Notes: Abstract Aciculin is a novel adherens junction antigen extracted from human uterine smooth muscle that is reported to associate biochemically with dystrophin. We attempted to determine (i) the immunostainability of anti-aciculin antibody for the 6 histochemically normal human muscles and seven muscles from boys with Duchenne muscular dystrophy(DMD) and 11 disease control muscles, (ii) the ultrastructural localization of aciculin in normal skeletal myofibers, (iii) aciculin’s spacial relationship with dystrophin and β-spectrin, and (iv) if the aciculin is ultrastructurally colocalized with dystrophin, the distance from the aciculin epitope to the epitope of the dystrophin N- or C-terminal domain. For this, rabbit anti-aciculin antibody was generated against the synthetic peptide of aciculin fragment D [4]. Immunohistochemical staining showed that the immunostainability of DMD muscles for anti-aciculin antibody was markedly decreased as compared with normal and disease control muscles. Single and double immunogold labeling electron microscopy of 6 histochemically normal human quadriceps femoris muscles revealed that aciculin was present along the inner surface of muscle plasma membrane and that aciculin formed doublets more frequently with dystrophin (23.5 ± 1.8%; group mean ± SE) than with β-spectrin (12.8 ± 1.1%; P 〈 0.01 two tailed t test). Rabbit anti-aciculin antibody frequently formed doublets with monoclonal antibodies against the N- or C-terminal domain of dystrophin at the muscle cell surface. These results suggest that aciculin is associated with dystrophin and may interact with both the N- and C-terminal domains of dystrophin.

Notes: Background, aims: The present study was undertaken to assess the periodontal status of a rural Japanese population and to study the correlation between the periodontal status and the serum antibody titers for Porphyromonas gingivalis (Pg) fimbriae.Method: A total of 236 individuals were examined for their periodontal conditions by the use of the community periodontal index for treatment needs (CPITN), and serum antibody titers for Pg fimbriae in their peripheral blood samples were evaluated using the enzyme-linked immunosorbent assay. Results: There was a substantially larger proportion of edentulous subjects in the age group older than 60 years. The remaining teeth were 24.1, 23.2, 11.1 and 10.1 per person in the 40–49, 50–59, 60–69 and 〈inlineGraphic alt="geqslant R: gt-or-equal, slanted" extraInfo="nonStandardEntity" href="urn:x-wiley:03036979:JCPE280311:ges" location="ges.gif"/〉70 age groups, respectively. The % of sextants with a CPITN code of missing sextant (MS) increased towards elderly and reached 〉60% in the age group of 〈inlineGraphic alt="geqslant R: gt-or-equal, slanted" extraInfo="nonStandardEntity" href="urn:x-wiley:03036979:JCPE280311:ges" location="ges.gif"/〉70 years, as the % of the CPITN 2, 1 or 0 sextant decreased. The % of CPITN 4 and 3 sextants did not differ between different age groups and were about 6–8% and 15–20%, respectively. The % of CPITN 1 or 0 sextants was higher in female subjects than in male subjects in the 60–69 and 〈inlineGraphic alt="geqslant R: gt-or-equal, slanted" extraInfo="nonStandardEntity" href="urn:x-wiley:03036979:JCPE280311:ges" location="ges.gif"/〉70 age groups, while the % of CPITN 4 or 3 sextants was higher in male subjects than in female subjects in all age groups. There was no significant difference between various age groups in the mean serum antibody titers for Pg fimbriae. The mean anti-Pg fimbriae antibody titers was significantly higher for the subjects with a maximum CPITN code 4 (max.-CPITN 4 subject) than for the subjects with lower maximum CPITN codes. The antibody titers varied extensively among the max.-CPITN 4 or 3 subjects, but not among the max.-CPITN 2/1/0 or MS subjects. Conclusions: The present study demonstrated that tooth loss is a remarkable event in elderly subjects and that oral prophylaxis and mechanical debridement should be mandatory in the population examined. It was also demonstrated that the serum antibody titers against Pg fimbriae could be useful for screening individuals with moderate to severe periodontitis.

