    Notes: There are several studies for developing web-based CAE applications to substitute existingCAE applications. CASOW is the web-based CAE system developed by the author. The approach forimplementation of the system is to use fundamental technologies, for instance, HTML, HTTP, CSSand so on, because we cannot know whether new technologies will be alive or not. Certaintechnologies bring specific advantages to the applications, but a life time of the application would beshort. Recently a great advantage of web interface is developed. Such a representative application isGoogle Maps which doesn’t require any special software components; however the applicationrealizes interactiveness of the application. The technology is called Ajax which drastically overcomesthe bad interactiveness on the web browser. On the other hand, high efficiency of analysis work withhigh performance computers produces many kinds of knowledge and a large amount of information.We are confronted with inefficient knowledge and information management in the network system.This paper presents the new web-based CAE system using Ajax and knowledge sharing frameworkwith compatibility to web interface. The system is presented, and advantages of the interface,effectiveness of the API in the system and knowledge sharing framework are discussed

    Notes: Fatigue crack growth under mixed mode loading conditions is simulated using S-FEM. Byusing S-FEM technique, only local mesh should be re-meshed and it becomes easy to simulate crackgrowth. By combining with auto-meshing technique, local mesh is re-meshed automatically, andcurved crack path is modeled easily. Plural fatigue crack problem is solved by this technique. Fortwo parallel crack problem, criteria of crack coalescence are proposed. By simulating this problem byS-FEM, it is verified these criteria are conservative ones

    Notes: Due to more complex and severe design restrictions, more effective and faster finiteelement analyses are demanded. There are several ways to compute FE analysis efficiently: parallelcomputing, fast iterative or direct solvers, adaptive analysis and so on. One of the most effectiveanalysis ways is the combination of adaptive analysis and multigrid iterative solver, because anadaptive analysis requires several meshes with difference resolutions and multigrid solver utilizessuch meshes to accelerate its computation. However, convergence of multigrid solver is largelyaffected by initial shape of each element. An effective mesh improvement method is proposed here. Itis the combination of mesh coarsening and refinement. A good mesh can be obtained by the method tobe applied to an initial mesh, and better convergence is achieved by the improved initial mesh

    Notes: A non-triangulation inserting particle method is presented to prevent numerical fractures ofSPH computations. The particles in those regions in which numerical fractures may occur are chosenat the beginning of SPH computations. And, the chosen particles are arrayed by the two different rulesrespectively according to the particle position relations. When the distance between each two chosenconnected particles exceeds a certain value and neither of the corresponding two particles hasfractured in SPH computations, a new particle is generated between those two ones and the positionrelation of particles is updated. The physical quantities that the new particle carries are obtainedaccording to some given rules. The method and the same type of method in the reference arecompared. The application of the method is analyzed. Conservation of mass and Conservation ofmomentum of the whole system are kept in this method. Several examples are given to validate theefficiency of the method to prevent numerical fractures

    Notes: Aim: To investigate the long-term efficacy of postoperative interferon-α (IFN-α) adjuvant therapy in preventing recurrence in non-metastatic renal cell carcinoma treated with radical nephrectomy and to identify related prognostic markers.Methods: Long-term follow-up was conducted to study rates of survival and non-recurrence in 88 subjects following radical nephrectomy for non-metastatic disease.Results: The overall survival rate was 90% at 5 years and 88% at 10, with corresponding non-recurrence rates of 81% and 74%. Survival rates reviewed by preadministration pT stage showed a falling tendency from T1 through to T3 in line with pathological progression; when cases at stage pT1b or below were compared with those at stage pT2 or above, the latter showed a tendency to lower survival rates (P = 0.0966, Breslow-Gehan-Wilcoxon). Similarly, non-recurrence rates tended to fall in line with pathological progression, with a significant difference found in the comparison of cases at stage pT1b or below with those at stage pT2 or above (P = 0.0265, log–rank, Mantel-Cox). Duration of IFN-α administration showed a tendency to positive correlation with long-term survival (P = 0.3765, Breslow-Gehan-Wilcoxon). Non-recurrence rate was not found to differ according to duration of administration. Comparison of groups with normal and abnormal preadministration immunosuppressive acidic protein values showed that the normal group tended to have higher rates of survival and non-recurrence (P = 0.3371, Breslow-Gehan-Wilcoxon).Conclusions: Immunosuppressive acidic protein values appear to be a useful predictive marker for recurrence. A randomized trial, examining long-term outcome according to tumor stage and variables such as duration of administration, dose, administration time, and dosing schedule is required.

