    Notes: Abstract We examined 64 normal Japanese chromosomes to determine the heterozygosities and allelic frequencies of 358 dinucleotide-repeat marker loci spanning the whole human genome. Comparisons of the data for each marker in the Japanese population sample with data for the same markers among Caucasian samples in the Genome Database (GDB) revealed a slightly lower average of heterozygosity in Japanese (71% vs 79%). Although the majority of the markers were as informative as in Caucasians, some in our sample were uninformative due to low heterozygosity; 38 loci revealed heterozygosities lower than 50% and 11 of these were less than 30%. Furthermore, allelic distributions at many of the marker loci were quite different in the two racial groups. Since such differences will influence statistical analyses between markers and disease loci, our data will be essential for linkage analyses, sib-ship pair analyses, and association studies involving the Japanese population.     Notes: Summary To clarify whether activity of the sympathetic nervous system is decreased in streptozotocin-induced diabetic rats, noradrenaline turnover, which is a reliable indicator of sympathetic nervous system activity, was measured in the interscapular brown adipose tissue, heart and pancreas of streptozotocin diabetic rats. Results from studies using inhibition of noradrenaline biosynthesis with α-methyl-p-tyrosine demonstrated significant reductions (p〈0.05-0.001) in sympathetic nervous system activity in the interscapular brown adipose tissue, heart and pancreas of streptozotocin (65 mg/kg) diabetic rats, compared with measurements in streptozotocin (35 mg/kg) diabetic and saline-control rats. The daily injections of neutral protamine Hagedorn insulin to streptoz/otocin (65 mg/kg) diabetic rats prevented the decrease of noradrenaline turnover in the interscapular brown adipose tissue and heart significantly (p〈0.02), but this was less marked in pancreas, compared with non-treated streptozotocin (65 mg/kg) diabetic rats. Furthermore reduced noradrenaline turnover was also observed in the control rats which showed comparable changes in body weight to the rats injected with streptozotocin (65 mg/kg). These results suggest that poorly controlled streptozotocin diabetic rats may have reduced sympathetic nervous function, and that insulin therapy might prevent this.

    Notes: Summary A case of congenital peripheral medullo-epithelioma arising in the sciatic nerve of a 6-month-old boy is reported. Histologically, areas of primitive neural tumor merged with areas showing differentiation into ependymoma, astrocytoma, oligodendroglioma, and ganglioneuroma. These lines of differentiation were confirmed by electron microscopy and immunohistochemistry. Despite the malignant nature of the tumor, 7 years after amputation of the affected leg, the child is alive without recurrence.

    Notes: Summary The distribution and ultrastructure of Alzheimer's neurofibrillary tangles (ANT) in the brain stem, hypothalamus, and Ammon's horn were studied in four patients with postencephalitic parkinsonism of Economo type (PEPE). The distribution of ANT was as previously reported; the pattern of distribution resembled to that of amine-containing nerve cells. Ultrastructurally, ANT revealed twisted tubules (TT), but straight tubules (ST) of 150 Å width were also found in the locus ceruleus of three cases; sometimes, TT and ST were mixed in a single neuron. Whether the coexistence of TT and ST in the locus ceruleus is a characteristic ultrastructural feature of ANT in PEPE or a regional peculiarity could not be determined. Ultrastructurally, ANT in PEPE were identical to those found in the brains of patients with Alzheimer's disease or senile dementia.

    Notes: Summary Experimental neurofibrillary change was produced in rabbit brains by daily subcutaneous aluminum tartrate injection for 40 days. The production of experimental neurofibrillary changes was confirmed by immunostaining with antibodies against neurofilament triplet proteins and the brain tissue was studied immunohistochemically with antibodies against microtubule-associated protein (MAP) 2 and ubiquitin. The hippocampal neurons of the chronically aluminum-intoxicated rabbit brain showed diminished staining of dendrites by anti-MAP2 antibody. The length of anti-MAP2-positive dendrites in hippocampus was significantly shorter than that of the control brain. In the cortex somata of a subset of pyramidal neurons were intensively stained by anti-MAP2 antibody, while the MAP2 immunoreactivity of distal dendrites was diminished. The immunostaining by anti-ubiquitin antibody revealed the positive staining of the neurons bearing experimental neurofibrillary changes in the lower brain stem nuclei. It is speculated that MAP2 dislocation and ubiquitination are accompanying phenomena of the production of experimental neurofibrillary changes in chronically aluminum-intoxicated rabbit brains.

