    Notes: Abstract Lectins from four marine algal species were examined for interaction with human platelets. The lectin designated hypnin A, from the red algaHypnea japonica, inhibited adenosine diphosphate (ADP)-or collagen-induced human platelet aggregation in a dose-dependent manner. Complete inhibition was observed at concentrations of 100 and 5 μg/ml of the lectin with, ADP (2 μM) and collagen (0.2 μg/ml)-induced platelet aggregation, respectively. At the inhibitory concentration of 0.5 to 100 μg/ml, the lectin did not induce aggregation of resting platelets. Lectins from the other three algal species also inhibited ADP-induced human platelet aggregation. These results indicate that the algal lectins are a new group of inhibitors and may be useful to study glycoconjugates on platelet membranes and to design novel platelet aggregation inhibitors.

    Notes: Abstract The tropical Indo-Pacific moray eel Lycodontis nudivomer possesses a mucous skin secretion which shows hemolytic, toxic and hemagglutinating activities. The active components were extracted from the mucous secretion with 0.02 M phosphate buffer (pH 7.5) and purified by DEAE-cellulose and Sephadex G-200 column chromatography. The component(s) responsible for the hemolytic-toxic activities differ(s) from that (those) causing the hemagglutinating activity. The hemolysin was unstable in the presence of heat, acidic and alkaline media, and several organic solvents. It was undialyzable throught a cellophane membrane, and showed an absorption maxium at 280 nm in the phosphate buffer (pH 7.5). The hemolytic activity was completely lost when treated with trypsin, indicating that the hemolysin is a protein. Histochemical examination on the epidermis of L. nudivomer revealed the presence of clubbed peculiar cells, in addition to common mucous cells, which we speculate produced the hemolytic-toxic component(s). Eight other species of morays were found to possess clubbed peculiar cells similar to those of L. nudivomer in their epidermis. There are marked differences, however, in the abundance of these cells among these muraenids.

    Notes: Abstract Serum amino acid pattern, blood ammonia, and serum valproic acid (VPA) levels were estimated in 75 epileptic children with and without VPA therapy. The incidence of patients with hyperammonaemia (〉60 μmol/l) in patients treated with only VPA or with a combination of VPA and other antiepileptic drugs was 19% and 20% respectively. Hyperammonaemia was not observed in patients receiving other drugs. Elevated levels of serum glycine, alanine and serine were more common in epileptic patients treated with VPA alone and with VPA in combination with other drugs than in patients receiving other drugs. A significant positive correlation was found between the serum levels of glycine and alanine, of glycine and serine, and of serine and alanine. These findings may indicate a mitochondrial dysfunction caused by VPA.

    Notes: Abstract We have applied atomic force microscopy (AFM) to the measurement of BAL 31 nuclease activities. BAL 31 nuclease, a species of exonuclease, is used to remove unwanted sequences from the termini of DNA before cloning. For cutting out only the appropriate sequences, it is important to know the nuclease properties, such as digestion speed and the distribution of the lengths of the digested DNA. AFM was used to obtain accurate measurements on the lengths of DNA fragments before and after BAL 31 nuclease digestion. We analyzed 4 DNAs with known number of base pairs (288, 778, 1818, and 3162 base pairs) for correlating the contour length measured by AFM with the number of base pairs under the deposition conditions used. We used this calibration for analyzing DNA degradation by BAL 31 nuclease from the AFM measurement of contour lengths of digested DNAs. In addition, the distribution of digested DNA could be analyzed in more detail by AFM than by electrophoresis, because digested DNA were measured as a population by electrophoresis, but were measured individually by AFM. These results show that AFM will be a useful new technique for measuring nuclease activities.

    Notes: Summary In order to investigate the developmental mechanism of cerebral aneurysms, the in vivo flow pattern around human cervical carotid bifurcations was studied by flow visualization using digital subtraction angiography with an isotonic contrast medium. The blood stream containing the medium impinged on the apex, then proceeded along the walls of the branches. After opacification of the whole lumen around the apex, most of the medium was carried away, while some remained for a few seconds at the carotid sinus. In the internal carotid artery, the blood struck the wall at an oblique angle near the tops of the arterial curvatures. In cases with atheromatous plaque or kinking of the branch, the blood passed through the stenosed segment and moved upstream, indicating turbulence. The study suggests that haemodynamic forces around the apex consist mainly of impingement on the apex and shear stress to the wall at and around the apex. In branches, high shear stress seems to exist. It might be possible that high shear stress causes degenerative changes in the endothelial layer, initiating the formation of saccular and fusiform cerebral aneurysms.

    Notes: Summary The distribution of125I-labelled recombinant mouse interferon-β (rMuIFN-β) in normal and glioma (203 glioma) bearing mice was studied by radioassay and macro-autoradiography at 15 and 30 min after a single intravenous injection. The level of rMuIFN-β in the spleen was about 20-fold higher than in serum. Concentrations higher than the serum level was detected in the lung, liver and kidney. The concentration of rMuIFN-β in the brain was 8% of the serum level and the concentration in the glioma 30 min after administration was about 10-fold higher than in normal mouse brain. Macro-autoradiographic study demonstrated a wide distribution range and selective uptake in glioma tissue. Furthermore, we found that mouse gliomas were sensitive to mouse IFN-β. Our findings demonstrate that in the mouse glioma model, intravenously administered interferon reaches the tumour.

    Notes: A new method for optimizing the glossy appearance of facial make-up is proposed. Reflections from skin make-up comprise two components, specular and diffuse. Glossiness of the skin can be optimized by controlling the magnitude of the specular component in applied cosmetic foundation. To determine optimal appearance of glossiness, a series of images of a woman's face with make-up varying in the magnitude of specular reflection were synthesized using computer-graphics techniques, and 15 female volunteers selected the most beautiful skin from among those simulated faces. For calculating the specular component of the facial images, six bidirectional reflectance distribution functions (BRDFs) were generated from the goniophotometric spectra of two foundation membranes that differed in reflectivity from each other. Results of the image evaluations on dark, normal or bright skin are shown. In addition, evaluation of images in which the specular component was shifted to blue caused by blue pearl pigments is shown.
