    Notes: To clarify the significance of basic fetoprotein (BFP) in lymphocytes, we investigated whether BFP is produced in lymphocytes during blastic transformation. Peripheral blood lymphocytes obtained from 14 adults were cultured under the stimulation of lectins. The concentration of BFP in the culture medium (extracellular BFP) was estimated serially. The incorporation of [6-3 H] thymidine was assayed simultaneously. The intracellular BFP was measured by dual flow cytometry for DNA and BFP. A lymph node was studies immunohistochemically. Serum BFP was measured in four cases of lumphocytic leukaemia. In two cases, dual staining was performed. The intracellular BFP of the mitogen-stimulated lymphocytes was increased within 24 h. The extracellular BFP was increased exponentially from 72 h. The extracellular BFP at 96 h did not correlate with the [3H]-thymidine incorporation. The intracellular BFP increase began in G1 phase. Immunostaining showed that the B cells also produced BFP. Theserum BFP level in leukaemia was high in 1 of 4 cases and the leukaemic cells in two cases showed high intracellular BFP content. These observations indicate that BFP is produced in activated human lymphocytes and in lymphocytic leukaemic cells. The production of BFP during blastic transformation will be a useful new in vitro model for studying the biological role of BFP, and BFP labelling may offer some new possibilities for study of lymphocytes.

    Notes: Summary Using the polymerase chain reaction (PCR), proviral DNA sequences of thepol andenv regions of human T-cell leukemia virus type I (HTLV-I) were directly amplified in paraffin-embedded and frozen tissue sections of active inflammatory central nervous system (CNS) lesions in three autopsy cases of HTLV-I-associated myelopathy (HAM) with serological confirmation. In parallel, the enumeration of UCHL-1 (monoclonal antibody reactive to T-cells) positive cells in the tissue sections subjected to PCR were carried out. Although the control DNA sequence of parathyroid hormone-like peptide gene was definitely amplified, no signals for HTLV-I proviral sequences were detected in these specimens. The number of UCHL-1 positive cell nuclei was almost on the border line of our PCR sensitivity in formalin-fixed tissue, which was estimated to be 20–200 copies. Therefore, it is unlikely that the central nervous system tissue damage in HAM/TSP is a consequence of productive infection of HTLV-I in the CNS tissue.

    Description / Table of Contents: Zusammenfassung Mit Hilfe der Gaschromatographie wurde die Kohlenmonoxid-Konzentration im Blut des rechten und linken Herzens vergleichend untersucht. Das Material stammt von Leichen mit antemortaler oder nicht-antemortaler Feuer- oder CO-Exposition. Das Konzentrationsverhältnis von COHb zwischen linkem und rechtem Herzblut (L/R-Quotient) scheint ein nicht brauchbares Kriterium zu sein, um festzustellen, ob der Tod sich vor oder nach der Feuer-Exposition ereignet hat; mit der Ausnahme von solchen Fällen, wo bei makrokopischer Untersuchung kein Ruß in den Atemwegen gefunden wird und wo die COHb-Konzentration innerhalb jener Grenzen ist, welche für Zigaretten-Raucher als normal angesehen werden.
    Notes: Summary Carbon monoxide hemoglobin (COHb) concentrations in left and right heart blood samples from cadavers both exposed and not exposed to fire or CO gas were analyzed by the gas chromatographic method. The COHb concentration ratio between samples of left and right heart blood (L/R ratio) does not appear to be useful for establishing whether death has occurred before or after exposure to fire with the exception of cases where no soot can be detected in the airways by the naked eye and the COHb concentration in the blood sample is within the level considered normal for tobacco smokers.

    Notes: Abstract Micron-size monodisperse crosslinked polymer microspheres having chloromethyl groups were prepared by seeded copolymerization of styrene, divinylbenzene, and chloromethylstyrene in the presence of 2.1-μ monodisperse polystyrene seed particles produced by dispersion polymerization. The modification reaction of chloromethyl groups on the surfaces with polyamines such as triethylenetetramine and ethylenediamine was carried out. From the measurements of ξ potential and the amount of chloride ion released, the introduction of a large number of cationic groups at the surfaces was confirmed.
