Notes: Summary Individual differences in developmental stages of mouse embryos of the two related strains C57 BL/6JHanFfm and C57BL/10JFfm during the period between day VIII and day X p.c. have been examined. The following results were obtained: 1. A remarkable variability in the number of somites could be stated in embryos of different litters but also in embryos of the same litter, especially at stages day IX and day IX +12 h. 2. There was a close relationship between the number of somites and progresses of rotation from the S- to the C-shape. Incidence of pre-rotation stages was higher in embryos of strain C57BL/6 than in embryos of strain C57BL/10. 3. Individual differences were stated in the developmental stages of neural groove and upper limb buds. The degree of differentiation and closure of the neural groove was more progressive in strain C57BL/10. 4. Individual differences were also registered in the appearance and development of the following primordia: eye, ear, heart, thyroid, lung, liver, ureter and the pronephric ducts. 5. The body length of the embryos was closely related to the number of somites.

Notes: Abstract: Objective and Design: Whether Mao-Bushi-Saishin-To (MBST), one of the formulas of classical Chinese medicine, is effective on 48-h passive cutaneous anaphylaxis (PCA) in rats and which substance in the formula is responsible for its inhibitory action were examined.¶Treatment: In the studies on PCA, MBST (hot water extract of the whole herbal formula), extracts of Ephedra herb (Mao), l-ephedrine and other reference drugs were orally administered immediately or at various times before or 5 min after the antigen challenge. In the experiments on anaphylactic histamine release from rat peritoneal mast cells, l-ephedrine and d-pseudoephedrine were added at 10-4-10-7 g/ml at 30, 10, 3 or 0 min before antigen provocation.¶Results: The time course study indicated that MBST produced a prompt and long lasting inhibition of PCA. Among the constituents of Mao, l-ephedrine exerted this prompt inhibitory activity, but d-pseudoephedrine did not. Neither pseudoephedrine nor l-ephedrine prevented the anaphylactic histamine release from isolated peritoneal mast cells.¶Conclusions: It is strongly emphasised that the rapid suppression of PCA by orally administered l-ephedrine must be exerted by a mechanism distinct from that of suppression produced following gastrointestinal absorption of the drug, because the time required for the inhibition was extraordinarily short. However, direct inhibition of anaphylactic histamine release from isolated mast cells was excluded in this inhibition of PCA.¶

Notes: Summary The critical period of Purkinje cell degeneration and cerebellar hypoplasia due to bilirubin was examined in rats with transient hyperbilirubinemia induced by a serial subcutaneous injection of novobiocin from 1 to 3, 5 to 7, 10 to 13, or 16 to 20 days after birth. Animals showing total plasma bilirubin levels of 5 to 7 mg/100 ml 6 h after the final injection were used for this study. In nearly midsagittal sections of the culmen, the percentage of the affected Purkinje cells was 0.9%, 17.1%, 0% and 0% at days 3, 7, 13 and 20, respectively. Thus, the Purkinje cells were most vulnerable to bilirubin between days 5 and 7. Cerebella from the rats which showed transient hyperbilirubinemia at day 3, 7, 13 or 20 were weighed at day 30. The cerebellar weight was significantly low only in rats showing hyperbilirubinemia at day 7. Thus, the critical period of the cerebellar hypoplasia due to bilirubin coincided with the period when the Purkinje cells were most sensitive to bilirubin. These results suggest that the Purkinje cell damage leads to the cerebellar hypoplasia in hyperbilirubinemia.

Notes: Summary Prenatal exposure of pregnant rats to methylazoxymethanol acetate (MAM), an anti-mitotic agent, on day 15 of gestation induces severe microcephaly in the offspring. The present study first investigated a developmental alteration of serotonin (5HT) neurons immunohistochemically in the dorsal and median raphe nuclei in serial sections in both control and microcephalic rats (MAM-rats) at 35 days of age. 5HT-immunoreactive neurons in the MAM-rats were reduced in number and irregularly distributed in the dorsal and median raphe nuclei compared with those in the control. The dendrites of neurons in these nuclei in the MAM-rats were very short and twisted. A follow-up observation on the development of the cerebral cortex at 5, 9 and 28 days of age was performed using Nissl-stained preparations, which revealed a disorganized cell arrangement in the cerebral cortex of the MAM-rats at the very early postnatal period. Furthermore, the distribution of 5HT-immunoreactive fibers into the cerebral cortex was also examined using brains of 28 days of age. In MAM-rats of this age, abnormally tortuous 5HT-immunoreactive fibers were observed in the cerebral cortex. 5HT neurons in the raphe nuclei are known to project their ascending axons widely into the entire cortical area during the 1st postnatal week. Thus, the association of disorganized cortical cell arrangement and the hyperdense and tortuous distribution of 5HT-immunoreactive fibers in the cerebral cortex support the idea of target-dependent secondary degeneration of 5HT neurons in the dorsal and median raphe nuclei of the MAM-rats.

Notes: By combining 1 MeV high-voltage-high-resolution electron microscopy with convergent-beam electron diffraction (CBED), the crystal structure of a perovskite-related compound, Ca4YFe5O13, is determined. The crystal has orthorhombic symmetry with lattice parameters a = 5.46, b = 37.4 and c = 5.54 Å. The space group is determined to be centrosymmetric Pnma by selecting the point group from the CBED patterns. The structure images taken from two principle-axis directions, which are interpreted on the basis of the calculated images, reveal that the structure consists of a succession of the stacking of FeO6 octahedra (O layer) and FeO4 tetrahedra (T layer) and is represented by the stacking sequence of ...OTOOTOTOOTO... along the b axis repeated at intervals of 37.4 Å. It is shown that the Ca4YFe5O13 crystal structure can be described in terms of unit-cell-level twinning of Ca2Fe2O5.
