    Description / Table of Contents: Résumé Sur 24 membres de cadavres la vascularisation du paquet adipeux antérieur d'Hoffa a été étudiée par injection. Sur certains sujets, nous avons réalisé l'intervention décrite par Clancy (opération plastique de remplacement du ligament croisé antérieur par un tiers de tendon rotulien pédiculé à partir du paquet adipeux antérieur d'Hoffa). Il apparaît que ce paquet adipeux antérieur d'Hoffa est directement vascularisé par les artères articulaires supéro-et inféro-externes et internes, l'artères articulaires moyenne du genou par le ligament adipeux et l'artère récurrente tibiale antérieure. Ces vaisseaux forment également le réseau vasculaire du tissu conjonctif du tendon rotulien d'où ce tendon tire sa vascularisation. Au niveau des insertions sur le tibia et sur la rotule aucun vaisseau sanguin provenant de l'os ne pénètre le tendon. Le tiers médian du tendon rotulien pédiculé sur le paquet adipeux antérieur reste normalement irrigué après la préparation nécessaire à l'opération. La majorité des vaisseaux sanguins du paquet adipeux antérieur d'Hoffa pénètre le ligament à son point d'attache.
    Notes: Summary In 24 fresh specimens the blood-supply of the infrapatellar fat-pad was investigated by injection. In some of the specimens, Clancy's pedicled patellar tendon graft for anterior cruciate ligament reconstruction had been performed before injection. The fat-pad is supplied by vessels forming a plexus supplying the tendon. In the tendon grafts injected after Clancy's procedure, no vessels were found penetrating the patellar tendon at its patellar and tibial attachments. Filled vessels were found by histologic methods and the Spalteholz technique.

    Notes: Summary Contractile responses of isolated coronary arteries from healthy and alloxan-diabetic dogs to prostaglandin F2α were enhanced by indomethacin and inhibited by prostaglandin I2. The potentiation by indomethacin was more prominent in diabetic vessels than in normal arteries.

    Notes: Abstract The structures of bacterial communities were studied in activated sludge samples obtained from the aerobic and anaerobic zones of a wastewater treatment plant showing enhanced phosphorous removal. Samples were analyzed by in situ hybridization with oligonucleotide probes complementary to selected regions of the 16S and 23S ribosomal RNA (rRNA) characteristic for defined phylogenetic entities (genera and larger groups). The microbial community structures revealed by molecular techniques were compared with the compositions of culturable bacterial communities, obtained from the characterization of 255 isolates from tryptone-soy (TS) agar and R2A agar. These isolates were characterized by 89 physiological tests and their cellular fatty acid patterns, and identified. Culture-dependent techniques indicated the following distribution: different Aeromonas spp. (2.7–8.3% on R2A agar; 45.0–63.7% on TS agar), Acinetobacter spp. (5.4–9.0% on R2A agar; 5.0–9.1% on TS agar), Pseudomonas spp. (up to 10% on R2A agar) and Shewanella putrefaciens (up to 3.0% on R2A agar), all members of the gamma subclass of Proteobacteria, were isolated most frequently. The relatively rare isolates of the beta subclass were identified as Acidovorax spp., Alcaligenes spp., and Comamonas spp., The Gram-positive bacteria (high DNA G+C) were assigned mainly to Arthrobacter spp., Microbacterium spp., and Mycobacterium phlei. In order to assess the in situ abundance of the most frequently isolated genus, Aeromonas, two rRNA-targeted oligonucleotide probes were developed. The two gamma proteobacterial genera Aeromonas and Acinetobacter constituted less than 5% of all bacteria. In situ, Proteobacteria belonging to the beta subclass and high G+C Gram-positive bacteria were dominant. From filamentous bacteria, Sphaerotilus spp. and Leptothrix spp. could be detected occasionally. In addition, one sample contained a high proportion of the morphologically distinct filaments of Microthrix parvicella. As for the genus Acinetobacter, the relative abundance of the most frequently gamma-proteobacterial genus Aeromonas was overestimated by the intrinsic selectivity of cultivation. Cultivation on nutrient-rich medium (TS-agar) especially supported an enhanced isolation of bacteria belonging to these two genera.

    Notes: Abstract Hereditary multiple exostoses (EXT) is a genetically heterogeneous bone disorder caused by genes segregating on human chromosomes 8, 11, and 19 and designated EXT1, EXT2 and EXT3, respectively. Recently, the EXT1 gene has been isolated and partially characterized and appears to encode a tumor suppressor gene. We have identified six mutations in the human EXT1 gene from six unrelated multiple exostoses families segregating for the EXT gene on chromosome 8. One of the mutations we detected is the same 1-bp deletion in exon 6 that was previously reported in two independent EXT families. The other five mutations, in exons 1, 6, 9, and the splice junction at the 3′ end of exon 2, are novel. In each case, the mutation is likely to result in a truncated or nonfunctional EXT1 protein. These results corroborate and extend the previous report of mutations in this gene in two EXT families, and provide additional support for the EXT1 gene as the cause of hereditary multiple exostoses in families showing linkage to chromosome 8.

    Notes: Summary Using the polymerase chain reaction (PCR) technique, we have screened the DNA of 42 patients with Duchenne or Becker muscular dystrophy for deletions within the DMD gene. Two regions within putative deletion “hot spots” of this gene were tested, and deletions were found in 16.6% of patients. The oligonucleotide primers employed in this study initiate the amplification of exon sequences and were used to test the suitability and reliability of PCR in deletion screening and prenatal diagnosis using various numbers of cycles and artificial contamination ratios. We compared our approach with both “multiplex DNA amplification” and Southern blot analysis. A comparative evaluation of currently available techniques is presented.
