    Notes: The complete nucleotide sequence of the xynA gene coding for a xylanase (XYLA) expressed by Pseudomonas fluorescens subspecies cellulosa, has been determined. The structural gene consists of an open reading frame of 1833 bp followed by a TAA stop codon. Confirmation of the nucleotide sequence was obtained by comparing the predicted amino acid sequence with that derived by N-terminal analysis of purified forms of the xylanase. The signal peptide present at the N terminus of mature XYLA closely resembles signal peptides of other secreted proteins. Truncated forms of the xylanase gene, in which the sequence encoding the N-terminal signal peptide had been deleted, still expressed an enzyme which was secreted in Escherichia coli. XYLA contains domains which are homologous to an endoglucanase expressed by the same organism. These structures include serine-rich sequences. Bal31 deletions of xynA revealed the extent to which these conserved sequences, in XYLA, were essential for xylanase activity. Downstream of the TAA stop codon is a G + C-rich region of dyad symmetry (δG = 24kcal) characteristic of E. coli Rho-independent transcription terminators.

    Notes: The results of the Midland Valley Investigation by Seismology (MAVIS) are described. The experiment consists of three refraction profiles of between 80 and 100 km length across the Upper Palaeozoic basins of the Scottish Midland Valley.Three refractors are recognized, thus defining four upper crustal layers. The top two layers are interpreted as Carboniferous plus Upper Old Red Sandstone, and Lower Old Red Sandstone plus Lower Palaeozoic. The structure of the refractor, an unconformity, separating the two layers mirrors surface structure. Deeper refractors, associated with Midland Valley crystalline basement, show no correlation with surface structure. The MAVIS survey allows the refinement of earlier gross interpretation of upper crustal structure in the Midland Valley, but confirms the existence of anomalously shallow (6.4 km s–1) basement, and shows, in conjunction with results of previous work, that a large area of the central and southern Midland Valley is characterized by one velocity signature, probably indicative of a single terrane.

    Notes: Nodulated winged bean [Psophocarpus tetragonolobus (L.) DC., cv. UPS 122] were grown under constant environmental conditions and supplied with mineral nutrient solution in which nitrogen was absent or was present as nitrate (12 mg N week-1 plant-1).Nitrate treatment dramatically promoted plant growth, increased fruit weight 1.6 fold, was necessary for tuberisation and enhanced nodulation. The in vitro accumulation of 14C into asparagine and aspartate components of excised nodules supplied with exogenous 14CO2 and [14C]-D-glucose was greater for nitrate-treated plants, whilst accumulation into ureides was reduced by nitrate treatment. Levels of amino acids in xylem sap were greater for plants supplied with a complete nutrient solution, than those grown without applied nitrate, particularly for asparagine, glutamine and proline. Xylem ureide levels were greater for plants grown in the absence of supplementary nitrate.Nitrogen accumulated in leaf, stem and petiole, and root nodule tissues for utilisation during fruit development; peak nitrogen levels and time of anthesis were retarded for plants grown without applied nitrate. The shoot ureide content increased during fruiting, coincident with decreases in the total nitrogen content, indicating that ureide pools are not utilised during the early reproductive phase. However ureide reserves, particularly allantoin, were utilised during the later stages of pod fill.Enzyme activity which metabolised asparagine was found throughout the plant and was identified as K+-dependent asparaginase (EC 3.5.1.1) and an aminotransferase. Apart from temporal differences in developmental profiles of enzyme activity, the activity of these enzymes and of allantoinase (EC 3.5.2.5) in developing tissues were similar for both treatments. The main differences were greater asparaginase and asparagine:pyruvate aminotransferase activities in root tissues and fruit of nitrate-supplied plants; allantoinase activity in the primary roots of plants grown without nitrate decreased during development, whilst activity in developing tubers (nitrate-supplied plants) increased.

    Notes: Abstract Cells of the diatomCyclotella meneghiniana were exposed in a closed system to 0.095 ppm pentachlorobenzene over a period of 5 days. Changes in fatty acid and morphological percent composition were monitored to determine the effect of the toxicant. The greatest morphological change observed was an increase in lipid volume. Most morphological changes occurred in the 1 hour and 5 day sampling periods. Few changes in morphological characteristics or fatty acid percent composition were observed at eight hours, when the cells were in the dark. The C18∶1 and C20∶5 fatty acids were most variable with exposure to pentachlorobenzene. Results suggest that at sublethal doses, lipophilic toxicants exert effects that are biphasic. That is, immediately measurable effects are observed in the cells that include increases in storage products and changes in membranous organelles. Long-term effects are postulated to be the result of mobilization of lipophilic toxicants that have partitioned into lipid stores and are more available when lipids are metabolized. Although pentachlorobenzene has a higher octanol/water partition coefficient, it appears to exert fewer cellular changes than any trichlorobenzene isomer.

    Notes: Abstract Cells of the diatomCyclotella meneghiniana were exposed in a closed system to 0.245 ppm 1,3,5-trichlorobenzene. Response of the diatom was measured by quantitative ultrastructure and fatty acid percent composition over a 5-day period. During that time, 28 significant morphological and 13 significant fatty acid percent composition changes occurred. Autophagic-like vacuoles were observed consisten9ly through the sampling periods. In comparison with exposure to 1,2,4-trichlorobenzene, 1,3,5-trichlorobenzene exerts fewer effects in the parameters studied, and these effects were observed most frequently in membranous components during the initial eight hours of exposure. It is suggested that the amount of cellular lipid and the relative reactivity of the isomer are responsible for the observed effects.
