    Notes: Abstract. Objective: To study the effect of interleukin (IL)-4 and IL-10 on nitric oxide (NO) production by macrophages.¶Materials and Methods: Elicited or resident peritoneal macrophages (PMO) and a macrophage cell line Raw264.7 were primed by IL-4 or IL-10 for 6 hours, and were further incubated in the presence of interferon (IFN)-γ and/or lipopolysaccharide (LPS) for 48 hours. NO2 - accumulation in the supernatant of cultured cells was used as an indicator of NO production and was determined by the standard Griess reaction adapted for microplates. The amount of tumor necrosis factor (TNF)-α in the culture supernatants was determined with a commercially available ELISA kit. The absorbance was measured at 450 nm with a microplate photometer.¶Results: IL-4 inhibited NO production by murine macrophages of different sources and the macrophage cell line Raw264.7. In contrast, different macrophage populations showed differential responses to IL-10. After stimulation with LPS or IFN-γ, IL-10 suppressed NO production by elicited PMO but enhanced NO production by resident PMO or by Raw264.7. Both IL-4 and IL-10 inhibited the production of TNF-α, which has been shown to play a crucial role in NO production. In the presence or the absence of blocking antibody to TNF-α, IL-10 always enhanced NO production by resident PMO. This result suggests that the inhibition of TNF-α production and the enhancement of NO production by resident PMO stimulated with IL-10 are independent, coexisting events.¶Conclusions: Factors other than TNF-α have been suspected to influence NO production by macrophages, and this study indicates that IL-10 may be a candidate cytokine for resident PMO.

    Notes: Abstract —The M 5.1 event (May 23, 1993) which occurred in one of the most active swarm areas of Japan was preceded by foreshock activity. We obtained precise hypocenters of the foreshock-mainshock-aftershock sequences with a temporary seismic network installed just above the source region twenty days before the mainshock. The foreshocks are very unique in their accelerating activity; the acceleration in the number of foreshocks enabled us to estimate the time of the mainshock with time-to-failure analysis proposed by . Although substantial snow remained in the swarm area, we quickly installed the network because the time-to-failure analysis disclosed that the mainshock was impending. The temporary network provided detailed information on both the temporal and spatial distribution of the foreshock-mainshock-aftershock sequences. Foreshocks started fifty days before the mainshock and were distributed linearly at the base of the seismogenic layer with a length of 5 km and horizontal and vertical widths of about 1 km. The temporal change of the number of foreshocks is approximated by a power law, and the time of the mainshock can be estimated by extrapolating plots of the inverse of the daily number of events. An area of seismic quiescence appeared 40 hours before the mainshock and propagated with a rate of 20 m/hour. The mainshock occurred 2 km westward from the primary foreshock area. It was located at the base of the aftershock region. This process can be interpreted as source nucleation; preslip on the fault prior to the mainshock.

    Notes: Summary Ling Zhi-8 (LZ-8), a novel and recently discovered immunomodulatory protein having in vivo immunosuppressive activity, was tested for in vivo effect against Type 1 (insulin-dependent) diabetes mellitus in the non-obese diabetic mouse, the disease having immunologically mediated aetiology in this animal. LZ-8 had mitogenic activity in vitro towards spleen cells of the non-obese diabetic mice as previously shown towards those of DBA/2 mice. Intraperitoneal administration of LZ-8 twice weekly into the mice (10.3–12.6 mg/kg body weight) from 4 weeks of age prevented insulitis and an almost normal number of insulin producing cells were observed. Extreme insulitis and reduction of the number of insulin producing cells were observed in the pancreata of the untreated non-obese diabetic mouse. No cumulative incidence of diabetes mellitus was observed in the LZ-8 treated group, while cumulative incidences of 70% and 60% were observed in an untreated group followed up to 42 weeks of age when the incidence of diabetes was defined as a plasma glucose level of greater than 11 mmol/l and as a urine glucose level of greater than 2 +, respectively. T cell subset population analysis was performed to further investigate the action of LZ-8 on the non-obese diabetic mouse which revealed that LZ-8 treatment increased in L3T4+/Lyt-2+ ratio.

    Notes: Abstract. ATP-sensitive potassium channels were found in frog ventricular myocytes using the inside-out patch-clamp technique. The channel was selectively permeable to K+. Single-channel conductance was 32.6 pS at 3.0 mm of [K+] o and 132 mm [K+] i and 77.3 pS at 114 mm [K+] o and 132 mm [K+] i . ATP did not affect single-channel conductance. The open probability of the channel was decreased by intracellular application of ATP in both the presence and absence of 2 mm MgCl2. The coexistence of Mg2+ with ATP shifts the dose-response curve for the open probability of ATP-sensitive K+ channel against ATP rightward. The shift of the curve indicates that Mg-ATP is less effective than free ATP in inhibiting the channel. An open-time histogram was fitted by a single exponential function with a time constant of 1.63 ± 0.17 msec (n= 5) in an ATP-free medium. Mean open time (1.57 ± 0.10 msec; n= 5) was not altered but the inter-burst time (closed time between bursts) lengthened in 10 μm ATP.