    Notes: Background: The presence of proliferating cell nuclear antigen (PCNA) has been suggested as a more important prognostic marker than either grade or mitotic in the prognosis of patients with renal cell carcinoma. We assessed the immunoreactivity of PCNA in primary lesions and pulmonary metastases from patietns with renal cell carcinoma and correlated the results with various histopathologic features and prognostic factors. Methods: We studied the relationship between PCNA expression and clinical prognostic factors from resected primary lesions and pulmonary metastases from 10 patients and primary lesions from 32 patients with renal cell carcinoma without metastases. The cells were immunohistochemically stained with PCNA monoclonal antibody (PC-10) and 1000 nuclei were counted. The results were expressed as a ratio of stained to total cells (PCNA labeling index, LI |X%). Results: The PCNA LI of pulmonary metastatic nuclei was significantly higher than the PCNA LI of renal lesions either from patients with (P 〈 0.05) or without (P 〈 0.01) metastases. Also, the mean PCNA LI of the pulmonary lesions in patients dying within 3 years of diagnosis was higher than the mean PCNA LI of patients surviving greater than 3 years. Conclusion: Our findings suggest that the PCNA LI, which was determined by immunohistochemical analysis, is an important market reflecting the biologic behavior of renal cell carcinomas. The degree of PCNA expression in this study was of prognostic significance.

    Notes: Abstract Osteocalcin (OC) is a major noncollagenous bone protein whose expression is limited almost exclusively to osteotropic tumors and mature calcified tissue (differentiated osteoblasts). The function of OC, a highly conserved gamma-carboxyglutamic acid-containing protein, relies in part on its ability to bind hydroxyapatite and act as a chemoattractant for bone-resorbing cells. Serum osteocalcin levels are used clinically as an index of active bone turnover. Research in our laboratory has revealed that OC is expressed in several solid tumors, including osteosarcoma and ovarian, lung, brain, and prostate cancers. Evidence arising from reverse-transcription polymerase chain reaction (RT-PCR; detection of OC mRNA), immunohistochemical staining (detection of OC protein), and transient transfection and reporter assay (detection of OC mRNA transcription) reveals that OC expression is up-regulated in numerous solid tumors, with its expression being further elevated in androgen-independent prostate cancers. A recombinant, replication-defective adenovirus, Ad-OC-TK (OC promoter-driven herpes-simplex-virus thymidine kinase) was constructed and, when combined with the appropriate prodrug, either ganciclovir (GCV) or acyclovir (ACV), was found to be effective at destroying prostate-cancer cell lines in vitro and prostate tumor xenografts in vivo in both subcutaneous and bone sites. Additionally, via use of the OC promoter the supporting bone stromal cells are cotargeted when the prostate cancer interdigitates with bone stroma at the metastatic skeletal sites. Thus, maximal tissue-specific cell toxicity is achieved by the interruption of cellular communication between the prostate cancer and the bone stroma. We describe herein the preclinical foundation as well as the design and implementation of an ongoing phase I clinical trial at the University of Virginia that targets androgen-independent metastatic prostate cancer using the Ad-OC-TK vector.

    Notes: MG (multigrid) method is one of the most promising solvers for large scale problems. Hexahedral mesh generation and its adaptation are not enough to use for practical applications, because its mesh generation is very difficult and still labor intensive work by hand. We have developed hexahedral local refinement technique controlled by posterior error estimation. We have proposed a MG technique for unstructured hexahedral meshes with local mesh refinement. In this paper, the proposed technique is evaluated to check its performance and severe analyses of bending cantilevers. Performance of MG for unstructured hexahedral meshes is compared with that of the PCG (preconditioned conjugate gradient) through several benchmark examples of 3-D static elastic analysis. Proposed MG is faster than PCG for all problems as number of freedoms increases. Finally limitation of the proposed technique is presented
