    Notes: Summary The origin of sympathetic and sensory innervation of the knee joint was investigated by the intra-axonal transport method. After injection of horseradish peroxidase (HRP) or horseradish peroxidase conjugated to wheat germ agglutinin (WGA-HRP) into the knee joint unilaterally in adult rats, labelled neurons were found ipsilaterally both in sympathetic and dorsal root ganglia at the lower thoracic and lumbosacral levels. A maximum of 1500 and 1000 peroxidase-positive cells were observed in the sympathetic and dorsal root ganglia, respectively; in the former they were mainly located at the levels of L2–L4, and in the latter mainly at L3–L5. The functional implications of the above observations are discussed in relation to pain mechanisms in joints and to the earlier hypothesis that a nervous component might be involved in the pathogenesis of chronic joint inflammation.

    Notes: Summary Lectin-conjugated horseradish peroxidase was either injected or implanted in crystalline form in various parts of the periaqueductal gray substance in the cat. After survival times ranging between 24 and 48 h the animals were fixed, and the mesencephalon and thalamus were sectioned and processed for peroxidase histochemistry, using tetramethylbenzidine as the chromogen. Light microscopic examination of the sections revealed that there exists a prominent reciprocal connection between the ventral and lateral parts of the periaqueductal gray matter on one hand, and the reticular nucleus of the thalamus on the other. The connections are mainly ipsilateral, and involve the entire rostrocaudal extent of the thalamic reticular nucleus, but mainly the ventrolateral sector of its caudal two thirds. There is a differential labelling within the thalamic reticular nucleus.

    Notes: Abstract  In order to investigate the changes in the somatosensory organization that occur after a peripheral nerve injury, a purely sensory nerve (radial nerve – superficial branch) was divided in adult monkeys (Macaca fascicularis). The nerve ends were immediately rejoined by an epineural suturing technique. After 6–21 months the nerve investigated was exposed to an intra-axonal nerve tracer (horseradish peroxidase conjugate) in order to label the primary afferent terminals within the cuneate nucleus of the brainstem. The non-transected nerve on the contralateral side was similarly exposed and served as a control. Terminal labelling was seen throughout the cuneate nucleus, mainly in the middle of its rostro-caudal extension, and in this part it showed a patchy appearance superimposed on cell clusters within the pars rotunda. This pattern of distribution was seen both on the experimental and control sides. On the experimental side there was an obvious loss of terminal labelling within the terminal field as estimated using an image-analysing system: Compared with the contralateral side the median loss (peroxidase activity) was 83% and between 6 and 21 months only minor restoration of the terminal intensity was observed. These results in the primate confirm earlier results in the cat that transection and microsurgical repair of a sensory nerve causes a considerable loss of neurons capable of intraaxonal transport.

    Notes: In patients, the long-term outcome of injuries to sensory nerves is poor. This is only partly due to mismatching of regenerating axons at the transection site. We found in the macaque monkey that 70% of the transganglionic labelling in the spinal dorsal horn was still significantly reduced 21 months after transection and suturing of the sensory radial nerve. The reduction was evenly distributed throughout the terminal field of nerve endings, which were labelled with a mixture of the intra-axonal nerve tracer wheat germ agglutinin-horseradish peroxidase conjugate and pure horseradish peroxidase.

    Notes: Summary The afferent fibres to the ventroposterolateral nucleus (VPL) of the contralateral thalamus from neurons in the dorsal column nuclei (DCN) and the lateral cervical nucleus (LCN) were labelled by anterograde transport of wheat germ agglutinin-horseradish peroxidase conjugate and subsequent histochemical processing with tetramethyl benzidine. In accordance with the results of previous light microscopical studies using the degeneration method or autoradiographic tracing technique, the distribution of the afferents from the DCN and LCN in the VPL differed considerably. Thus the DCN terminals, which were calculated to constitute about 7–8% of the total number of boutons in the VPL, were found throughout the entire VPL, whereas the LCN terminals were mainly located in its dorsal and dorsolateral parts, where they made up about 1% of the total number of boutons. However, the morphology and synaptic organization of the terminals from the DCN and LCN were virtually identical. Thus the synaptic terminals of the two afferent pathways seemed to be represented by large boutons of a similar type, which had large, slightly oval and loosely packed synaptic vesicles and contained numerous mitochondria. Both DCN and LCN terminals synapsed preferentially on medium-sized to large dendrites, but were also presynaptic to other vesicle-containing profiles, probably of internuncial origin, which in turn were in synaptic contact with the same dendrites as the labelled ones. It is suggested that the differences in physiological properties between the somatosensory information that is transmitted to the somatosensory cortex via the dorsal column-medial lemniscus pathway and the spino-cervico-thalamic tract do not seem to have a counterpart in differences in the synaptic organization of their relay in the VPL.
