    Notizen: The witches’ broom disease caused by the fungus Crinipellis perniciosa is the main limiting factor for cocoa production in South America and the Caribbean. In Brazil, this disease affects almost all cocoa-growing regions, causing serious economic, social and ecological damage. The aim of this study was to map genomic regions associated with resistance to C. perniciosa using an F2 population derived from a cross between ‘Scavina-6’(resistant) and ‘ICS-1’(susceptible). The phenotypic index was determined as the average number of vegetative witches’ brooms per canopy area of each plant, the witches’ brooms were counted and eliminated during six field evaluations between May 1998 and August 1999. A total of 124 random amplified polymorphic DNA (RAPD) and 69 amplified fragment length polymorphism (AFLP) markers were mapped along 25 linkage groups covering 1713 cM of cocoa genome. After employing single factor and composite interval mapping analyses, a major quantitative trait loci (QTL) flanked by the marker AV14.940 was identified in the linkage group 11, explaining almost 35% of the resistance to witches’ broom. The present result suggests that this QTL acts as a major dominant component of resistance to this pathogen, with great potential for use in marker-assisted selection procedures in cocoa breeding programmes.

    Notizen: Chub Squalius torgalensis and nase Chondrostoma lusitanicum, in a Mediterranean stream, showed important differences in life-history traits and population dynamics. Both species reached mean maturity at age 2 years. Chub lived up to age 5 years, spawned in March to June, grew at a maximum rate of 0·59 mm mm−1 year−1 and showed a low reproductive allocation, with fecundity and egg size increasing with body size. Nase lived up to age 4 years, spawned in January to April, grew at a maximum rate of 0·46 mm mm−1 year−1 and showed a high reproductive allocation, with egg size independent of body size. Both chub and nase showed moderate fluctuations in population size during 1991–1998, but differed in factors driving density at age. Density of age 1 year juvenile chub decreased following severe summer droughts and proportionate survival prevailed thereafter. Density of age 2 year adult nase decreased following severe spring floods, but neither environmental nor parental stock effects were detected for juveniles and older fishes. The results illustrated the interplay between life history and environmental variability in driving fish population dynamics, with impacts of both summer droughts and spring floods being contingent on species-specific patterns of spawning and reproductive investment.

    Notizen: Squalius alburnoides is a complex of minnows common to the Iberian Peninsula, consisting of two distinct forms. The most common form is comprised of diploid and polyploid asexual hybrids heterozygous for several diagnostic allozyme loci contributed by Squalius pyrenaicus or Squalius carolitertii and a missing ancestor. The other form is diploid and homozygous for the allele contributed by the missing ancestor at these same loci. Present results from analyses of sex ratio and cytochrome b sequences are not consistent with the evolutionary distinctiveness of this non-hybrid form and suggest that it represents an all-male lineage imbedded within an almost all-female complex. This all-male lineage allowed preservation of the ancestral paternal nuclear genome after the paternal ancestor became extinct in all or most drainages, withimportant evolutionary implications.

    Notizen: Nowadays trace organic pollutants represent a major concern in water treatment systems. Activated carbon has been used for most applications aiming at the reduction of these kind of compounds in aqueous effluents, but regeneration needs and high operation costs led to a renewed interest in the search for alternative sorbents. Pine bark is an excedentary raw material from sawmills in Portugal, and therefore a profitable natural resource that has already been successfully tested in the adsorption of organochlorines from contaminated water. This study aims at characterizing the pine bark surface structurally and chemically, to understand the nature of sorption occurring when a trace organic contaminant is present in aqueous effluents. Pentachlorophenol (PCP) was the trace contaminant used in the experiments. Scanning electron microscopy (SEM), mercury porosimetry, Fourier transform infrared spectroscopy (FTIR) and X-ray photoelectron spectroscopy (XPS) were the techniques used, in addition to classical chemical analysis and solid phase micro extraction (SPME) prior to gas chromatography with mass spectrometry (GC-MS) for PCP quantification. The pine bark proved to be a material of very low porosity, low specific surface area, strong carbon aromatic content probably relative to polyphenols and lignin composition. Sorption experiments showed a good correlation for the linear adsorption isotherm, as well as the desorption experiments. In the conditions tested, the average PCP removal after 24?h was above 98 per cent. This material proved to be an encouraging sorbent for cheap water remediation solutions.

    Notizen: Ultrastructural observations of principal cells of the epithelium lining of the proximal caput epididymis in experimental alcoholic albino rats at 180 days of treatment showed pyknotic nuclei, ill-defined cellular organelles and clusters of electrondense bodies, perhaps lysosomes. It was also verified for a progressive accumulation of lipid droplets initially in the basal and perinuclear cytoplasm and finally in the apical cytoplasm of principal cells at 60, 120 and 180 days of experimentation, respectively. The clear cells of alcoholic rats at 180 days showed the cytoplasm totally filled with lipid droplets. These findings were taken comparatively with the morphological features of the same epididymal cells in control (normal) rats.

    Notizen: This work was carried out to determine the effect of overload cycles on the fatigue life of a structural steel used for offshore applications. Single and multiple overloads were adopted and the corresponding fatigue crack growth retardation was evaluated. Residual stress fields were measured in the vicinity of the crack tip using an X-ray diffraction technique and their size compared with that of the overload cyclic plastic zone. In regard to crack growth retardation, the results indicated that the extension in fatigue life increases with an increase in overload, as a consequence of the generation of higher compressive residual stress levels over a larger distance ahead of the crack tip. The effect of two equal and consecutive overloads, with the second one applied at different intervals of crack propagation from the first, was also considered. Larger intervals were shown to lead to a longer residual fatigue life.

    Notizen: From 228 recent Leptospira isolates from mainland Portugal and Azorean wild mammals, 149 were characterized at the serovar level by monoclonal antibodies (MAbs), a quick serological method in epidemiological studies. In order to compare this antigenic information with that from new genetic techniques, a sample of isolates was analyzed through pulsed-field agarose gel electrophoresis (PFGE) (n=71), mapped restriction site polymorphisms (MRSPs) in PCR-amplified rRNA genes (n=45, including 13 saprophytes) and arbitrarily primed polymerase chain reaction (AP-PCR) fingerprinting (n=32). MRSP and AP-PCR lead to species identification of the studied 32 pathogenic isolates: Leptospira interrogans (n=3), Leptospira kirschneri (n=8) and Leptospira borgpetersenii (n=21). MAbs and PFGE characterized pathogenic isolates at the serovar level and resulted mainly in agreement (64%) although many discrepancies (35%) were observed.

    Notizen: The ability for biomass colonization of four porous mineral microcarriers (sepiolite, clay, pozzolana and foam glass-Poraver), was studied and related to their surface properties. The surface hydrophobicity of the mineral carriers was a more important factor influencing colonization by the anaerobic consortium than was surface charge. It was possible to correlate linearly the degree of hydrophobicity with the biomass retention capacity. Although the thermodynamic theory did not explain adhesion, an increase in cell attachment was directly related to the decrease of the positive values of the free energy of adhesion. Surface roughness, porosity and the amount of surface Mg2+, were also determinant factors in bacterial immobilization.
