    Notes: Abstract Several mutant hTNFα genes were constructed by deletion and stepwise reconstitution of regions coding for C-terminal sequences. The mutant hTNFα proteins behaved differently from native hTNFα when expressed in Escherichia coli. They were either sensitive to proteolytic degradation or formed insoluble aggregates depending on the strains and conditions used for expression. By contrast, native hTNFα was always present in a soluble form and had a tendency to associate with the cytoplasmic membrane. It was even transported to the periplasmic space in E. coli as shown by both cell fractionation and immunoelectron microscopy. The different behaviour of mutant hTNFα proteins probably results from a disturbance of protein folding.

    Notes: Abstract Post-embedding labelling techniques with colloidal gold-IgG or -protein A complexes were used to determine the subcellular location of IFNα1 and staphylokinase secreted from Bacillus subtilis GB500 cells. Both proteins were present in the cytoplasma and the cell enveloped pointing to a posttranslational mode of translocation across the cytoplasmic membrane. 5- to 10-fold higher concentrations of gold particles per 0.1 μm2 were found on the cell enveloped and clustering was observed suggesting preferential regions for secretion sites. Several control experiments ensured the specificity of the labelling data.

    Notes: A sialic acid-specific lectin was isolated from the albumin glands of the garden snail Cepaea hortensis by affinity chromatography on fetuin-Sepharose following gel filtration on Superdex 200. The purified native lectin showed a molecular mass of about 95 kDa by gel filtration and 100 kDa by SDS electrophoresis. It was cleaved by boiling in buffer containing SDS in three serological identical bands corresponding to molecular masses of about 24, 20 and 16 kDa, respectively. From these three fragments, only the 24- and the 20-kDa bands were found to be glycosylated. Only the three sugars mannose, galactose and N-acetylglucosamine could be detected in a molar ratio of 3:8.6:2. The oligosaccharide moieties seem to be N- and partially O-glycosidic bound. Isoelectric focusing (IEF) of the purified lectin revealed a heterogeneous pattern with bands in the pH range of 4.3–5.0. Isolated bands of different isoelectric points showed in SDS electrophoresis the same three fragments with molecular masses of 24, 20 or 16 kDa. The heterogeneity of the lectin was revealed either by IEF or amino acid sequencing of internal tryptic peptides.

    Notes: The localisation of phosphate-starvation-induced phosphodiesterase PhoD from Bacillus subtilis was studied by analysing processing, release and immunogold labelling of the sections. Although the processing of the pre-protein was extremely slow, the major fraction of PhoD could be detected at the surface of the cell wall. The results indicate that inefficient processing of the translocated pre-protein keeps PhoD in a cell wall-associated location. The uncleaved signal peptide might function as a membrane anchor.

    Notes: A potent basic superantigen (designated streptococcal pyrogenic exotoxin X, SPEX/SMEZ3) was purified to homogeneity from culture supernatants of a Streptococcus pyogenes scarlatina strain of type 12 (genotype speA−, speC−) and characterized. Sequence alignments revealed SPEX to be an allele of the streptococcal mitogens type Z (SMEZ). The N-terminal amino acid sequence of SPEX was found with LEVDNNSLLR to be identical to the recently described acidic superantigen SMEZ. Although SPEX/SMEZ genes were present in all of the streptococcal strains tested, a toxin production could only be detected in a small number of strains. The produced toxin concentration in the culture supernatants of positive strains differed between 0 and 20 ng ml−1. The purified SPEX stimulated human T-lymphocytes with Vβ8 specificity at extremely low concentrations (lower than 100 pg ml−1).

