    Description / Table of Contents: Summary 1. The proteinase-inhibitor of beef-liver was isolated out of extracts over deproteinising by perchloric acid, cationexchange-chromatography, and the trypsin-inhibitor-complex (0,277 μg protein/ImU). 2. The identity of the functional groups of both proteinase-inhibitors was demonstrated by the following results: there are no differences in the proportion of the inhibition of trypsin, and kallikrein; both inhibitors do not inhibit kallikrein of dogs, and rats; pancreas trypsin cannot displace the kallikrein from the liver-inhibitor-compound in one hour; the same is true for the complexes with Trasylol. 3. In the blood-coagulation-test with normal blood, hemophilia A- and hemophilia B-blood Trasylol, and liver-inhibitor retard the coagulation to the same degree. 4. The inhibitors inhibit the coagulation of hemophilia B-blood much more than that of hemophilia A-blood. 5. The rough extracts of bovine liver retard the coagulation of blood more than Trasylol does, they also have a small inhibiting affect against pepsin. 6. In pig-liver extracts won in the same manner an inhibiting effect against the mentioned proteinases, and blood coagulation was not to be demonstrated.
    Notes: Zusammenfassung 1. Der Proteinasen-Inhibitor aus Rinderleber wurde über Enteiweißung mit Perchlorsäure, Kationenaustausch-Chromatographie und den Trypsin-Inhibitor-Komplex rein dargestellt (0,277 μg Protein/ImE) und mit Trasylol verglichen. 2. Die Identität der funktionellen Gruppen beider Inhibitoren wurde an Hand folgender Ergebnisse erwiesen: Im Hemmungsverhältnis für Trypsin und Kallikrein bestehen keine Unterschiede; beide Inhibitoren hemmen Pankreas-Kallikrein von Hund und Ratte nicht; Trypsin kann Kallikrein innerhalb 1 Std. aus der Bindung mit Leber-Inhibitor nicht verdrängen; Kallikrein kann Trypsin aus der Bindung mit Leber-Inhibitor nicht verdrängen; das entsprechende gilt für die Trasylol-Komplexe. 3. Im Nativ-Venenblut-Gerinnungs-Test mit Normalblut, Hämophilie-A-Blut und Hämophilie-B-Blut verzögern Trasylol und gereinigter Leber-Inhibitor die Blutgerinnung gleich stark. 4. Die Inhibitoren verzögern die Gerinnung von Hämophilie-B-Blut erheblich stärker als die von Hämophilie-A-Blut. 5. Ungereinigte Rinderleberextrakte verzögern die Blutgerinnungszeit stärker als Trasylol; sie haben auch eine geringe Hemmwirkung für Pepsin. 6. In gleichartig gewonnenen Schweineleberextrakten war eine Hemmwirkung gegen die untersuchten Proteinasen und die Blutgerinnung nicht nachweibar.

    Description / Table of Contents: Summary For the estimation of the secretin half life time an indirect method was chosen, as the concentration of secretin in the blood cannot directly be determined till now. The tests were performed on dogs. The “indirect half life time of secretin” hereby corresponds — by definition — with the period, in which the activity of secretin is half reduced. This activity was determined by pancreatic secretion — as parameter — transformed by the dose response curve. The decrease of secretion — following an exponential function — was transformed by the dose response curve into a dose time curve which allows to estimate the indirect half life time. The indirect half life time of secretin is not constant, but changes continuously because of the double logarithmic decrease in activity. With a medium dosage (1 unit/kg) a half life time of approx. 2.9 min was attained. When doubling the dose a half life time of about 2.2 min results as against 4.4 min when using one half of the dosage (1/2 unit/kg). The average half life time — the mean value of these periods — is about 3.2 min.
    Notes: Zusammenfassung Zur Bestimmung der Halbwertszeit des Secretins wurde eine indirekte Methode gewählt, da die Secretinkonzentration im Blut zur Zeit noch nicht direkt bestimmt werden kann. Die Versuche wurden an Hunden vorgenommen. Die „indirekte Secretinhalbwertszeit“ entspricht dabei per definitionem der Zeitspanne, in der die Aktivität des Secretins, gemessen am Parameter der Pankreassekretion und transformiert über die Dosiswirkungskurve, um die Hälfte abnimmt. Der einer exponentiellen Funktion folgende Sekretionsabfall wurde über die Dosiswirkungskurve in eine Dosiszeitkurve übertragen, aus der die indirekte Halbwertszeit des Secretins abgelesen werden kann. Die indirekte Secretinhalbwertszeit ist dabei nicht konstant, sondern ändert sich wegen des doppelt logarithmischen Aktivitätsabfalls stetig. Bei mittlerer Dosierung (1 E/kg) wurde eine Halbwertszeit von ungefähr 2,9 min gefunden. Bei Verdoppelung der Dosis resultiert aus der Aktivitätsverlaufskurve eine Halbwertszeit von rund 2,2 min, während sie bei halber Dosierung (1/2 E/kg) bei ungefähr 4,4 min liegt. Die mittlere Halbwertszeit, ein Durchschnittswert aus diesen Zeitspannen, liegt bei 3,2 min.