    Notes: Summary Tissue from seven patients with tuberous sclerosis and subependymal giant-cell tumors was examined with special stains, immunohistochemistry, and electron microscopy. Immunoreactive glial fibrillary acidic protein (GFAP) was not found in the giant cells of four tumors, but was present in some tumor cells in the other three. Immunoreactive S-100 protein was present in tumor cells of six cases; it was also seen in more tumor cells than was GFAP. Electron microscopy was similar in all cases and showed that the tumor cells had numerous organelles — many dense bodies thought to be primary lysosomes, swollen mitochondria, Golgi complexes, rough and smooth endoplasmic reticulum, free ribosomes, and sparsely distributed intermeadiate filaments. In one case, neurosecretory granules, microvilli, and synapses were observed. In another subject, prominent, thick bundles of glial filaments were seen. These findings suggest that the tumor is made up of unique cells in addition to cells with recognizable neuronal or astrocytic features.

    Notes: Abstract In cultures of Alaria crassifolia Kjellman, unfertilized eggs developed normally into haploid sporophytes which differentiated into a holdfast, stipe and blade with a midrib. The terminal cells of a male gametophyte grew apogamously into haploid sporophytes with narrow blades which lack the midrib. Further, diploid gametophytes were formed by apospory from vegetative cells of a diploid sporophyte. They were monoecious and their fertilized eggs developed into tetraploid sporophytes. Nuclear phases of the sporophytes and gametophytes concerned were confirmed by cytological observations.

    Notes: Abstract The distribution and abundance of Japanese common squid [Todarodes pacificus (Steenstrup, 1880)] paralarvae off southern Kyushu Island near the Kuroshio were examined in relation to water types in the region. Surveys were conducted in 1996 and 1997 using paired, 70 cm diameter Bongo nets. Temperature–salinity plots from 0, 50 and 100 m depths were used to assign sampling stations to three water types: inshore, mixing and Kuroshio. In total, 4103 T. pacificus paralarvae were collected at 59 of 72 stations. Catch densities at positive stations ranged between 8 and 4282 ind./50 m2 sea surface area. Catches during both years were highest in mixing waters, followed by inshore and Kuroshio waters. Total catches were highest just north of the oceanographic front at the inshore edge of the Kuroshio and declined with increasing distance away from the front. Catches of paralarvae ≤1.0 mm mantle length also peaked near the front. In both years, ≥85% of all paralarvae and ≥69% of those ≤1.0 mm mantle length occurred less than 25 km from the front. We conclude that most spawning off southwest Kyushu occurs near the frontal zone, where Kuroshio and inshore waters meet.

    Notes: Abstract As part of our effort to isolate and characterise the von Hippel-Lindau (VHL) disease gene, we constructed a physical map of chromosome 3p25-26 by fluorescence in situ hybridisation (FISH) studies on a panel of cytogenetic rearrangements involving this region. Biotinylated cosmid and lambda probes were hybridised to metaphase chromosome spreads and positioned with respect to each cytogenetic breakpoint. These studies unequivocally established the order of five loci linked to the VHL disease gene: cen-(RAF1,312)-D3S732-D3S1250-D3S601-D3S18-pter and determined the position of three other probes within this map. These results ordered RAF1 and D3S732 for the first time, confirmed the localisation of D3S1250 between RAF1 and D3S601 and determined the position of D3S651 with respect to other chromosome 3p25-p26 loci. The establishment of an ordered set of cytogenetic aberrations will enable the rapid assignment of polymorphic and nonpolymorphic cloned sequences within the chromosome region 3p25-p26.
