    Notes: Summary Several thrombogenic abnormalities are associated with diabetes. To investigate the underlying molecular mechanisms, we examined the effects of advanced glycation endproducts (AGE), non-enzymatically glycated protein derivatives, on the production of prostacyclin (PGI2), an anti-thrombogenic prostanoid, and of plasminogen activator inhibitor-1 (PAI-1), a fast-acting serine protease inhibitor of fibrinolysis, in human microvascular endothelial cells (EC). Firstly, AGE-bovine serum albumin (BSA) but not non-glycated BSA, was found to considerably decrease the production of PGI2 to about two-thirds of the control value. Secondly, quantitative reverse transcription-polymerase chain reaction showed that AGE-BSA increased the EC levels of mRNA coding for PAI-1, this being associated with a concomitant increase in the immunoreactive PAI-1 contents and the anti-fibrinolytic activity. Thirdly, the effects of AGE on PGI2 and PAI-1 syntheses in EC were found to be mediated by a receptor for AGE (RAGE) because antisense DNA against RAGE mRNA could reverse the AGE effects. Further, it was found that AGE decreased the intracellular cyclic AMP concentrations in EC and that cyclic AMP agonists such as dibutyryl cyclic AMP, forskolin and PGI2 analogue reduced the AGE-stimulated PAI-1 production, suggesting the involvement of cyclic AMP in the AGE-signalling pathway. The results thus suggest that AGE have the ability to cause platelet aggregation and fibrin stabilization, resulting in a predisposition to thrombogenesis and thereby contributing to the development and progression of diabetic vascular complications.     Description / Table of Contents: Résumé Ce cas est celui d'une enfant qui présentait à la naissance une large masse rétroauriculaire que le diagnostique clinique identifiait comme étant un hémangiome. La radio thérapie eut de très bons résultats. A onze ans on pratiqua l'ablation d'un sarcome cérébelleux qui ne semblait pas avoir été causé par la tumeur rétroauriculaire. Après traitement à moutarde nitrogénée et après trois appliquations de rayon X s'échelonnants pendant neuf mois et demi-soit une dose de radiation d'environ 7,953 r — son état s'aggrava. Pendant quatre traitements de «neutron capture», dirigés principalement sur la région sous-occipitale, on remarqua une amélioration spectaculaire, la malade pouvant s'asseoir et étant même capable de converser pendant quelque temps. Après ce traitement, la tumeur qui s'étendait dans la région sous-occipitale et dans la cou disparut, ainsi que presque toute la tumeur qui se trouvait dans le tiers dorsal du cervelet particulièrement dans la région la plus proche de l'aire sous-occipitale. Dans le tiers moyen cérébelleux on trouva également des agrégats de tumeurs plus ou moins grands, dont quelques-uns calcifiés, qui étaient isolés par un tissu conjonctif hyperplasique. Dans le tiers ventral du cervelet la tumeur ne cessa de croître dans la région non-irradiée par les neutrons. La tumeur était un sarcome d'origine périvasculaire située principalement dans la couche réticulaire cérébelleuse. La vie de la malade fut prolongée d'environ huit mois par le traitement de «neutron capture».
    Notes: Summary This paper deals with a girl who at birth had a large retroauricular mass diagnosed clinically as hemangioma. It responded well to X-ray therapy. When she was 11 years old, a sarcoma was removed from the cerebellum. No evidence could be found that the sarcoma had originated from the retroauricular tumor. After the use of nitrogen mustard locally and after three courses of X-ray therapy to the tumor over a period of 9 1/2 months — a total radiation dose of approximately 7,953 r — the patient was growing moribund. During 4 neutron-capture treatments, which were directed chiefly toward the suboccipital region, striking improvement occurred — to such an extent that the patient was able for a time to sit up in a wheelchair and converse. As a result of the therapy, all tumor which had spread suboccipitally and into the neck vanished, as did also virtually all tumor in the dorsal third of the cerebellum, i.e., in the region receiving the largest concentration of thermal neutrons. In the middle third of the cerebellum large and small tumor aggregates, some of them calcified, were necrotic and were walled off by hyperplastic connective tissue. In the ventral third of the cerebellum, in a region presumably out of the range of an effective concentration of thermal neutrons, the tumor grew unimpeded. Evidence indicated that the sarcoma in our case originated in the vascular sheath, chiefly about vessels in the granular layer of the cerebellum. Life was prolonged approximately 8 months by the neutron-capture therapy.

