    Notes: Abstract. To identify individual chromosomes of a frog karyotype by their fluorescence banding patterns, chromosomes were stained with actinomycin D and 4,6-diamidino-2-phenylindole (DAPI) after incorporation of BrdU during the late S-phase. The chromosomes of three Rana species which were selected for this study (R. ridibunda, R. lessonae and R. japonica) showed well-defined late replication bands. The fluorescence patterns obtained were the reverse of those produced by a 4Na-EDTA Giemsa-staining technique. Fluorescence patterns of the two water frog species (R. ridibunda and R. lessonae) were similar to each other, except for the different fluorescence of the centromeric heterochromatin, which gave extremely bright signals in R. ridibunda but no signal in R. lessonae. Experiments also showed differences between the fluorescence patterns of R. lessonae chromosome 13 in the Italian and Luxembourgian populations. These results sho w that the fluorescence replication banding using actinomycin D and DAPI is very effective in identifying individual frog chromosomes and detecting their structural changes.

    Notes: Abstract SnO2—glass composites are promising materials for nitrogen-fireable thick film resistor systems. However, the SnO2—glass composite has many undesirable properties which should be improved for industrial application, such as a high electrical resistivity and a large negative temperature coefficient of electrical resistance. This work was undertaken to make a survey of the methods for improvement in the electrical properties of the SnO2—glass composites. The effect of the addition of Cu particles, with a large positive temperature coefficient of electrical resistance, on the electrical properties of SnO2—glass composites has been investigated. Cu particles have been coated with semiconductive SnO2 fine particles by hydrolysing tin and antimony ethoxides then firing. The coated particles have been applied as conductive components in the glass composite. Using SnO2-coated Cu particles, Cu and SnO2 particles are homogeneously dispersed in a glass matrix, and the electrical properties of the glass composites largely depend on the volume fraction of Cu in the glass composite. The volume fraction of Cu in the glass composite is determined by a Cu/SnO2 volume ratio in the coated particles. From the experimental evidence, it is thought feasible to produce the glass composite having well-controlled electrical properties by the suitable selection of the Cu/SnO2 ratio in the coated particles.

    Notes: Abstract To determine whether heat shock proteins can be utilized as a biomarker for cytotoxicity of dental materials the induction of synthesis of heat shock proteins by mercuric chloride was examined. To analyse the synthesis of heat shock proteins, HeLa cells were labelled with [35S] methionine, and the labelled proteins were separated by SDS-PAGE and autoradiographed. Incubation of the cells in a medium containing 1.25 to 40 μM mercuric chloride markedly increased the synthesis of HSP70. At 20 or 40 μM mercuric chloride in medium, HeLa cells synthesized HSP70 at 2 h after exposure, maximally at 4–7 h, and gradually diminished thereafter. Examination of the cytotoxicity of mercuric chloride by the conventional neutral red uptake assay revealed a reduction of uptake of the dye in the presence of mercuric chloride at 40 μM and above. These findings suggest that the induction of synthesis of HSP70 is one of the most sensitive cellular responses caused by mercury ion, and the heat shock protein assay can be utilized for evaluation of the cytotoxicity of dental materials.

    Notes: Abstract The stability constans,β 1, of each monochloride complex of Eu(III) have been determined in the methanol and water mixed system with 1.0 mol·dm−3 ionic strength using a solvent extraction technique. The values ofβ 1 increase with an increase in the mole fraction of methanol (X S ) in the mixed solvent system when 0≤X S ≤0.40. The, distance of Eu3+−Cl− in the mixed solvent system was calculated using the Born-type equation and the Gibbs' free energy derived fromβ 1. Calculation of the Eu3+−Cl− distance and the preferential solvation, of Eu3+ by water proposed the variation of the outersphere complex of EuCl2+ as follows: (1) [Eu(H2O)9]3+Cl−, [Eu(H2O)8]3+Cl− and [Eu(H2O)7(CH3OH)3+Cl− inX S≤0.014, (2) [Eu(H2O)8]3−Cl− and [Eu(H2O)7(CH3OH)]3+Cl− in 0.014〈X S 〈0.25 and (3) [Eu(H2O)7(CH3OH)]3−Cl− and [Eu(H2O)6(CH3OH)[2 3+Cl− in 0.25〈X S ≤0.40.

