    Notizen: Comparative analysis was carried out for hydrogen peroxide (H2O2) accumulation in response to powdery mildew (Blumeria graminis[Erysiphe graminis] f.sp. tritici) primary germ tube (PGT) and appressorial germ tube (AGT) contact, and in papilla (cell wall apposition) and hypersensitive responses (HR) in wheat (Triticum aestivum). Using primary leaves of three susceptible wheat lines (Bainong 3217; Beijing 837; Jingshuang 16) and five resistant lines [Mardler (Pm2 + Pm6); Ulka/8 × Cc (Pm2); Mardler/7 × Bainong 3217 (Pm2); Pm16; Pm16/7 × Beijing 837 (Pm16)], early H2O2 accumulation between 10 and 48 h after inoculation was studied. Strong H2O2 accumulation was found in effective papillae and associated cytosolic vesicles in both susceptible and resistant wheat lines, suggesting the important role of H2O2 in effective papillae formed as a general plant defence against powdery mildew. High frequency of effective papilla formation was observed in all five resistant lines. Among resistant lines, hypersensitive cell death was detected earlier in Mardler and Ulka/8 × Cc than in Pm16 and Pm16/7 × Beijing 837. In all cases this was associated with H2O2 accumulation in attacked epidermal cells. Interestingly, penetration resistance but not HR appeared to be mediated by the Pm2 gene in the Mardler/7 × Bainong 3217 line, suggesting that Pm2 may govern an HR-independent defence pathway in this genetic background. As effective papillae and HR did not occur in the same cells, papilla deposition may be independent of the HR response, despite the fact that both defence mechanisms were associated with high H2O2 accumulation. In wheat powdery mildew-incompatible interactions, HR acts as a second line of defence to contain infection when the papilla defence fails.

    Notizen: Summary.  Factor X (FX) deficiency is a rare bleeding disorder inherited as an autosomal recessive trait. In this study, we investigated the molecular basis of FX deficiency in a Chinese pedigree. The proposita showed a markedly prolonged activated partial thromboplastin time and a mild prolongation of prothrombin time. The levels of FX antigen and FX activity were 58.6% and 2.5%, respectively. Molecular analysis revealed that the proposita was compound heterozygous for two novel mutations: IVS1 + 1G 〉 A and G1185A (Arg347His). The aberrant transcripts from the IVS1 + 1G 〉 A mutant allele were not detected by analyzing the splicing pattern of ectopic transcripts in leukocytes of the patient with nested polymerase chain reaction after reverse transcription. We thus hypothesize that the mRNA molecules originating from the IVS1 + 1G 〉 A mutation were rapidly destroyed in vivo. Site-directed mutagenesis of FX cDNA was used to introduce FXG1185A mutation, and wild-type as well as mutant FX proteins were expressed by transient transfection in HEK 293 cells. Normal FX antigen levels both in the conditioned media of cells expressing the mutant and in cell lysates were detected by an enzyme-linked immunoadsorbent assay. Evaluation of wild-type and mutant coagulant activity demonstrated that the FX molecules carrying the Arg347His mutation have dramatically decreased activity.

    Notizen: M3:2 high speed steels with and without carbon addition were prepared by using powdermetallurgy at sintering temperature between 1210 and 1280 °C. Densification, microstructure andmechanical properties of M3:2 high speed steels were investigated. Experimental results show thatwith 0.4wt% carbon addition, full density high speed steels were obtained at temperatures in therange 1240-1260 °C which is 40 °C lower than that of the undoped counterparts, leading to a sinteringwindow expanded by 10 to 20 °C. By the addition of 0.4wt% carbon, the sintered steels showattractive combinations of bend strength and hardness over those of M3:2 steels without carbonaddition. The results reveal that the addition of carbon will not only lower the sintering temperatureand oxygen content, but also improve the mechanical properties of the sintered steels

    Notizen: Conditions are reported for the reverse-phase HPLC analysis of gossypol and of some of its derivatives, including anhydrogossypol, gossypolone, gossypol hexaacetate, and Schiff's base derivatives. It is shown that field desorption (FD) and chemical ionization (CI) mass spectrometry have advantages over electron impact (EI) mass spectrometry for the characterization of these compounds and that combined high-performance liquid chromatography/mass spectrometry (LC/MS), especially when used in the CI mode, is particularly effective.
