Zusammenfassung Das TRH-Derivat wurde aus den geschützten Aminosäuren Z-L-Gln(Mbh)-OH,L-Phe-OMe und L-Pro-NH2 hergestellt. Aus dem Tripeptid Z-L-Gln(Mbh)-L-Phe-L-Pro-NH2 wurden die Schutzgruppen durch Kochen in Trifluoressigsäure/Anisol abgespalten. Die biologische Aktivität von TRH und dessen Derivat wurden indirekt durch die J125-Konzentration im Blut gemessen. 3 Std nachdem 0,5 µg TRH eingespritzt wurden, wurde ein Mittelwert von 67% gemessen und 8 Std danach einer von 29%. 8 Std nach Einspritzen von 0,5 µg TRH-Derivat konnte nur eine geringe Zunahme festgestellt werden (9%). 3 Std nach Injektion von 10 µg TRH stieg die J125-Konzentration auf 121% und 8 Std danach auf 70% an. Die entsprechenden Werte für das TRH-Derivat betrugen 53 bzw. 9%. Es scheint also, daß das Derivat unter den gegebenen Bedingungen fast die Hälfte der biologischen Aktivität des TRH hat.

Summary The TRH derivative was prepared from the protected amino acids Z-L-Gln(Mbh)-OH,L-Phe-OMe and from L-Pro-NH2. From the tripeptide derivative Z-L-Gln(Mbh)-L-Phe-Pro-NH2 the protecting groups were cleaved off by boiling in trifluoroacetic acid/anisole. The biological activity of TRH and its derivative were measured indirectly as I125 concentration in the blood. Three hours after 0.5 µg TRH were injected a mean value of 67%, and 8 hours later of 29% was measured. Eight hours after injecting 0.5 µg TRH derivative only a slight increase (9%) was observed. The injection of 10 µg of TRH caused the I125 concentration to raise to 121% after 3 hours and to 70% after 8 hours. The corresponding values for the TRH derivative showed to be 53 respectively 9%. It seems therefore that the derivative has nearly half the biological activity of TRH for the conditions described here.