Depressor response, stimulation of gastric secretion and increase of histamine concentration in the blood strongly support the view that Epontol® (Propanidid, dissolved in cremophor-EL) releases histamine. 2. Cremophor-EL, the dissolver of propanidid, could be excluded as histamine releasing substance in man. 3. Atropine nor inhibits the release of histamine nor the stimulation of gastric secretion. Histamine stimulates the gastric secretion directly and does not need acetylcholine as transmitter. 4. Before Epontol® injection not only atropine, but also an antihistaminie drug should be used for premedication. The rapid injection of Epontol® should be avoided.
    Notes: Zusammenfassung 1. Anhand von Blutdruckabfall, Magensaftstimulierung und Zunahme des Bluthistamingehalts wird bewiesen, daß Epontol® beim Menschen Histamin freisetzt. 2. Cremophor-EL, der Lösungsvermittler des Propanidids, konnte als Ursache der Histaminfreisetzung ausgeschlossen werden. 3. Atropin verhindert weder die Histaminfreisetzung, noch die Wirkung auf die Magensaftsekretion. Histamin stimuliert die Magensaftsekretion direkt und ohne Vermittlung durch Acetylcholin. 4. Bei Epontol®-Narkosen wird neben Atropin und bei entsprechender Disposition die Verabreichung eines Antihistaminicums empfohlen. Schußinjektion ist zu vermeiden.

    Description / Table of Contents: Zusammenfassung Menschliches Nasensekret enthält unter physiologischen Bedingungen einen spezifischen, relativ niedermolekularen Proteaseninhibitor mit Hemmwirkung gegen Trypsin, Chymotrypsin und Pronase. Die inhibitorische Kapazität des Nasensekrets, untersucht an 20 gesunden Patienten und bezogen auf sezerniertes Protein, entspricht nahezu der des Blutplasmas. An 66 Patienten wurden die Veränderungen des Inhibitorspiegels bei verschiedenen Rhinopathien untersucht, Erniedrigung des Inhibitorspiegels auf 20% war bei allen frischen Nasen- und Nasennebenhöhleninfekten zu beobachten. Um 55% erniedrigt ist der Inhibitorgehalt des Sekrets bei chronischen Nasennebenhöhlenerkrankungen, wobei sich besonders niedrige Werte bei den Fällen finden, die erst in späteren Stadien der Infektion untersucht wurden. Die Höhe des Inhibitorspiegels im Nasensekret ist offensichtlich vom Funktionszustand der Nasenschleimhaut abhängig.
    Notes: Summary Under physiological conditions human nasal mucus contains a specific low molecular protease inhibitor which inhibits trypsin, chymotrypsin and pronase. The inhibitory capacity of nasal mucus was examined in 20 healthy subjects and correlated to secreted protein. This was found to be comparable with the blood serum. The variations in inhibitor levels were examined for the different forms of rhinopathy (66 subjects). A decrease to 20 p. c. of the normal inhibitor activity was found in acute nose-sinus-infections, an average decrease to 55 p. c. in chronic infections, whereas the activity was particulary low in those cases examined in late phase of infection. Thus the inhibitor level seems to depend on the state of the nasal mucus membrane.

    Description / Table of Contents: Summary Histalog in a dose of 2 mg/kg which is used for maximal stimulation of the human gastric acid secretion, showed not only directly histamine-like actions, but also an enhancement of the action of kinins and seotonin in different species. This seems to be the reason why antihistaminic drugs without antikinin and antiserotonin action do not prevent the side effects of histalog during the test. 13-fold higher histamine levels in human gastric juice than in plasma support the hypothesis, that histalog stimulates gastric secretion by a release of histamine.
    Notes: Zusammenfassung In der beim maximalen Histalogtest üblichen Dosis von 2 mg/kg, zeigt Histalog bei verschiedenen Tierspezies neben dirketen histaminartigen Wirkungen auch noch eine Steigerung der Serotonin- und Kininwirkung. Wahrscheinlich sind deshalb Antihistaminica ohne Antikinin- und Antiserotoninwirkung gegenüber Nebenerscheinungen des maximalen Histalogtestes wirkungslos. 13fach höhere Histaminspiegel im Magensaft als im Blutplasma des Menschen sprechen für eine Histaminliberierung durch Histalog.
