    Notes: Summary Primary afferent projections from cervical and lumbar levels were studied in the turtle Pseudemys scripta elegans. Injections of radioactive amino acids, wheat germ agglutinin and horseradish peroxidase were made into the dorsal root ganglia or the spinal cord. Previous reports on the terminal distribution of primary afferents within the ipsilateral segment of entry were confirmed (Kusuma and ten Donkelaar 1979, 1980) and additional dorsal root projections were demonstrated to the contralateral side and to several neighboring spinal segments. The primary afferent projections to the brainstem were essentially restricted to a dorsolateral area that appears to be homologous to the main dorsal column nuclei (n. gracilis and n. cuneatus medialis) in mammals. While exhibiting a similarly extensive rostro-caudal span, the projections originating from lumbar injections terminated more medially, those from cervical injections more laterally. The labeling pattern suggested that terminations are mainly on dorsally extending dendrites.

    Notes: Summary The distribution, organization and origin of the ipsilateral descending limb of the Brachium Conjunctivum (B.C.), have been studied in the rat by using anterograde and retrograde tracing techniques. After injections of tritiated leucine/proline into the lateral cerebellar nucleus, covering both its medial part, corresponding to the dorsolateral hump (DLH) of Goodman et al. (1963) and its lateral part, (designated here as the lateral dentate, LD), and the neighboring interposed nucleus (NI), emerging fibres are numerous and leave laterally from the B.C. On the contrary, injections restricted to LD reveal very few such fibers. Within the lateral parvocellular reticular formation (LPRF) terminal labelling is heavy, and moderate to sparse within the adjacent trigeminal complex. Rostro-caudally, silver grain accumulation within the LPRF extends from the level of the motor trigeminal nucleus (VM) to the pyramidal decussation, exhibiting a cephalocaudal decrease of grain density. Within the trigeminal complex, labelling occurs in the caudal VM, the dorsal portion of the principal sensory nucleus, and within and around the trigeminal spinalis oralis. In addition, the area surrounding the VM (in part corresponding to the supratrigeminal region of Lorente de Nó 1922, 1933) is moderately labelled. After injections of HRP into various levels of the ipsilateral descending B.C.'s projection field, retrogradely labelled cells are numerous within the DLH. A slightly lesser amount of labelled cells are found in the lateral half of the NI, primarily concerning the nucleus interpositus posterior. Within the LD, only a few labelled cells are observed: these are mainly restricted to the dorsal portion at rostral levels of the nucleus. The results obtained by both the anterograde and retrograde studies suggest an absence of a topographic organization within this descending B.C. component. The possible functional meaning of these results is discussed.

    Notes: Carnation (Dianthus caryophyllus L. cv. White Sim) petals contained two distinct invertases (EC 3.2.1.26) based on chromatographic behavior on DEAE-cellulose. Both are soluble in 20 mM sodium phosphate buffer (pH 6.5) and exhibit acid pH optimum of 5.5. Extraction of a cell wall preparation from petals with 1 M NaCl released little additional activity. Furthermore, only traces of activity remained associated with the NaCl-extracted cell wall preparation. One of the soluble invertases, representing over 75% of the total activity, was partially purified by (NH4)2SO4 fractionation and sequential chromatography over diethylaminoethyl-cellulose, concanavalin-A sepharose and polyacrylamide P-200. The enzyme was purified 38-fold with a recovery of 12%. It had an apparent native molecular weight of 215 kDa. The partially purified invertase is a β-fructofuranosidase (EC 3.2.1.26) based on its specificity for sucrose. The Km for sucrose was 3.3 mM. Accumulation of reducing sugars and increased invertase activity during expansive petal growth indicates that sucrose is the major source of carbon for petal growth.

    Notes: Developmental changes in polypeptide and mRNA popultions in carnation (Dianthus caryophyllus L. cv. White Sim) petals were investigated during the senescence of harvested flowers. Total proteins were extracted from flower petals at various stages of senescence and subjected to separation by sodium dodecyl sulfate polyacrylamide gel electrophoresis. Analysis of the Coomassie blue stained gels revealed polypeptides with apparent molecular weights of 76, 62, 35.5 and 24 kDa which increased, while those with molecular weights of 70.5, 67.5, 46.5 and 31 kDa decreased during petal senescence. Changes in mRNA populations were investigated by translating poly (A)+RNA, isolated from carnation petals, in vitro using the rabbit reticulocyte lysate system. Polypeptides synthesized in vitro were separated by one- and two-dimensional gel electrophoresis and visualized by fluorography. Three classes of mRNA's were associated with the senescence of carnation petals. The majority of the mRNA's were constitutive at all stages of senescence. Another class of mRNA's increased with the climacteric rise in ethylene production, which accompanied the onset of senescence. Their translation products were 81, 58, 42, 38 and 35 kDa. In addition, several mRNA's appeared to decrease in abundance during the course of petal senescence. These results indicate that senescence of carnation flower petals is associated with changes in gene expression.
