    Notizen: Abstract— The distribution of some monoamines in the tissues of an echinoderm, the sunflower starfish, Pycnopodia helianthoides, has been investigated in order to ascertain whether monoamine levels are similar to those found in other Deuterostomia. Dopamine, noradrenaline and octopamine were present in the arm nerves at concentrations of 5954 ng/g, 2133 ng/g and 260 ng/g respectively. The octopamine/ noradrenaline ratio for the arm nerve was 0.12 and thus similar to the typical mammalian (deuteros-tome) ratio rather than the higher invertebrate (protostome) ratio. Trace amounts of p-tyramine, β-phenylethylamine were also present. 5-Hydroxytryptamine was not detected but tryptamine was present in high concentrations (1251 ng/g).

    Notizen: Abstract— An enzyme radiochemical assay for p-octopamine, m-octopamine (norphenylephrine) and phenylethanolamine based on the N-methylation of these amines by the enzyme phenylethanolamine N-methyl transferase (S-adenosyl-l-methionine: phenylethanolamine N-methyl transferase (EC 2.1.1.28) has been developed. [3H]Methyl-S-adenosyl-l-methionine was used as methyl donor. The reaction products are converted to their dansyl derivatives and separated by TLC in three different solvent systems prior to liquid scintillation counting. The method exhibits a sensitivity of less than 10 pg for each amine and is suitable for the measurement of endogenous p-octopamine levels in mammalian brain. The highest levels of p-octopamine were found in the hypothalamus (3.4 ng/g) but despite the sensitivity of the assay, neither phenylethanolamine nor m-octopamine could be detected. After MAO inhibition, however, both of these amines were found to be present. p-Octopamine was increased substantially in all brain regions following the administration of an MAO inhibitor, whereas pretreatment with reserpine produced a significant decrease in the hypothalamus.

Octopamine and dopamine are present in high concentration but the noradrenaline content is only 1/25 that of octopamine.     Notizen: Summary Cocaine, a catecholamine agonist, has been shown to produce a transient induction of the immediate-early gene c-fos and its protein product Fos in the striatum of normal rats. In the present study we report that the expression of Fos can be induced by cocaine challenge in intrastriatal grafts derived from cell suspensions of embryonic striatal primordia. Fos-like immunoreactivity in the nuclei of grafted neurons was detected 2 hr after the injection of 50 mg/kg cocaine into the host rats. Neurons with Fos-immunoreactive nuclei tended to form clusters in the striatal grafts. The Fos-rich clusters were aligned with acetylcholinesterase (AChE)-rich and tyrosine hydroxylase (TH)-rich patches demonstrated in adjoining sections. Previous studies have shown that presynaptic and postsynaptic cellular markers of the dopaminergic system in the striatum, including immunostaining for TH and dopamine- and adenosine 3′:5′-monophosphate-regulated phosphoprotein (DARPP-32), and binding for high affinity dopamine uptake sites and for dopamine D1 and D2 receptor sites, are all concentrated in the AChE-rich patch regions (P regions) of such embryonic striatal grafts. The preferential expression of Fos in neurons of the P regions of the grafts thus implies that the induction of Fos was cell-type specific in being concentrated in the parts of the grafts that express striatal phenotype and that are innervated by catecholamine-containing fibers. This specificity strongly suggests that the activation of Fos expression in neurons of the P regions of the grafts reflects dopaminergic interactions between the grafts and host nigrostriatal fibers. We conclude that the cellular messenger systems and transcriptional activation mechanisms responsive to dopaminergic stimulation by the host can be activated in the embryonic striatal grafts, and that the grafts are thus functionally integrated into the host brain at the level of cellular signaling systems.

    Notizen: Summary The salivary glands of the moth,Manduca sexta, are described, emphasizing correlations between structure and function in an attempt to explain the production of a dilute saliva. Each of the paired glands consists of five distinct regions: protein secreting, fluid secreting, thin duct, bulbous duct, and common duct. Each region constists of a single, ultrastructurally distinct, cell type. It is proposed that the protein and fluid secreting regions produce an enzyme-containing primary saliva isosmotic with the haemolymph; this saliva is modified in the remaining regions of the gland to yield a dilute saliva.
