    Notes: The adiabatic electron affinities of five second row atoms (Al, Si, P, S, Cl) and their monoxides and dioxides were determined using six different density functional or hybrid Hartree–Fock/density functional methods. The 15 species selected form a convenient closed set for which reliable experimental electron affinities exist for 13 of the species. Zero-point vibrational energy corrected electron affinities are also reported. Equilibrium geometries and vibrational frequencies were determined with each density functional method. The method based on the Becke exchange functional and the Lee–Yang–Parr correlation (BLYP) functional reproduced the experimental electron affinities most accurately, having an average absolute error of 0.15 eV. Using this functional, the electron affinities were predicted for SiO and SiO2, molecules for which electron affinities are not known experimentally, as 0.11 eV and 2.03 eV, respectively. It is concluded that the accuracy observed for density functional theory methods applied to first row atoms and molecules extends to molecules containing second row atoms and that density functional theory continues to provide a computationally affordable means of producing electron affinities reliable to within a few tenths of an eV of definitive experimental values. © 1999 American Institute of Physics.

    Notes: High-level coupled-cluster theory with large basis sets was used to determine the optimized geometries and harmonic vibrational frequencies for the ground and low-lying excited electronic states of the CH2N and CH2P radicals. Additionally, isotropic hyperfine splitting constants were determined for the C2v CH2X, trans-HCXH and cis-HCXH (where X=N and P) isomers as a gauge of the delocalization of the unpaired electron. The geometrical parameters of X˜ 2B2 CH2X, the trans-HCXH and cis-HCXH conformers, and the first three excited states are reported at the coupled-cluster level with single, double, and perturbatively applied triple excitations [CCSD(T)] using Dunning's correlation consistent polarized valence quadruple-ζ basis set (cc-pVQZ). The C2v structures on the ground state surface are predicted to lie 9.3 and 13.5 kcal/mol lower than the trans- and cis-isomers, respectively, for CH2N and 28.1 and 30.0 kcal/mol, respectively, for CH2P. There are many other important properties of CH2N and CH2P which are not known from experiment. The geometrical parameters of the CH2N ground state [r(C–N)=1.2462 Å, r(C–H)=1.0921 Å, and θ(HCH)=119.4°] and the CH2P ground state [r(C–P)=1.6583 Å, r(C–H)=1.0842 Å, and θ(HCH)=118.9°] agree well with the C–N and C–P bond distances of the r0 structures derived from microwave data, although notable differences were observed in the C–H bond distance and HCH bond angle. This research resolves an earlier discrepancy between theory and experiment for the ground state C–P distance in CH2P. The Fermi contact terms for 1H, 13C, 14N, and 31P were determined at CCSD(T) level of theory with the cc-pVTZ and cc-pVQZ basis sets and are in reasonable agreement with the experimental values with a maximum deviation of 26 MHz for CH2N and 11 MHz for CH2P. The excited states of CH2N are predicted to lie 33 000 cm−1 (A˜ 2B1), 36 000 cm−1 (B˜ 2A′), and 38 000 cm−1 (C˜ 2A1) above the ground state, and the excited states of CH2P to lie approximately 21 000 cm−1 (A˜ 2A′), 26 000 cm−1 (B˜ 2B1), and 33 000 cm−1 (C˜ 2A1) above the ground state. © 2001 American Institute of Physics.

    Notes: We have investigated in C6 glioma cells the involvement of protein kinase C (PKC) in the regulation of serotonin-2A receptor (5-HT2A receptor) expression by agonist treatment. Comparison of the time-courses of agonist-induced downregulation of receptor number and mRNA indicate that a decrease in the number of 5-HT2A receptor binding sites in response to serotonin (5-HT) treatment is preceded by a decrease in 5-HT2A receptor mRNA. This decrease in 5-HT2A receptor mRNA as a result of agonist exposure was not due to a change in the stability or half-life of the transcript. Pretreatment of cells with the PKC inhibitor bisindolylmaleimide blocked the decrease in 5-HT2A receptor mRNA levels, and attenuated the down-regulation of 5-HT2A receptor binding sites induced by treatment with 5-HT. Experiments performed with the PKC inhibitors calphostin C and Gö 6976 confirmed that PKC was involved in the regulation of 5-HT2A receptor mRNA by agonist and implicate the conventional subgroup of PKC isoforms. Western blot analysis, using isoform-specific anti-PKC antibodies showed that under our culture conditions C6 glioma cells express the conventional isoforms PKC α, PKC γ, as well as the novel isoforms PKC δ, PKC ε, and the atypical isoforms PKC λ and PKC ι. Upon treatment with 5-HT for 10 min levels of the conventional isoforms PKC α and PKC γ increased in the nuclear fraction. Taken together, our results implicate PKC α and/or PKC γ in the regulation of 5-HT2A mRNA receptor and binding sites in response to agonist treatment.