    Notes: Abstract Lipopolysaccharides (LPS) of Salmonella djakarta and Salmonella isaszeg, as well as of a spontaneous R-mutant of S. djakarta were investigated as to their content in neuraminic acid (Neu) and its individual linkage. The two Salmonella serovars both belong to the O:48 serogroup of Salmonella, but to two different subgroups. LPS of both S-forms contained high amounts of Neu, although in different quantities, whereas the R-form was completely devoid of it. Methylation analysis indicated that Neu is exclusively terminally linked in S. djakarta whereas both terminal and 4-linked Neu were recognized in S. isaszeg. Although terminally linked, a sialidase from Arthrobacter ureafaciens was unable to split Neu even after prolonged incubation from both S-type LPSs. When LPS was first treated by mild alkali, however, the total amount of Neu from S. djakarta LPS and about 50% from that of LPS of S. isaszeg could be removed. In contrast, alkali-treated LPS, but also the non-treated one, proved to be effective inhibitors for a sialic acid-binding lectin from Cepaea hortensis. The resistance of terminal Neu towards sialidase may be due to the presence of an O-acetyl group which would be removed during the methylation analysis but would, especially when linked to C-4, not interfere with the reactivity of the lectin.

    Notes: Abstract In a Rheotens experiment, the tensile force needed for elongation of an extruded filament is measured as a function of the draw ratio. For thermo-rheologically simple polymer melts, the existence of Rheotens-mastercurves was proved by Wagner, Schulze, and Göttfert (1995). Rheotens-mastercurves are invariant with respect to changes in melt temperature and changes in the average molar mass. By use of purely viscous models, we convert Rheotens-mastercurves of a branched and a linear polyethylene melt to elongational viscosity as a function of strain rate. The resulting elongational viscosity from constant force extension experiments is found to be in general agreement with what is expected as steady-state viscosity of polyethylene melts measured in either constant strain-rate or constant stress mode.

    Description / Table of Contents: Summary The authors report on the immuno electron microscopic demonstration of Concanavalin A-receptors on the erythrocyte membrane. Immuno freeze-etching studies evidenced a focal arrangement of Concanavalin A-receptors on intact erythrocytes as well as on erythrocyte ghosts. Obviously hemolysis by distilled water does not produce changes in the pattern of distribution of Concanavalin A-receptors whereas slight aggregation of plasma membrane intramembraneous particles occured. No unambigous spatial relationship between Concanavalin A-receptors and the plasma membrane intramembraneous particles (intrinsic proteins) of erythrocytes could be observed.
    Notes: Zusammenfassung Es wird über den immun-elektronenmikroskopischen Nachweis der Concanavalin A-Rezeptoren der Erythrozytenmembran berichtet. Die Immun-Gefrierätzungsuntersuchungen zeigten, daß die Concanavalin A-Rezeptoren sowohl der intakten Erythrozyten als auch der Erythrozytenschatten herdförmig verteilt sind. Die Aqua dest.-Hämolyse führt offenbar zu keiner Umverteilung der Concanavalin A-Rezeptoren, während eine geringfügige Aggregation der intramembranösen Partikeln der Plasmamembran eintrat. Eine eindeutige räumliche Beziehung der Concanavalin A-Rezeptoren zu den intramembranösen Partikeln (intrinsic proteins) der Plasmamembran der Erythrozyten konnte nicht beobachtet werden.

    Notes: Abstract By generalizing the Doi-Edwards model to the Molecular Stress Function theory of Wagner and Schaeffer, the extensional viscosities of polyolefin melts in uniaxial, equibiaxial and planar constant strain-rate experiments starting from the isotropic state can be described quantitatively. While the strain hardening of four linear polymer melts (two high-density polyethylenes, a polystyrene and a polypropylene) can be accounted for by a tube diameter that decreases affinely with the average stretch, the two long-chain-branched polymer melts considered (a low-density polyethylene and a long-chain branched polypropylene) show enhanced strain hardening in extensional flows due to the presence of long-chain branches. This can be quantified by a molecular stress function, the square of which is quadratic in the average stretch and which follows from the junction fluctuation theory of Flory. The ultimate magnitude of the strain-hardening effect is governed by a maximum value of the molecular stress, which is specific to the polymer melt considered and which is the only free non-linear parameter of the theory.