    Notes: Abstract The stability constants, α1, of each monochloride complex of Nd(III) and Tm(III) have been determined in the mixed system of methanol and water with 1.0 mol·dm−1 ionic strength using a solvent extraction technique. The values of α1 of Nd(III) and Tm(III) increase as the mole fraction of methanol in the mixed solvent system (X s) increases. However, the variation mode of α1 againstX s in the region of 0.00≤X s≤0.40 differs from each other, a concave curve for the Nd(III) and a convex curve for the Tm(III). The LnCl2+ formed is present as a solvent-shared ion-pair. Since Cl− is a structure breaking ion, it was assumed that the primary solvation sphere of Ln3+ directly contacted with Cl−. Calculation of Ln3+−Cl− distance using Bom-type equation revealed the followings: (1) for Tm3+ with coordination number 8, the estimated distance between Tm3+ and Cl− increases linearly withX s in 0.00≤X s≤0.40. The results mean an increase of the primary solvation sphere size of Tm3+ withX s. (2) For Nd3+, the distance between Nd3+ and Cl− decreases linearly withX s in 0.00≤X s〈0.13, where both coordination numbers of 9 and 8 coexist, while it increases withX s in 0.13〈X s≤0.40. The results mean a decrease of the primary solvation-sphere size of Nd3+ withX s in 0.00≤X s〈0.13 and an increase of that withX s in 0.13〈X s≤0.40.

    Notes: Abstract The initial appearance and the development of Leydig cells (LCs), the sites of steroid hormone production in the testis, were investigated ultrastructurally during testicular differentiation in the Japanese eel, Anguilla japonica. In addition, the effects of a single injection of human chorionic gonadotropin (HCG; 5 IU g body weight-1) on histological changes of the testes and serum 11-ketotestosterone (11-KT) were examined at various stages (15–18, 20–23, 26–29, 32–35, 38–41 and 46–50 cm body length (BL)) of testicular differentiation. Testicular differentiation was morphologically characterized by the development of loose connective tissue on the medial side in animals 18–29 cm in BL. Ultrastructurally, LCs were first identified in the loose connective tissue of the testis of the 23 cm fish. In the testes of fish over 32 cm, clusters of LCs were distributed throughout the interstitial region accompanying the increase in number of spermatogonia. In fish larger than 32 cm, spermatogenesis was induced by administration of HCG; serum 11-KT levels were also raised. On the other hand, there was no effect on spermatogenesis or serum 11-KT levels in fish less than 29 cm, or in the controls. These result suggests that morphological differentiation of LCs occurs in testis of the 23 cm eel, and subsequently, the testes of eels of BL more than 32 cm acquire the capability to produce steroid hormones.

    Notes: Abstract The in vitro identification of copigmentation was carried out using anthocyanins, such as malvidin 3RGac5G, petunidin 3RGac5G and delphinidin 3RGac5G and the flavone isovitexin. These are major pigments of Iris ensata. These anthocyanins brought about copigmentation, i.e., the bathochromic shift (bluing effect) of visible λ max due to increased concentrations of isovitexin, and 32 to 35 nm were estimated as the magnitude (Δλ max) of each shift. In addition, the absorption spectrum of 0.1 mM malvidin 3RGac5G, 0.07 mM petunidin 3RGac5G and 0.7 mM isovitexin mixture closely matched those of the fresh outer perianths of the bluish purple cultivars, ‘Suiten-isshoku’, ‘Hekikai’ and ‘Yakonotama’ which belong to the malvidin 3RGac5G - petunidin 3RGac5G type of I. ensata. Therefore, these results indicated that the bluing effect on the flower color of the bluish purple cultivars of this species was caused at least in part by the copigmentation between these anthocyanins and the flavone isovitexin. For the copigment effects of isovitexin among malvidin 3RGac5G, petunidin 3RGac5G and delphinidin 3RGac5G, λ max and Δλ max of delphinidin 3RGac5G was slightly higher than those of malvidin 3RGac5G or petunidin 3RGac5G, and the copigmentation of delphinidin 3RGac5G was characterized by a higher concentration of isovitexin. Finally, the breeding for blue flowers due to copigmentation of delphinidin 3RGac5G with isovitexin in I. ensata was discussed.

    Notes: Abstract Ischemia-reperfusion injury by free radicals and lipid peroxides is observed in various organs. Ascorbic acid (AsA) or glutathione (GSH) in various doses (AsA:2, 0.5, 0.1 mmol/kg, GSH:2 mmol/kg) was intraperitoneally administered to male Wistar rats. The entire small intestines were resected just before ischemia, after ischemia, and after 20 min of reperfusion (n = 7–10 at each time point). At each time point, the specimens were subjected to assays of lipid peroxides, GSH, and glutaminase activity of the tissues; they were also examined histologically. In the AsA group, the production of lipid peroxides after reperfusion was significantly suppressed in a dose-dependent manner, and the ratio of oxidized GSH to total GSH was also significantly low. Tissue glutaminase activity decreased to a lesser extent, and the degree of injury was apparently less marked in the AsA group. This study indicates that AsA acts as an antioxidant against peroxidative tissue injury, possibly by scavenging radicals, preserving reduced GSH, and reducing the peroxidative reaction.