    Notes: Al2O3-ZrO2 eutectics containing 0 to 12.2 mol% Y2O3 (with respect to zirconia) were produced by directional solidification using the laser floating zone (LFZ) method. Processing variables were chosen to obtain homogeneous, colony-free, interpenetrating microstructure for all of the compositional range, optimum from the viewpoint of mechanical properties. The amount of cubic, tetragonal, or monoclinic zirconia phases was determined using a combination of Raman and X-ray diffraction techniques. Monoclinic zirconia was present up to concentrations of 3 mol% Y2O3, while the amount of tetragonal zirconia gradually increased with yttria content up to 3 mol%. Cubic zirconia was the only phase detected when the yttria content reached 12 mol%. The residual stresses in alumina were measured using the shift of the ruby R lines. Compressive stresses were isotropic when measured in the samples containing tetragonal and cubic zirconia, while higher tensile, anisotropic stresses were found when monoclinic zirconia was present. They were partially relieved in the eutectic sample without yttria. These results were compared with a thermoelastic analysis based on the self-consistent model.

    Notes: : Nitrate levels in the Ocklawaha River Basin in north central Florida were reviewed over a 50-year period. Data were obtained from the literature, U.S. Environmental Protection Agency (USEPA) STOrage and RETrieval (STORET), and U.S. Geological Survey (USGS) databases. The study objective was to determine whether nitrate concentrations are increasing and if so, whether this increase is linked to land use changes. Increasing nitrate levels were seen at 5 of the 14 stations, while other stations showed no trend or a decreasing trend. Median nitrate concentrations in the Ocklawaha River increased from 0.07 mg-N/L to 0.78 mg-N/L at sites downstream from the Silver River. Throughout the Rodman Reservoir, median nitrate concentrations decreased from 0.48 mg-N/L to 0.01 mg-N/L and increased to 0.04 mg-N/L after the Kirkpatrick Dam. Flow and concentration relationships were correlated for five stations. At four of the five stations nitrate concentrations decreased in response to increasing flow, likely the result of dilution with nitrate poor water. Changes in land use over a 20- year period (1970 to 1990) also were monitored. Sources of nitrate have been linked by isotopic analysis to organic and inorganic fertilizers, which appear to be related to increased urbanization and an increase in lawns that require nutrient fertilization.

    Notes: Previous discussions of the evolution of the self have diverged greatly in their estimates of the date at which the capacity for self-thought emerged, the factors that led self-reflection to evolve, and the nature of the evidence offered to support these disparate conclusions. Beginning with the assumption that human self-awareness involves a set of distinct cognitive abilities that evolved at different times to solve different adaptive problems, we trace the evolution of self-awareness from the common ancestor of humans and apes to the beginnings of culture, drawing upon paleontological, anthropological, biological, and psychological evidence. These data converge to suggest that that modern self-thought appeared just prior to the Middle-Upper Paleolithic transition, approximately 60,000 years ago.Recto running head: Evolution of the Self.

    Notes: We have recently shown that phototactic movement in the unicellular cyanobacterium Synechocystis sp. PCC6803 requires type IV pilins. To elucidate further type IV pilus-dependent motility, we inactivated key genes implicated in pilus biogenesis and function. Wild-type Synechocystis sp. PCC6803 cells have two morphologically distinct pilus types (thick and thin pili). Of these, the thick pilus morphotype, absent in a mutant disrupted for the pilin-encoding pilA1 gene, appears to be required for motility. The thin pilus morphotype does not appear to be altered in the pilA1 mutant, raising the possibility that thin pili have a function distinct from that of motility. Mutants disrupted for pilA2, which encodes a second pilin-like protein, are still motile and exhibit no difference in morphology or density of cell-surface pili. In contrast, inactivation of pilD (encoding the leader peptidase) or pilC (encoding a protein required for pilus assembly) abolishes cell motility, and both pilus morphotypes are absent. Thus, the PilA1 polypeptide is required for the biogenesis of the thick pilus morphotype which, in turn, is necessary for motility (hence we refer to them as type IV pili). Furthermore, PilA2 is critical neither for motility nor for pilus biogenesis. Two genes encoding proteins with similarity to PilT, which is considered to be a component of the motor essential for type IV pilus-dependent movement, were also inactivated. A pilT1 mutant is (i) non-motile, (ii) hyperpiliated and (iii) accumulates higher than normal levels of the pilA1 transcript. In contrast, pilT2 mutants are motile, but are negatively phototactic under conditions in which wild-type cells are positively phototactic.
