    Notes: Conclusion It is clear that calcium ions are of considerable importance as a second messenger in insulin secretion. There is increasing evidence that calmodulin, a ubiquitous intracellular regulatory protein that mediates calcium-dependent processes, has a fundamental role in stimulus-secretion coupling. Calmodulin is present in the B cell and the secretion of insulin is inhibited by phenothiazines which bind to and inhibit the action of calmodulin. The evidence strongly suggests that phenothiazines influence insulin secretion by their effect on calmodulin which probably mediates calcium-dependent insulin release. It seems likely that calmodulin acts at several points in stimulus-secretion coupling, influencing cyclic nucleotide metabolism, protein phosphorylation and exocytosis. The discovery of calmodulin and the increasing clarification of its roles in cellular metabolism represent major steps towards our understanding of the mechanisms which influence the secretion of insulin.

    Notes: Summary Previous studies demonstrating reduced plasma concentrations of ascorbic acid (AA) in diabetes and interactions between this vitamin and biochemical mechanisms such as synthesis of structural proteins, oxidative stress, polyol pathway and non-enzymatic glycation of proteins suggest that disturbed AA metabolism may be important in the pathogenesis of diabetic microangiopathy. However, limited information is available on the concentration of AA in tissues which develop diabetic complications. This study demonstrates reduced renal but not sciatic nerve or plasma AA concentration in two animal models of insulin-dependent diabetes mellitus, namely the STZ-diabetic rat and the spontaneously diabetic BB rat. Decreased lens AA concentration was also observed in STZ-diabetic rats. Improvement of glycaemic control by insulin treatment (albeit insufficient to achieve normoglycaemia) partially corrected lens and renal AA concentration in STZ-diabetic rats. AA treatment increased kidney and lens AA concentrations of STZ-diabetic and non-diabetic rats and corrected the abnormalities observed for untreated diabetic rats. Sciatic nerve AA concentration was not increased by AA treatment in any group. Tissue ratios of dehydroascorbic acid (DHAA)/AA, one index of oxidative stress, were not different between the diabetic and non-diabetic groups and were unaltered by AA supplementation. AA treatment of STZ-diabetic rats had no effect on elevated tissue concentrations of glucose, sorbitol and fructose or reduced myo-inositol concentration. The effect of reduced tissue AA levels in diabetes on either collagen synthesis or ability to combat increased free radical production is not known. However, correction of abnormal kidney and lens AA concentrations in experimental diabetes by AA supplementation suggests that if AA does have a role in the development or progression of the renal and ocular complications of diabetes, this treatment could be beneficial.     Notes: An Acton Research VM 521-SG UV monochromator has been equipped with an intensified diode array detector for use as a mid-UV spectrometer. 600 and 2400 gr/mm gratings are used in a turret assembly for either low-resolution surveys or high-resolution Doppler ion studies. The detector is a CsI coated MCP with a P-11 output phosphor. The 1024 diode array is scanned at a 1-MHZ video rate allowing a complete spectra to be obtained in 1.0 ms. System performance and preliminary operation on the OHTE plasma will be presented.

    Notes: A brief review of the history of laboratory testing of information retrieval systems focuses on the idea of a general-purpose test collection of documents, queries and relevance judgements. The TREC programme is introduced in this context, and an overview is given of the methods used in TREC. The Okapi team's participation in TREC is then discussed. The team has made use of TREC to improve some of the automatic techniques used in Okapi, specifically the term weighting function and the algorithms for term selection for query expansion. The consequence of this process has been a very good showing for Okapi in terms of the TREC evaluation results. Some of the issues around the much more difficult problem of interactive evaluation in TREC are then discussed. Although some interesting interactive experiments have been performed at TREC, the problems of reconciling the requirements of the laboratory context with the concerns of interactive retrieval are still largely unresolved.

    Notes: The Water Framework Directive was implemented in December 2000 and has to be transposed into Scottish legislation by 2003. The Directive provides a major opportunity to enhance the water environment of Scotland and to improve its management through the implementation of river-basin management planning with stakeholder and public engagement and the delivery of a programme of environmental-improvement measures to deliver good ecological status. Whilst the Directive provides unparalleled opportunities, there are significant challenges to its successful implementation in Scotland, where a step-change in managing the water environment will be required. This paper explores some of the challenges which include technical aspects, scale and timing issues, and capacity limitations.

Hailey Hailey disease – a significant hazard to consider when patch testing (2004)

    Notes: A 42 year old man was referred to the Contact Dermatitis Investigation Unit because of a 12 month history of an inflammatory eruption affecting his neck. He was patch tested to our standard series, textile series and his own medicaments. At the 48 hour reading the patches were removed and there were large areas of unusual painful superficial erosions where the patch test tape – Scanporä had been in contact with the skin. This appearance was difficult to explain. He denied interfering with the patches. The erosions were dressed with non-adherent dressings held in place by a different tape- Microporeä. At the 96 hour reading these dressings were removed and in the areas under the Microporeä tape there were further erosions around the edge and also some thin walled vesicles and small bullae. There were ++ allergic reactions to sodium metabisulphite, disperse blue 35, 124 and 106 all of which were felt to be relevant as he had used Trimovateâ cream and regularly wore dark nylon clothing when refereeing lacrosse matches. A biopsy was taken for histology and immunofluorescence from an area of blistering. This demonstrated suprabasal acantholysis with vesicle formation. There were occasional corps ronds. The immunofluorescence was negative. Anti-epidermal (skin) antibodieswere also negative. This striking and alarming clinical presentation after removal of patch tests had been encountered once previously in our Contact Dermatitis Investigation Unit. The possibility that the disorder was artefactual either due to an irritant introduced under the tapes or perhaps due to a contaminant within the tape adhesive was considered but no conclusion was reached In this case the clue to diagnosis became apparent at the second patch test reading when small vesicles and bullae could be seen at the edge of where the Microporeâ had been. A diagnosis of Hailey Hailey disease (HHD) trigered by the trauma caused by the adhesive tapes and their removal was made. The relatively persistent nature of his presenting problem makes Grover’s disease less likely. The affected sites healed after two weeks. Allergic contact dermatitis particularly to medicaments is reported to be a frequent complication and routine patch testing has been advocated1. There has been a previous report of blistering at the site of patch testing however he was an individual already known to have HHD2. Despite having patch tested around 30 000 people in the last 22 years this complication had not been encountered until this year. We present this case whose clinical features are distinctive as we feel similar instances may have been or will be seen by colleagues. Diagnosis may be difficult if there are no blisters in association with the painful erosions.
