    Notizen: Background : The acid suppressive effect of lansoprazole is influenced by the P450 2C19 (CYP2C19) polymorphism.Aim : To investigate whether the CYP2C19 genotype is related to the healing of erosive reflux oesophagitis during treatment with lansoprazole.Methods : Eighty-eight Japanese patients with erosive reflux oesophagitis were treated with a daily oral dose of 30 mg lansoprazole for 8 weeks. The CYP2C19 genotype, Helicobacter pylori infection status and serum pepsinogen I/II ratio were assessed before treatment. At 4 and 8 weeks, the healing of erosive reflux oesophagitis was evaluated endoscopically.Results : The healing rates were 57.1%, 69.2% and 72.7% at 4 weeks and 77.4%, 95.0% and 100% at 8 weeks in homozygous extensive metabolizers, heterozygous extensive metabolizers and poor metabolizers, respectively. At 8 weeks, the healing rate of erosive reflux oesophagitis was significantly lower in homozygous extensive metabolizers than in the other two groups (P 〈 0.05). The H. pylori status and serum pepsinogen I/II ratio had less influence than CYP2C19 polymorphism on the healing rate of erosive reflux oesophagitis.Conclusions : The therapeutic effect of lansoprazole on erosive reflux oesophagitis is influenced by the CYP2C19 genotype status. Therefore, a test of CYP2C19 genotype may be useful for the medical treatment of reflux oesophagitis with lansoprazole.

    Notizen: Three-dimensional small-angle neutron scattering (3D-SANS) analysis is derived. This analysis using neutrons, which can penetrate matter easily, is a powerful technique to investigate the anisotropic nanoscale structures in single crystal bulk materials, although it takes a long time to get a complete data set. This technique is applied to the analysis of the anisotropic neutron scattering from a superconductor CeRu2. It suggests that planar defects correlate well with the lattice symmetry of the crystal and influence superconducting properties as pinning centers. It is also shown by simulation that the 3D-SANS analysis is useful even at intense short pulse spallation sources that give a wider band width of neutron wavelength, if the SANS profile is highly anisotropic.

    Notizen: Abstract Matrix metalloproteinases (MMPs) and tissue inhibitors of metalloproteinases (TIMPs) are important in any process of tissue remodeling. However, there is no report evaluating their expression in cardiac allografts in human or nonhuman primates. Heterotopic cardiac transplantation was performed on Japanese monkeys. Subjects were treated with chimeric anti-human lymphocyte function-associated antigen-1 monoclonal antibody for 2 weeks. Heart grafts were harvested at days 1–95 (n = 7). Native monkey hearts were used as controls (n = 2). We examined expression of MMP-2, MMP-9, TIMP-1, and TIMP-2 using immunohistochemistry and in situ reverse transcriptase polymerase chain reaction (RT-PCR). In the myocardium, the expression of MMP-2 was increased in the spindle-shaped cells of acutely rejected myocardial interstitium and prior to the presence of mononuclear cell infiltration at days 1–41. TIMP-1 and 2 expression was enhanced in association with the progression of fibrosis at days 40–95. In the coronary arteries of chronically rejected allografts, enhanced MMP and decreased TIMP expression was observed in both thickened intima and media at days 40–95. The medial MMP mRNA expression was observed before the development of intimal thickening occurred at days 7–28. MMPs are critical for the progression of acute and chronic rejection, and TIMP predominance plays important roles in fibrosis in association with acute rejection. Expression of MMPs and TIMPs is a sensitive indicator of acute and chronic cardiac rejection.
