    Notizen: Abstract  Since orally given cytotoxic agents may cause intestinal disfunction, the effect of oral administration of three cytotoxics, i.e., methotrexate (MTX), cyclophosphamide (CPA), and ftoral, a derivative of 5-fluorouracil (5-FU), on the gastric, liver, and small-intestine cells of C57B1 mice was studied by transmission electron microscopy. Although no ultrastructural alterations could be detected in the cells of the first two organs, the epithelial cells of the small intestine showed a marked increase in size of their mitochondria. In the control animals the mitochondrial size was in the range of 0.04–1.8 μm (mean ±SE 0.54±0.01 μm). In the treated animals the size of the mitochondria ranged between 0.15 and 4.33 μm (mean ±SE 0.73 μm) for those treated with MTX, 0.24–2.88 μm (mean ±SE 0.80±0.02 μm) for those given CPA, and 0.28–5.3 μm (mean ±SE 1.18±0.48 μm) for those treated with 5-FU. These findings were significantly different from those obtained in controls (P〈0.0001). In addition, in animals treated with MTX the mitochondria of the jejunal cells were surrounded by channels of rough endoplasmic reticulum. The cytoplasm contained long, winding channels of smooth endoplasmic reticulum, vacuoles, and myelin figures. Fluid retention in the small intestine due to administration of cytotoxic drugs is suggested as a possible mechanism for distention of the mitochondria.

    Notizen: Summary A 65-year-old woman developed severe chorea as the presenting symptom of the hyperviscosity syndrome together with polycythemia vera and an ovarian cyst. Whole blood viscosity was 69.5 centipoise at a shear rate of 1.1 s−1 (normal 12.0–22.0 centipoise). After four phlebotomies the chorea subsided completely, and the blood viscosity became normal. Hemoglobin level was stable at normal range and then increased to 18.0 g/dl with recurrence of the faciobuccal dyskinesia that disappeared promptly following the phlebotomies. This case emphasizes that in elderly patients with neurological disorders, such as chorea, polycythemia should be strongly considered as one of the treatable causes.

    Notizen: Summary A patient with acute lymphoblastic leukemia (ALL) with cells containing two types of cytoplasmic inclusions is described. The inclusions appeared as glubular bodies containing electron dense material with homogenous structure and as crystalloid formations confined in organelles with structure similar to that of the surrounding mitochondria. In distinction to other reports, these structures were not related to the endoplasmic reticulum. The possibility that some of them represented altered mitochondria is discussed.

Die Morphologie der Zellen im peripheren Blut und im Knochenmark deutet darauf hin, daß diese der lymphatischen Reihe angehören, jedoch bleibt der Typ der Leukämie unsicher. Elektronenmikroskopische Untersuchung zeigt konzentrische oder spiralförmige lamellare Strukturen im Zytoplasma einer Anzahl dieser Zellen, was auf die Diagnose einer Lymphosarkom-Zell-Leukämie hindeutet.

The morphology of the cells in the peripheral blood and in the bone marrow suggested that they were of the lymphocytic series, but the type of leukemia was uncertain. Electron microscopic studies revealed concentric or spiraloid lamellar formations in the cytoplasm of a substantial number of the cells, suggesting the diagnosis of lymphosarcoma cell leukemia.

    Notizen: The present study describes a new low molecular weight factor released by muscle cells, which inhibits proliferation of tumor cells in vitro and in vivo, is highly specific towards tumor cells, and has no observable effect on normal cells' proliferation. What first prompted us to investigate this factor was the observation that tumor metastases are extremely rare in striated muscles. Co-culturing of striated muscle cells with malignant cells led to marked morphological alterations in the latter, in contrast to the same cells when incubated without muscle cells. A conditioned medium of striated muscle cells was prepared and its effect tested on a variety of cells. This conditioned medium (CM) inhibited proliferation of tumor cell lines of murine (B16 melanoma, Madison 109 lung carcinoma, MCA-105 sarcoma, ESB lymphoma), or of human origin (HTB-38 adenocarcinoma, T47D breast carcinoma, CX1 colon carcinoma). The proliferation of normal cells (bone marrow cells, fetal liver erythroid cells) was not affected by the CM. Flow cytometric analysis of B16 melanoma cells following incubation with the CM revealed that 63% ± 12 of the cells were in the G0/G1 phase of the cell cycle, compared to 47.8% ± 8 of cells incubated with a medium (not conditioned) only. The activity of the CM and of certain fractions thereof was also demonstrated in vivo: they prevented tumor growth in mice inoculated intraperitoneally with MCA-105 sarcoma cells. Partial purification of the CM revealed that the active component was a non-proteinaceous compound with a molecular weight of about 500 D. The results clearly suggest that muscle cells produce a low molecular weight factor which can selectively inhibit the proliferation of tumor cells in vitro and in vivo. This factor is neither species nor tumor specific.
