Notizen: To determine: 1) the minimum amounts of sleep subjects would tolerate, 2) the changes in EEG sleep measures, and 3) whether subjects would revert to baseline sleep after study termination, 4 couples gradually reduced their sleep. Three couples reduced their TST in 30-min steps from a baseline of 8 hrs and one couple from a baseline of 6.5 hrs. Subjective estimates of sleep time, sleep quality, and mood were collected daily. Home EEG sleep recordings were obtained 3 nights a week. Two of the 8-hr sleepers reduced their sleep to 5.5 hrs, 2 to 5.0 hrs, and 2 reached 4.5 hrs. These 6 subjects continued sleeping 1 to 2.5 hrs below baseline amounts a year after reduction terminated. The 6.5-hr baseline couple reached 5.0 hrs and returned to 6.5 hrs TST during follow-up.Stages W, 2, and REM decreased significantly in absolute amounts. Percentage of stages W and 2 also decreased significantly. REM percent remained constant. Stage 3 was constant while stage 4 increased in both absolute and relative amounts. REM cycle length remained constant. Stage 4 rebound on 7-hr nights was not observed during times of greatest sleep reduction. Occurrences of stage REM within 10 min of stage 1 onset were observed in 2 subjects when their TST was below 6.5 hrs.Our results are consistent with other studies of shortened sleep, indicating that TST is the major determinant of sleep-stage characteristics.

Notizen: Long-term gradual sleep reduction effects were investigated on 4 young adult collegiate couples. The battery of assessment tools included a sleep log, Stanford Sleepiness Scale, Profile of Mood States, Feeling Tone Checklist, a measure of circadian oral temperature, Williams Word Memory test, Digit Span test, Wilkinson Auditory Vigilance task, Wilkinson Addition task, Minnesota Multiphasic Personality Inventory, Rapid Alternation task, psychiatric and medical examinations, and a subjective effects questionnnaire. It was concluded that 6–8 months of gradual sleep restriction, down to 4.5–5.5 hrs per night, does not result in behavioral effects measurable by the instruments used. Subjective fatigue appears to be the limiting factor in determining tolerability of gradual sleep restriction. At the end of an additional 12-month follow-up period, total sleep time was still 1–2.5 hrs below baseline, but measures of well-being had returned to baseline levels.

Notizen: The effects of exercise and sleep loss on recovery sleep were studied in young male naval volunteers. For 1 hr out of every 4 hrs during a 40-hr period, 20 subjects rested in bed and 10 subjects bicycled. Eleven measures of recovery night sleep were selected for comparison of the bedrest and exercise groups. Only one reached significance under the conservative Dunn-Bonferroni criterion: the exercise group had a higher percent total sleep time. The results indicate that exercise does increase the effects of sleep loss on recovery sleep, but that there is no simple, direct effect on specific sleep stages.

Notizen: Abstract The significance of partial deficiency of erythrocyte adenine phosphoribosyltransferase (APRT), reported in a number of subjects with gout, has been investigated by studying its incidence in 700 normal blood donors. Three clearly deficient subjects were found, an incidence not significantly different from that in patients with abnormalities of urate metabolism. A new assay method for APRT is described in which an erythrocyte lysate is incubated with adenine and phosphoribosylpyrophosphate (PRPP) for a given time; both hemoglobin and adenine nucleotide (AMP) are then precipitated with lanthanum phosphate; the change in absorbance of adenine at 260 nm reflects the extent of its conversion to AMP by APRT.

Notizen: Abstract In a dose-response study, a formulation of chenodeoxycholic acid (chenic acid) with complete bioavailability was fed at doses of 0,250, 500, 750 mg for randomized 6-week periods to 13 patients with radiolucent gallstones. The proportion of chenic acid in biliary bile acids increased in direct relation to dose. Lithocholic acid also increased, but ursodeoxycholic acid showed no significant change. Bile saturation decreased with increasing dose, but response was variable and achieved significance only at 750 mg/day. Cholesterol saturation was negatively correlated with the proportion of chenic acid in biliary bile acids: When biliary bile acids contained greater than 70% chenic acid (and ursodeoxycholic acid), bile became unsaturated, on the average. The data suggest that a dose of at least 10–15 mg/kg may be necessary to obtain desaturation in many gallstone patients.

Notizen: Improved techniques in preparing and sizing dental alloy powders containing Ag, Sn, and Cu have led to amalgams with higher strengths and less spread in dimensional change but very poor shelf life. Since maximum strengths occurred for amalgams containing about 15% more Sn than previously reported, and consequently presumably more of the weak γ2 phase, the higher strengths are difficult to understand. The poor shelf life may be explained by oxidation of the highly reactive splatcooled alloy or possibly by the existence of highly reactive metastable phases which transform fairly rapidly to the less reactive equilibrium ones.
