    Notes: From a combined theoretical and experimental study of 1,2,4,5-tetrachlorobenzene (TCB) we conclude that this crystal shows many interesting effects. In agreement with earlier optical measurements, which probe the complete vibron band structure of TCB, we calculate that several of the vibron modes have the dispersion of a one-dimensional crystal with stacks of molecules along the a axis. The inclusion of fractional atomic charges in the atom–atom potential used in the calculations is absolutely necessary to obtain the correct vibron bandwidths. Also the sign of the vibrational coupling matrix elements, which is given correctly by the calculations, is determined by these charges. For other properties, such as the crystal stability, the phonon frequencies, the site splitting in the vibron bands, and also the dispersion of the lower frequency vibrations, the interactions between the "one-dimensional'' stacks are essential, however. The calculations predict further, in qualitative agreement with the Raman spectra, that the splitting between the pairs of inequivalent vibron bands in α-TCB (the site splitting) is significantly larger than the factor group splitting in β-TCB. Finally we have found that, for those vibrations where the conditions are shown by the calculations to be favorable, the 35Cl/37Cl isotope effects are clearly visible in the Raman spectra.

    Notes: The degradation of a panel of monoclonal antibodies (MoAb) bound to surface IgM (sIgM) was studied in three human Burkitt's lymphoma cell lines. The panel included MoAb that recognize several distinct epitopes associated with the F(cμ)5 domain, the cμ2 domain, and δ or λ light chains. The amount of degraded MoAb and the rate of their degradation varied considerably between the various antibodies. Properties of MoAb such as avidity or ability to cross-link sIgM did not significantly influence their degradation. The most consistent correlation between rate of degradation and MoAb used was the location of the epitope recognized by the individual MoAb. Thus, 7 out of 8 anti-light chain MoAb were degraded at a higher rate than 5 out of 5 anti-F(cμ)5 MoAb. One anti-cμ2 MoAb was degraded at a rate similar to the majority of anti-light chain MoAb. The intracellular transport of an anti-δ light chain MoAb and an anti-F(cμ)5 MoAb was studied in detail by subcellular fractionation in sucrose gradients. We found that the anti-δ light chain MoAb was transported more rapidly to lysosomes than the anti-F(cμ)5 MoAb, showing that they were sorted differently intracellularly.

    Notes: This was a multicentre, double-blind, randomized group comparative study in which 77 children, aged 6–16 years, received 2% nedocromil sodium eye drops and 72 received placebo, one drop into each eye twice daily. The treatment period was 4 weeks, covering the peak birch pollen season. Prior to the start or the season, patients who had mended the clinic the previous 2 years because of seasonal allergic conjunctivitis (SAC) to birch pollen, entered a one week baseline period during which symptoms were assessed, diary cards completed, and routine sampling of blood and urine earned out. The double-blind treatment period then commenced at the onset of the birch pollen season. Patients parents kept daily diary record cards of eye symptom severity and concomitant therapy. Conjunctivitis was mild in both treatment groups but nedocromil sodium was more effective than placebo in controlling symptoms. During the 2–3 weeks of peak pollen counts, this therapeutic effect was statistically significant for itching (P 〈0–01), watering (P 〈0.05) and total symptom score (P 〈0.01), but was not significant for grittiness (P= 0.08) or redness (P = 0.06). Global opinions of efficacy showed no difference between treatments, due to a high placebo effect (however, the diary card data indicated a significant improvement with nedocromil sodium). We therefore conclude that nedocromil sodium 2% eye drops, administered twice daily, is an effective treatment for SAC in children.

    Notes: The intracellular pathway of a endocytosed glycoprotein has been studied in rainbow trout (Oncorhynchus mykiss) liver by means of differential and isopycnic gradient centrifugation. When 125I-tyramine-cellobiose-labelled asialoorosomucoid was injected intravenously, the protein was removed rapidly from the blood by hepatic endocytosis. The tracer was localized initially in a small, slowly sedimenting organelle (endosome) and transferred to the denser lysosomes where degradation took place. This process took several hours at the acclimated temperature of 10 ° C. The transport could be accelerated markedly or retarded by transfer of fish to higher or lower temperatures respectively. The intracellular transport steps were more susceptible to changing temperature than the internalization process. In particular, the degradation was temperature sensitive. These results may have implications for the effects of changing temperature on immune function in fishes. The impairment of transport upon a rapid temperature decrease, will reduce the rate of antigen processing and presentation in these animals.

