    Notes: Abstract The decomposition of three different 14C-labeled cellulose substrates (plant holocellulose, plant cellulose prepared from 14C-labeled beech wood (Fagus sylvatica) and bacterial cellulose produced by Acetobacter xylinum) in samples from the litter and mineral soil layer of a beechwood on limestone was studied. In a long-term (154 day) experiment, mineralization of cellulose materials, production of 14C-labeled water-soluble compounds, and incorporation of 14C in microbial biomass was in the order Acetobacter cellulose 〉 holocellulose 〉 plant cellulose in both litter and soil. In general, mineralization of cellulose, production of 14C-labeled water-soluble compounds, and incorporation of 14C in microbial biomass were more pronounced, but microbial biomass 14C declined more rapidly in litter than in soil. In short-term (14 day) incubations, mineralization of cellulose substrates generally corresponded with cellulase and xylanase activities in litter and soil. Pre-incubation with trace amounts of unlabeled holocellulose significantly increased the decomposition of 14C-labeled cellulose substrates and increased cellulase activity later in the experiment but did not affect xylanase activity. The sum of 14CO2 production, 14C in microbial biomass, and 14C in water-soluble compounds is considered to be a sensitive parameter by which to measure cellulolytic activity in soil and litter samples in short-term incubations. Shorter periods than 14 days are preferable in assays using Acetobacter cellulose, because the decomposition of this substrate is more variable than that of holocellulose and plant cellulose.

    Notes: Abstract A typical, small encatchment (catena Bölkendorf) in the moraine, northeast German agricultural landscape Schorfheide-Chorin was studied with respect to summit, midslope, and foot-slope positions at northern and southern slope exposure, respectively, including a central noncultivated kettle hole position (pot hole). Across the sequence of seven distinct sampling positions, soil organic carbon and total nitrogen contents, soil gravimetric water content, and soil microbial biomass displayed maxima at the kettle hole position. Soil pH revealed a decreasing trend at the northern exposed slope and a minimum at the kettle hole position. Against this background, the population density of total culturable bacteria clearly displayed a minimum at the kettle hole position, whereas the population density of carboxymethylcellulose decomposing bacteria was not clearly differentiated in relation to sampling positions. To study the phylogenetic diversity of culturable cellulolytic bacteria, 311 isolates were obtained from the sampling positions across the entire encatchment and examined by restriction analysis of PCR-amplified 16S rDNA. Using the restriction enzyme ScrFI, isolates were classified into 31 pattern groups. Although the ratio of actinomycetes within total isolates ranged from 0.73 to 0.94, only 16 pattern groups originated from actinomycetes, but 15 from other bacteria. At all sampling positions, a dominant pattern group was identified, containing 38 to 65% of total isolates. Two site-specific pattern groups could be identified, representing significant parts of the total population, which were highly specific for the kettle hole (19% of total isolates) and for foot- and midslope positions (15–18% of total isolates), respectively. In general, the composition of cellulolytic isolates across the encatchment displayed differences with respect to slope positions, but was not significantly affected by soil properties. Based on 16S rDNA sequence analysis, isolates of the dominant as well as the specific pattern groups could be assigned to the genus Streptomyces. Furthermore, sequencing of 16S rDNA of isolates of another three pattern groups revealed a high phylogenetic diversity among these isolates, including cellulomonads and bacilli.

    Notes: Abstract Perinatal transmission of hepatitis B virus (HBV) occurs in about 10%–20% of anti-HBe seropositive mothers. The babies are at risk of developing fulminant hepatitis. In most cases no viral DNA has been detected in the sera of mothers and children by conventional hybridisation techniques. Thus, the aim of our investigation was to demonstrate HBV DNA in three children with liver failure and their anti-HBe positive mothers by more sensitive molecular hybridisation techniques. The babies were healthy at birth and did not receive vaccination. At 3 months of age they developed acute liver failure and died from liver insufficiency. Only in one child serum HBV DNA was detected by dot blot hybridisation, but polymerase chain reaction (PCR)-detectable HBV DNA was present in all sera. The liver specimen was negative for HBV DNA by Southern blot hybridisation, but showed a focal distribution of viral sequences as determined by in situ hybridisation. This finding was confirmed by PCR. Our results prove that chronic anti-HBe positive HBsAg carrier mothers and their babies show a low level virus replication. Fulminant hepatitis is due to vertical transmission of very small amounts of viral DNA, only detectable by most sensitive techniques like PCR and in situ hybridisation. Our findings underline the necessity to vaccinate all babies of HBsAg positive mothers regardless of HBeAg/anti-HBe status.