    Notes: Summary This study presents measurements of serum vitamin D metabolites, calcium and phosphorus as well as measurements of the equilibrium dissociation constant for duodenal 1,25(OH)2D3 receptor in 15-, 18-, 19-, and 20-day chick embryos in comparison to that in 1- and 118-day-old chicks and to vitamin D-deficient chicks. The present results showed that: (a) serum 1,25(OH)2D and 24,25(OH)2D levels rise from 15 and 18 to days 19 and 20 of embryonic development while serum phosphate levels are stable; (b) serum calcium levels rise at hatching to adult levels; (c) the duodenal 1,25(OH)2D3 receptor is detectable in 15-day-old embryo and has a Kd similar to that of 118-day-old vitamin D-replete chicks; and (d) the activity of 1,25(OH)2D3 receptor in chick duodenal cytosol is maximal at hatching.

    Notes: Abstract A 16-year-old girl with disseminated molluscum contagiosum (MC) was found to have a very low level of serum IgM, elevated levels of IgG and IgA, and a high level of IgE. She had normal numbers of peripheral blood IgM+, IgG+ and IgA+ B-lymphocytes but their terminal differentiation into plasma cells could not be induced by pokeweed mitogen (PWM) in vitro. On the other hand, the patient's T-cells showed normal helper functions in the PWM system and normal interferon (IFN) production in vitro. However, the IgM+ B-cells can be induced to differentiate into IgM secreting cells by Epstein-Barr virus (EBV), suggesting that the genetic mechanism for synthesis of the component immunoglobulin proteins is present. T-cell functions were impaired, as shown by delayed type cutancous hypersensitivity (DTH) and mitogen response. The data suggest that the selective IgM deficiency of the patient is due mainly to defects in B-cells at the terminal differentiation stage, but immunological abnormalities are present in both B and T-cell systems. Neutrophil functions examined were normal. MC was treated by intravenous injection of IFN without any side effects; however, no clinical improvement was achieved.

    Notes: Abstract In order to clarify the complex interrelationship between serum calcium, 1,25-dihydroxyvitamin D (1,25(OH)2D), and parathyroid hormone (PTH), and the urinary excretion of cyclic AMP (cAMP) in response to exogenous PTH in pseudohypoparathyroidism (PHP) and related diseases, we investigated 3 patients with parathyroid disorders before and after treatment with 1α-hydroxyvitamin D3 (1α-OH-D3). Low plasma 1,25(OH)2D before treatment increased after giving 1α-OH-D3 (0.1 μg/kg/day), where-as high plasma PTH measured by the C-terminal assay (C-PTH) decreased in all 3. No response in urinary cAMP was found before or after treatment in 2 patients with PHP type I, despite the fall of plasma C-PTH. However, in one patient with extremely high plasma C-PTH but normal N-PTH (measured by a homologous radioimmunoassay using 1–34 human PTH), urinary cAMP response to exogenous PTH was increased after treatment with 1α-OH-D3. We suggest that he had pseudopseudohypoparathyroidism (PPHP) with Albright's hereditary osteodystrophy and a partial deficiency of renal 1α-hydroxylase. In this patient secondary hyperparathyroidism is thought to be due to 1,25(OH)2D deficiency, and the decreased responsiveness to exogenous PTH before treatment due to excess PTH occupying renal receptors.

    Notes: Abstract In 1974, a 2-year-old boy was diagnosed as having X-linked hypophosphataemic rickets (XLH) because of severe rickets and hypophosphataemia. The vitamin D metabolite concentrations, blood and urine chemistry and renal 25-hydroxyvitamin D3 (25 OHD3)-1α-hydroxlase were measured in 1982 (about 2 weeks after withdrawal of medication). 1α-hydroxylase was 392 pg/mg tissue/20 min in the patient, which was high compared with aged-matched controls (69.7±28.5 pg/mg tissue/20 min, mean ±SD, n=7). Our present studies showed that the 1α-hydroxylase activity in the patient with XLH was elevated. Therefore, the normal or low 1,25-dihydroxyvitamin D3 (1,25-(OH)2D3) concentrations in XLH patients could be due to accelerated catabolism of 1,25-(OH)2D3 or abnormally regulated 25OHD3-1α-hydroxylase in response to hypophosphataemia, although significantly elevated above that in normal controls.
