    Notes: Abstract. We developed a new two-chamber system for the coculture of hepatocytes and fecal microflora under aerobic and anaerobic conditions, respectively, to investigate the sequential metabolism of chemicals by the liver and microflora in vitro. The culture device consisted of two chambers separated by a permeable polycarbonate membrane. In the aerobic compartment, hepatocytes were cultivated as a monolayer on the membrane and in the anaerobic compartment fecal microflora as a suspension. To characterize the metabolic capacity of the microflora and hepatocytes, various marker enzymes were studied. Azoreductase, nitroductase, β-glucuronidase, β-glucosidase and sulphatase were tested in the microflora of the feces from three volunteers who had had significantly different eating habits for years (daily meat, mixed diet, vegetarian). The microflora exhibited significant activities and the various enzymes differed only moderately in the samples from the three volunteers. For rat hepatocytes the activities of various cytochrome P450 forms and conjugating enzymes served as markers. The enzyme activities were tested in the coculture system during a 4-h culture period intended for the test protocol. Deethylation of ethoxycoumarin and 2α-, 6β- and 16α-hydroxylation of testosterone decreased by about 30%, 25%, 40% and 20%, respectively, while there was no loss of glucuronidation and sulphonation of 3-OH-benzo(a)pyrene nor of glutathione conjugation of 1-chloro-2,4-dinitrobenzene during the 4-h culture period. The activities of the tested hepatic phase I and II enzymes were not changed after coculture of the hepatocytes with the microflora for 4 h. The applicability of the in vitro system for studying the metabolic interaction of liver and microflora was demonstrated using 7-ethoxycoumarin and the developmental drug EMD 57033, a thiadiazinon derivative from Merck KGaA, as model compounds. Both compounds were oxidized and conjugated by liver cells. In the coculture of hepatocytes and fecal microflora the resulting glucuronides and sulphoconjugates were split by hydrolytic enzymes of the intestinal microflora.

    Notes: This paper presents results from quasi-static and high rate tensile testing of threealuminum sheet alloys, AA5754, AA5182 and AA6111, all of which are candidates for replacingmild steel in automotive bodies. Tests were performed at quasi-static rates using an Instronapparatus and at strain rates of 600 to 1500 s-1 using a tensile split Hopkinson bar. Additionally, anin-depth investigation was performed to determine the levels of damage within the materials and itssensitivity to strain rate.The constitutive response of all of the aluminum alloys tested showed only mild strain ratesensitivity. Dramatic increases in the elongation to failure were observed with increases in strainrate as well as greater reduction in area. Additionally, the level of damage was seen to increase withstrain rate

    Notes: The cytokine profile of CD4+, CD8+ T cells, γδ+ T cells and natural killer (NK) cells (CD94+CD3–) was studied in a patient with visceral leishmaniasis (VL). The otherwise healthy, human immunodeficiency virus-negative patient acquired the disease in Tuscany, Italy. Diagnosis was made by demonstration of high concentrations of antibodies against Leishmania antigens in serum. Flow cytometry for the detection of intracellular interferon-γ (IFN-γ), interleukin (IL)-2, IL-4, IL-6, IL-10, IL-13 and tumour necrosis factor (TNF)-α expression in peripheral blood mononuclear cells stimulated with phorbol 12-myristate 13-acetate and ionomycin was performed, followed by treatment with liposomal amphotericin B. CD4+ cells were identified as major cytokine-expressing cells, capable of producing both type 1 and type 2 cytokines. A high frequency of IL-4- and IL-13-expressing CD8+ cells was noted. NK cells and γδ+ T cells, thought to be involved in innate host defences against Leishmania, expressed IFN-γ and TNF-α. Ten per cent of γδ+ T cells expressed IL-10, predominantly together with IFN-γ, suggesting additional immune-regulatory roles for this T-cell subset in VL.

    Notes: Abstract— The magnetic emission (ME) technique is used to determine the onset of crack propagation in brittle and ductile steels under dynamic loading. Brittle fracture is directly indicated by the ME signal; however the onset of stable crack propagation can usually not be seen in this signal. A new result is that the integrated ME signal, which is proportional to the magnetic field at the site where the probe is located, is capable of detecting this event. Critical values of the J-integral derived with this method, in different test series, are compared with conventional R-curve and stretch zone methods. The ME-derived critical dynamic J-integral data fit very well to the stretch zone results which describe physical crack initiation. It is therefore concluded that the easy-to-apply and low-cost ME technique is capable of indicating material failure even with stable crack propagation and can therefore provide critical material parameters.