    Notes: Abstract 3-Hydroxyacyl-CoA dehydrogenase deficiency is a newly recognised fatty acid oxidation disorder with a usually fatal outcome. We present a further patient who presented with hypoketotic hypoglycaemia, hepatopathy, secondary carnitine deficiency and increased plasma long-chain acylcarnitines. 3-Hydroxydicarboxylic aciduria was present and the diagnosis confirmed in cultured skin fibroblasts. Our patient is compared with those reported in the literature with respect to clinical symptoms, differential diagnosis and possible therapeutic regimens.

    Notes: Abstract Following the cloning and sequencing of the hepatitis B virus genome, molecular hybridization techniques have been established to detect hepatitis B virus (HBV) DNA in serum and liver tissue. Analyses can be performed by dot blot, Southern blot and in situ hybridization. HBV DNA is regarded to be the most sensitive marker of viral replication and infectivity which was previously related to the presence of hepatitis B e antigen in serum and hepatitis B core antigen in liver cells. In liver tissue different molecular patterns can be recognized as free viral DNA and integrated sequences. Furthermore, introduction of the polymerase chain reaction allows the detection of very small amounts of viral DNA and has markedly improved diagnostic sensitivity. Thus the study of HBV DNA has become a valuable part of the routine diagnosis in chronic hepatitis B, providing a more reliable evaluation of virus replication and infectivity, and facilitating more precise statements about course and prognosis of the disease.

    Notes: Abstract More than 50% of children with chronic hepatitis B do not respond to treatment with alpha-interferon. Since these patients continue to display high viral replication and progressive liver disease, retreatment should be considered. To date it has not been well evaluated whether a second course of treatment could increase the response rate. In two alpha-interferon retreatment trials in adult patients the response rate, defined by seroconversion from HBeAg to anti-HBe, ranged between 11% and 44%. One beta-interferon retreatment study in children reported a seroconversion rate of 32%. Regrettably, none of the studies included a control group observing the `spontaneous' seroconversion rate after a first interferon cycle. Thus, a nonrandomized alpha-interferon retreatment study in children including control patients was performed. Alpha-interferon for retreatment was administered 3 times a week for 16–24 weeks in 15 children (5–16 years) at least 6 months after ceasing the first cycle. Four children received 5 MU/m2 of a natural alpha-interferon and 11 children 9 MU/m2 recombinant alpha-interferon 2b. Follow up was 18–47 months after initial treatment. In parallel, a control group of 19 un-retreated children with comparable clinical and demographic data was followed for 12–39 months. HBeAg seroconversion was observed in 5 (33%) of the retreated children and in 5 (26%) of the control patients during follow up. The difference is not significant. In the initially nonresponding children, those with high ALT levels before the first treatment showed late HBeAg seroconversion more frequently than those with low ALT levels (P = 0.017) irrespective of retreatment. The ALT level before retreatment was not a predictor for response. Conclusions A second cycle of alpha-interferon during the 3 years following the first treatment in nonresponding children with chronic hepatitis B can be safely performed but did not increase HBeAg/anti-HBe seroconversion compared with the spontaneous seroconversion rate of patients without retreatment.

    Notes: Abstract We report on a 9-year-old boy with a congenital renal arteriovenous fistula of the aneurysmal type, a form previously not observed in childhood. The clinical picture was unusual with severe arterial hypertension, excessive polyuria and decreased levels of serum sodium and chloride as main signs. Clinical and biochemical findings normalised after nephrectomy of the kidney involved.

    Notes: Abstract Cell-mediated immune reactions play the most important role in the pathogenesis of chronic viral and auto-immune hepatitis. Cellular cytotoxicity (CC) of peripheral blood lymphocytes against autologous hepatocytes isolated from liver biopsies was studied in 29 children with different types of hepatitis B surface antigen (HBsAg)-positive hepatitis. Children with chronic hepatitis B showed higher cytotoxicity than control patients. However, a correlation of cytotoxicity to serum amino-transferases, HBeAg-/Anti-HBe-status, and hepatitis B virus DNA in serum could not be found. Children with a higher percentage of hepatitis B core antigen (HBcAg) expression in their liver tissue presented lower CC values, and vice versa. This supports the hypothesis that virus elimination occurs via T-cell attack against HBcAg expressing hepatocytes. Furthermore, children with a longer duration of chronic hepatitis B had considerably higher cytotoxicity values. Possibly, CC plays a role in perpetuating liver damage after infection with hepatitis B virus.

    Notes: A measuring procedure for the determination of the switching behavior of hard magnetic particles is presented. For this end, the remanence vector is determined for increasing fields. In the first part of the presented analysis, the angular dependence of the switching field is calculated, neglecting the switching field distribution. In the second part, the portion of switching particles depending on the strength of the applied field and its orientation with respect to the particle easy axes is determined. This switching portion includes information on the angular dependence of the switching field and the switching field distribution for each particle orientation. The method is demonstrated on a magnetically diluted barium ferrite sample with noninteracting single-domain particles.
