    Notes: Laser spot scanning studies of single-crystalline p-InSe in contact with a neutral aqueous solution reveal a dramatic difference in lateral resolution between the material "as cleaved'' and after platinization by brief dipping in a dilute H2PtCl6 solution. A model is developed to explain these observations, and the resolution is calculated as a function of the diffusion coefficient and the life time of minority carriers, and of the charge transfer rate. The improvement of the resolution is found to be due to the increase of the rate of hydrogen evolution at the illuminated semiconductor by Pt catalyst. The model also provides numerical values for the charge transfer rate in the noncatalyzed and the catalyzed cases.

    Notes: We have investigated a Josephson effect heterodyne mixer for the millimeter wave band employing high-Tc GdBaCuO point contacts. Mixer performance was in qualitative agreement with theory. A mixing response was observed up to 55 K, the highest operating temperature achieved for such a device to date. The voltage separation of rf-induced steps gave a value of h/2e=2.08×10−15 V s which is in excellent agreement with the value expected for Cooper pairs. In addition, the temperature dependence of the I0R product was found to agree with Bardeen–Cooper–Schrieffer theory in the weak coupling limit.

    Notes: Cerebrospinal fluid (CSF) lymphocytes from two patients with tuberculous meningitis proliferated stronger than the corresponding peripheral blood lymphocytes (PBL) when stimulated with tuberculin purified protein derivative (PPD) in the lymphocyte transformation test after 3 days of culture. This might indicate an accumulation of specifically primed lymphocytes within the central nervous system. CSF lymphocytes and PBL from nine of ten patients with acute aseptic meningitis investigated as controls showed no or low responses when stimulated with. PPD, whereas the remaining patient displayed a significant proliferation of CSF lymphocytes, which was more pronounced than that of PBL. Stimulation with the mitogens phytohaemagglutinin, concanavalin A, and pokeweed mitogen gave lower proliferation of CSF lymphocytes compared with PBL in tuberculous and aseptic meningitis. Evaluation of the proliferative response of CSF lymphocytes compared with PBL on stimulation with PPD might be a useful complement in the diagnosis of tuberculous meningitis.

    Notes: We present a case of chronic liver disease with selective and exclusive hepatocyte endoplasmic reticulum storage of δ1-antichymotrypsin in the form of granules, detected by specific immunohistochemistry at the light microscopy level and corresponding to material found in dilated endoplasmic reticulum of hepatocytes by electron microscopy. The patient had intermediate deficiency of δ1-antichymotrypsin. Thus, the hepatocyte accumulation of δ1-antichymotrypsin may indicate the presence of an export block resembling that of a closely-related protein, namely PiZ δ1-antitrypsin. It is proposed that hepatocyte storage of δ1-antichymotrypsin may be an expression of an inborn error of metabolism bearing the characteristics of endoplasmic reticulum storage diseases such as PiZ δ1-antitrypsin deficiency and hereditary hypofibrinogenaemia.

    Notes: The pathogenicity of Salmonella enterica serovar Typhimurium has traditionally been correlated with its ability to survive and grow in macrophages. Macrophage-derived production of nitric oxide (NO) has been implicated as a major innate defence, restricting bacterial proliferation both in macrophage cultures and in mice. In the present study, we show that the ability of primary murine dendritic cells (DCs) to ingest Salmonella is low, but greatly enhanced by serum complement. Ingestion of bacteria was followed by the expression of inducible nitric oxide synthase (iNOS), as well as by NO production. iNOS mRNA was detected as early as 6 h post infection and production of NO 12 h post infection, rising further at 16 h post infection. Inhibition of the iNOS activity with the inhibitor N-monomethyl-l-arginine or using DCs from iNOS–/– mice resulted in increased intracellular bacterial yields. To further define the potential defensive role of DC-derived NO, the actual intracellular replication rate of S. Typhimurium in DCs was measured. DC-derived NO was shown to exert a bactericidal effect, whereas the effect of NO in macrophage-like J774-A.1 cells was found to be bacteriostatic. These results identified an important role for NO in restricting S. Typhimurium survival in DCs, indicating that DCs may actively participate in the innate defence against intracellular pathogens.

    Notes: In a case-control study we compared the prevalence of heterozygous deficiency of two closely related anti-neutrophil protease inhibitors, α1-antitrypsin and α1-antichymotrypsin, in 172 consecutive children with asthma. In a cohort study the clinical spectrum and severity were compared. On the basis of family studies 5/172 (2.9%) were classified as heterozygotes for α1-antichymotrypsin deficiency, a high prevalence compared with that of an unselected adult population (prevalence ratio 4.5 (1.7–11.9), P〈0.005). This finding suggests that the carrier state of this rare allele (prevalence 0.64%) may predispose to asthma in children. Among these heterozygous patients the prevalence of positive RAST tests for foodstuffs was significantly increased (prevalence ratio 4.8 (1.7–13.2). P〈0.005) and 2/5 manifested food allergy with Quincke oedema. Either the PiMZ or SZ phenotype of α1-antitrypsin deficiency was found in 12 (7.0%) of the 172 patients, a prevalence similar to that of a normal population (prevalence ratio 1.3 (0.67–2.6), P= 0.44). However, the asthma was more severe among the Z allele carriers, judged by the number of hospital admissions, compared with the non-Z asthmatic children (mean 2.92 vs. 1.72, P〈0.05). The results indicate that heterozygous deficiency of protease inhibitors directed against neutrophil proteases may affect the severity and clinical spectrum of childhood asthma, and to some degree be predisposing.
