    Description / Table of Contents: Summary Lymphocytes of three patients suffering from chronic lymphocytic leukemia were studied electromicroscopically before and during ALS-treatment and after in-vitro-incubation with ALS. During the treatment, no qualitative or quantitative changes were shown. After incubation with ALS different changes could be demonstrated. The findings are discussed.
    Notes: Zusammenfassung Bei drei ALS-behandelten Patienten mit chronischer Lymphadenose wurden die zirkulierenden Lymphozyten vor und während der ALS-Gabe sowie nach Inkubation mit ALS elektronenmikroskopisch untersucht. Die Lymphozyten zeigten während der Behandlung weder qualitative noch quantitative (Einteilung in 4 Gruppen, Ausdifferenzierung) Veränderungen. An den ALS-inkubierten Lymphozyten waren verschiedenartige Schäden nachzuweisen, die mit entsprechenden Befunden anderer Untersucher verglichen werden.

    Description / Table of Contents: Zusammenfassung Es wurde versucht, mit dem IBM-Zellseparator an gesunden Personen für eine Transfusion ausreichend große Mengen an Granulozyten bei optimalem Reinheitsgrad gegenüber den anderen Blutzellen zu isolieren. Es konnten maximal 1,5×109 Granulozyten bei einem Spender isoliert werden, was etwa dem Gehalt einer Frischblutkonserve entspricht. Der Grund für die geringe Ausbeute wird in der Antikoagulierung des Blutes mit ACD gesehen. Bei Verwendung von Heparin als Antikoagulans ist eine wesentliche Verbesserung der Ausbeute zu erwarten.
    Notes: Summary In summary only that number of granulocytes can be obtained from normal persons as are contained in an amount of 500 ml fresh blood. But contrary to fresh blood, there is only a very small contamination with lymphocytes and thrombocytes. As known from the literature, the amount of 1.5×109 granulocytes is not sufficient to produce a therapeutic effect. On the other hand, the transfusion of donor granulocytes obtained in this way can only be repeated as often as the transfusion of such amount of erythrocytes is allowed. So the isolation of a sufficient amount of granulocytes from normal subjects for transfusion is not possible with the cell separator by this method.

    Description / Table of Contents: Zusammenfassung Bei der Blutzellisolierung mit dem NCI-IBM-Zellseparator wurde an Normalpersonen die maximale Ausbeute an Thrombocyten, Lymphocyten und Granulocyten sowie der optimale Reinheitsgrad gegenüber anderen Blutzellen untersucht. Insgesamt wurden 40 Zentrifugationen an 15 Normalpersonen durchgeführt. Die Anticoagulierung erfolgte ausschließlich mit ACD. An Stichproben wurde die Zellverteilung in der Zentrifuge in Abhängigkeit von der Drehzahl bestimmt und daraus die für Zellausbeute und Reinheitsgrad günstigste Entnahmeschicht und Zentrifugendrehzahl ermittelt. Als günstigste Bedingungen ergaben sich zur Gewinnung von Thrombocyten die Entnahme aus der Leukocytenschicht bei 1 750 Upm (220 g), zur Gewinnung von Lymphocyten ebenfalls dic Entnahme aus der Leukocytenschicht bei 1 750 Upm (220 g) und zur Gewinnung von Granulocyten die Entnahme aus der äußersten Erythrocytenschicht bei 1 750 Upm (220 g). Mit diesen Bedingungen wurden Dauerentnahmen von Thrombocyten, Lymphocyten und Granulocyten vorgenommen. Die Thrombocytenausbeute betrug 4,8×1011 (Entnahmedauer 2 Std, Reinheitsgrad Thrombocyten : Erythrocyten=1:0,0003; Thrombocyten:Lymphocyten=1:0,007; Thrombocyten : Granulocyten=1:0,0004). Die Lymphocytenausbeute betrug 3,55×109 (Entnahmedauer 2 Std, Reinheitsgrad Lymphocyten : Erythrocyten=1:0,3; Lymphocyten : Granulocyten=1:0,03; Lymphocyten : Thrombocyten=1:100). Die Ausbeute an Granulocyten war 1,5×109 (Entnahmedauer 1/2 Std, Reinheitsgrad Granulocyten : Thrombocyten=1:2; Granulocyten : Lymphocyten=1:0,01). Eine Trennung der Granulocyten von den Erythrocyten war bei der angewandten Methode nicht möglich.
    Notes: Summary In normal subjects the maximum yield of platelets, lymphocytes and granulocytes, and the optimum degree of purity in regard to the other blood cells was examined by separation with the NCI-IBM blood cell separator. 40 centrifugations were carried out with blood from 15 normal subjects. For anticoagulation ACD was used exclusively. First, the distribution of the cells in the centrifuge depending on g-forces was calculated. The best layer and the most suitable g-force were evaluated for the yield of cells and the degree of purity. Platelets and lymphocytes could be withdrawn from the buffy coat at 1750 rpm (220 g). Granulocytes could be withdrawn from the outer layer of red blood cells at 1750 (220 g). Under these conditions the yield of platets was 4.8×1011 (withdrawal time 2 hours, degree of purity platelets: red blood cells=1:0.0003; platelets:lymphocytes=1:0.007; platelets:granulocytes=1:0.0004). The yield of lymphocytes was 3.55×109 (withdrawal time 2 hours, degree of purity lymphocytes:red blood cells=1:0.3; lymphocytes:granulocytes=1:0.03; lymphocytes:platelets=1:100). The number of granulocytes was 1.5×109 (withdrawal time 30 min, degree of purity granulocytes:platelets=1:2; granulocytes:lymphocytes=1:0.01). A separation of granulocytes from red blood cells was not possible with the method applied.

    Notes: Geobacter pelophilus is capable of dissimilatory Fe(III)-reduction on solid phase Fe(III)-oxides by means of surface attachment and direct electron transport to Fe(III), in part mediated by outer membrane c-type cytochromes. A study was undertaken to characterize surface colonization patterns, gene expression, and mineral transformation by this organism. The gene ferA (Geobacter sulfurreducens outer membrane Fe(III) reductase cytochrome c) was used as a target for PCR based molecular detection methods for visualizing G. pelophilus surface colonization. Protein extracts were prepared from solid-phase cultures, and cytochrome c content assessed. Mineral transformations were followed by X-ray photoelectron spectroscopy (XPS). Results of in situ (IS) RT-PCR experiments demonstrate that G. pelophilus attaches and grows at ferrihydrite mineral surfaces. Fluorescently-labeled cells were observed after IS-RT-PCR experiments, suggesting that G. pelophilus contains a cytochrome c sequence similar to ferA in G. sulfurreducens which is expressed in the presence of ferrihydrite. Protein extracts possessed high mass c-type cytochromes of similar size to those found in G. sulfurreducens. In addition, unique high-mass c-type cytochromes were also detected. XPS analysis demonstrated mineral transformation to occur, mediated by the surface associated population. This study demonstrates that G. pelophilus attaches to Fe(III)-oxide surfaces, reduces the Fe(III) oxides at the surface, produces c-type cytochromes under these growth conditions, and expresses cytochrome c-encoding genes as measured by in situ molecular detection techniques.