    Notes: The catabolic fate of circulating collagen (Col) in the Atlantic salmon (Salmo salar) was studied. Serum t1/2 and organ distribution of circulating Col in salmon were determined using Col conjugated with l25I-tyramine cellobiose (125I-TC), a low molecular weight adduct which is trapped intralysosomally at the site of uptake. Intravenously administered l25I-tyramine cellobioselabelled Col type I was prepared either from salmon skin (sCol) or rat skin (rCol). Biphasic clearance kinetics of l25I-TC-sCol in salmon were apparent, with 78% being removed from the circulation in an initial rapid α-phase (t1/2(α) = 2.4 min), and 22% being removed more slowly in a terminalβ-phase (t]2(β) = 25.8 min). Serum half life of 125I-TC-rCol was found to be 5.4 min (in this type of experiment the number of data points allow the determination of only a monophasic decay slope). Approximately 90% of recovered radioactivity was found in the kidney of the fish. In comparative experiments, 74% of administered 125I-TC-sCol was cleared from the circulation of rats during an initial rapid α-phase with tl/2(α) = 0.8 min, and 26% was eliminated in the terminal β-phase with t1/2(β) = 7.2 min l25I-sCol was endocytosed and degraded in pure cultures of ral liver endothelial cells, which are the main site of clearance of circulating Col in the rat. Moreover, Col from the two species competed for the same receptor on cultured rat liver endothelial cells, Intravenous administration of tetramethyl rhodamine isothiocyanate-labelled sCol (TRITC-sCol) in salmon, and subsequent examination of sections of kidney in the fluorescence microscope, revealed that the fluorochrome was accumulated exclusively in discrete vesicles of sinusoidal lining cells. Analyses of kidney tissue 24h after intravenous administration of a mixture of fluorescein isothiocyanate-labelled latex beads and TRITC-sCol revealed no codistribution of the two fluorochromes, suggesting that the injected Col was taken up in cells different from macrophages. Purified pronephros macrophages prepared after simultaneous injections of stained beads and Col contained only fluorescein-labelled latex particles. Interestingly, the cells which had accumulated TRITC-sCol appeared to be equally distributed in both pronephros and the part of the kidney containing tubuli. We conclude that Col which gains access to the circulation of the Atlantic salmon is cleared mainly by uptake into sinusoidal lining cells of the kidney. These cells are distinct from phagocytosing macrophages, and morphologically similar to the highly specialized scavenging endothelial cells of mammalian liver sinusoids.

    Notes: The rigor mortis development of large Atlantic salmon was evaluated at a commercial plant using a sensory method, a mechanical rigorometer and low-frequency vibrations. The fish were classified according to rigor state using a neural network. Assessments were done with fish subjected to two different levels of handling stress prior to slaughter. White muscle high-energy phosphates and inosine monophosphate were used as indices of stress showing that handling stress may be considerable during commercial slaughter. Results indicate rigor assessment should be nondestructive and classificaton of rigor state was possible using a lowfrequency vibration method combined with a neural network.

    Notes: Xanthomonas campestris is a seedborne bacterium that causes black rot of crucifers. Substantial crop losses may result from the rapid spread of the bacteria under favourable conditions, especially those occurring during seedling production. A PCR-based method has been developed for the rapid and sensitive detection of the pathovars of X. campestris that affect crucifers. Primers were designed to specifically amplify a 619 bp fragment of the hrpF gene from X. campestris. Amplification products were not detected from other Xanthomonas species, or from other pathogenic or epiphytic bacteria occurring on these plants. To avoid false-negative results arising from the presence of amplification inhibitors in plant extracts, primers targeting a 360 bp section of the internal transcribed spacer (ITS) region from Brassica spp. were included in a multiplex PCR. The assay readily detected X. campestris pv. campestris infections in diseased plants and from bacterial colonies isolated on growth media, and was more sensitive and specific than traditional plating methods and a commercially available ELISA. A seed-washing protocol was optimized to allow the detection of a single artificially infected seed among 10 000 healthy seeds using the multiplex PCR.