    Notes: Summary Interspecies difference in expression of the c-myc gene between two human and three rat glioma cell lines was studied with use of a human c-myc probe. The c-myc deoxyribonucleic acid (DNA) fragments detected at higher stringency in Southern blotting, showed a difference in size and gene copy number between human and rat glioma cells. The c-myc transcript was detected at both higher and lower stringencies in Northern blotting in human glioma cells, whereas it was demonstrated only at lower stringency in rat glioma cells, and the c-myc transcript was seen in cytoplasms of both glioma cells by in situ hybridization. The c-myc protein, if examined with anti-human c-myc protein monoclonal antibody, was observed as two separated components in Western blotting and localized immunocytochemically in nuclei in human glioma cells, whereas it was detected as three separate forms in Western blotting and shown in both nuclei and cytoplasm in rat glioma cells. The above discrepancy in manifestation of c-myc DNA fragments, transcript and protein could be due to the difference in nucleotide sequence of c-myc gene between human and rat glioma cells.

    Notes: Abstract. Nuclear magnetic resonance (NMR) quantum computation in a crystal lattice holds more promise for scalability than its solution NMR counterpart, but dephasing is a severe concern. Pulse sequence refocusing can help bring the qubit dephasing time closer to the limit of the intrinsic decoherence time, but the intrinsic transverse relaxation time (T2) and the longitudinal relaxation time (T1) of the crystal must be sufficiently long for a successful implementation. We discuss these time scales and their relation to parameters relevant to quantum computation for several crystal types, discussing in detail the examples of CaF2, MnF2, and CeP. Included in the calculation of coherence times for CeP is the development of spin-wave spectra in a type-1 antiferromagnetic FCC lattice.

    Notes: Abstract In our previous study, $${1 \mathord{\left/ {\vphantom {1 {f^\beta }}} \right. \kern-\nulldelimiterspace} {f^\beta }}$$ -type power spectrum with the spectral exponent β significantly greater than zero was found in the variability of soleus H-reflex amplitudes. This result indicated that the H-reflex variability was time-correlated owing to fractal characteristics. Furthermore, it was also suggested that the fractal characteristics were generated at the spinal level. The purpose of the present study was to investigate whether the fractal nature of the H-reflex variability was influenced by the loss of supraspinal input. Six healthy normal subjects and seven patients with spinal cord injury participated in this study. Soleus H-reflexes were evoked every l s from both legs simultaneously (stimulation intensity: motor threshold) and 1050 successive amplitudes of the H-reflex were recorded. The H-reflex sequence evoked from each leg was analyzed by “coarse graining spectral analysis” to calculate the spectral exponent β. The value of β was used to evaluate the level of time-correlation (fractal correlation). Cross-spectral analysis was used to evaluate the degree of synchronization between the H-reflex sequences evoked from both legs. The β values for normal subjects (0.84±0.33, left leg; 0.88±0.34, right leg) were significantly greater (P〈0.001) than those for patients (0.31±0.18, left leg; 0.32±0.14, right leg), suggesting that the H-reflex sequences for normal subjects were more time-correlated than for patients. In the frequency range less than 0.2 Hz, the coherence of both legs was high (0.41±0.14) for normal subjects as compared to 0.20±0.12 for patients (P〈0.005). In this frequency range, the phase was almost 0 rad for normal subjects, indicating that the H-reflex variabilities of both legs were synchronized. These results suggested that (1) the strong fractal correlation observed in the H-reflex sequences for normal subjects was associated with supraspinal input, and (2) such supraspinal input had equal influence on the reflex arcs of the soleus of both legs.
