delayed stiffness increase effected by a restretch to the initial length. 4. The rate and the extent of the stiffness drop after release increased with increasing amplitude of the release and with increasing temperature. 5. After the deactivation, i.e., after tension and stiffness achieved a new steady level after the release, the attached cross bridges are already in the same state of strain as they were before the release. This finding is interpreted to indicate that within the deactivation phase all cross bridges attached prior the release are replaced by cross bridges attached after the release. 6. The rate of tension and stiffness decay after release does not depend on the absolute muscle length but on the amplitude of the release which induced the deactivation.

    Notes: Abstract A probabilistic model of a flow of fluid through a random medium,percolation model, provides a typical example of statistical mechanical problems which are easy to describe but difficult to solve. While the percolation problem on undirected planar lattices is exactly solved as a limit of the Potts models, there still has been no exact solution for the directed lattices. The most reliable method to provide good approximations is a numerical estimation using finite power-series expansion data of the infinite formal power series for percolation probability. In order to calculate higher-order terms in power series, Baxter and Guttmann [6] and Jensen and Guttmann [33] proposed an extrapolation procedure based on an assumption that thecorrection terms, which show the difference between the exact infinite power series and approximate finite series, are expressed as linear combinations of the Catalan numbers. In this paper, starting from a brief review on the directed percolation problem and the observation by Baxter, Guttmann, and Jensen, we state some theorems in which we explain the reason why the combinatorial numbers appear in the correction terms of power series. In the proof of our theorems, we show several useful combinatorial identities for the ballot numbers, which become the Catalan numbers in a special case. These identities ensure that a summation of products of the ballot numbers with polynomial coefficients can be expanded using the ballot numbers. There is still a gap between our theorems and the Baxter-Guttmann-Jensen observation, and we also give some conjectures. As a generalization of the percolation problem on a directed planar lattice, we present two topics at the end of this paper: The friendly walker problem and the stochastic cellular automata in higher dimensions. We hope that these two topics as well as the directed percolation problem will be of much interest to researchers of combinatorics.

    Notes: The elastic properties and their pressure dependence of four B1-type alkaline earth oxides, MgO, CaO, SrO, and BaO, are calculated using the ab initio full-potential linear muffin-tin-orbital (FP-LMTO) generalized gradient approximated (GGA) method to elucidate their systematics. The calculated results agree quite well with the comparable experimental data. The large pressure dependence of c11 and c44 of MgO observed over 25 GPa is not predicted as well as the previous local-density approximation (LDA) calculations. It is inferred that the high-pressure measurement of elastic constant is quite sensitive to nonhydrostaticity. The deviation from the Cauchy relation and the elastic anisotropy are investigated. It is found that the interatomic interaction in SrO is nearest to the two-body force and the many-body contribution is largest in MgO. The elastic anisotropy in SrO and BaO are almost the same and only MgO has a large positive anisotropy under low pressure. The normalized elastic constants cij′ are introduced for more detail analysis. It is found that while there is no substantial difference in c11′ and c12′ of the four oxides, only c44′ and its pressure dependence vary systematically. The difference in the Cauchy deviation and elastic anisotropy of alkaline earth oxides under pressure is characterized by c44. © 2001 American Institute of Physics.