Notes: To construct an amplified-fragment length polymorphism (AFLP)-based molecular linkage map of zoysiagrass, the selfed progenies of a clone consisting of 78 individuals were analysed using 471 AFLP markers derived from 126 PstI/MseI primer combinations. Of these markers, 364 were grouped into 26 linkage groups. The maps covered a total length of 932.5 cM, with an average spacing of 2.6 cM between markers. This information proves useful for gene targeting, quantitative trait loci mapping, and marker-assisted selection in zoysiagrass.

Notes: The nematic liquid-crystal director-orientation in the vicinity of the interface between a liquid-crystal layer and a glass substrate is evaluated in various orientations under an applied electric field, using shear horizontal (SH) wave propagation. The measured acoustic phase delay is related to numerical analysis of the propagation characteristics of the SH wave in a trilayer structure containing a nematic liquid-crystal layer (4-cyano-4′-pentylbiphenyl: 5CB) between two glass plates. This technique using SH wave propagation in a liquid-crystal cell is promising for evaluating the nematic liquid-crystal director angle in the vicinity of the interface. © 2002 American Institute of Physics.

Notes: The electrorheology (ER) effect of ferroelectric liquid crystals is observed under shear-horizontal (SH) wave propagation. The ER effect is strongly dependent on the orientation direction of the liquid-crystal molecule. The apparent viscosity is evaluated through the numerical analysis result on the propagation characteristics of a SH wave in a trilayer structure including a ferroelectric liquid-crystal (Chisso, CS1014) layer between two glass plates. The ER effect is examined by the relation between the orientation of smectic layers and the propagation direction of the SH wave. © 2001 American Institute of Physics.

Notes: This letter describes the memory effect of an AlGaAs/GaAs heterojunction field-effect transistor that contains InAs nanodots in the barrier layer. The device experiences a shift of threshold gate voltage, as a function of the amount of the electrons trapped in the nanodots. These trapped electrons can be injected by applying a positive gate voltage and be erased by a visible light illumination at negative gate bias. Although the shift of the threshold gate voltage volatilizes with the time after the memory programing operation, a considerable part of the shift is retained even after 100 h at room temperature. © 2000 American Institute of Physics.

Notes: Background : Omeprazole 10 mg is used as maintenance therapy for gastro-oesophageal reflux disease, but previous reports have not mentioned the potency of its acid suppression.Aim : To evaluate the potency of acid suppression with omeprazole 10 mg, in relation to CYP2C19 genotypes. Methods : Eighteen healthy subjects without Helicobacter pylori participated. After a 7-day regimen of omeprazole 10 mg, 20 mg, lafutidine 20 mg (a novel H2-receptor antagonist) or water only (baseline data), intragastric pH was measured for 24 h.Results : With omeprazole 10 mg, greater differences were observed than 20 mg in median pH values and pH 〉 4 holding time ratios between poor metabolizers (PMs, n = 6) and the others [homozygous extensive metabolizers (homo-EMs, n = 6) and heterozygous extensive metabolizers (hetero-EMs, n = 6)]. With lafutidine 20 mg, these parameters were not influenced by the genotype. The potency of acid suppression was: omeprazole 20 mg ≈ lafutidine 20 mg 〉 omeprazole 10 mg in homo-EMs, omeprazole 20 mg 〉 omeprazole 10 mg ≈ lafutidine 20 mg in hetero-EMs, and omeprazole 20 mg ≈ omeprazole 10 mg 〉 lafutidine 20 mg in PMs.Conclusions : Omeprazole 10 mg strongly suppresses acid secretion, but depending on the CYP2C19 genotypes shows greater interindividual variations in suppression than 20 mg.

Notes: Background : Insufficient acid suppression is one of the main causes of proton pump inhibitor-refractory gastro-oesophageal reflux disease.Aim : To achieve more potent and long-lasting acid suppression, different dosage regimens of rabeprazole were compared in relation to the CYP2C19 genotype status. Methods : In a cross-over study, 18 healthy Helicobacter pylori-negative males (six homozygous extensive metabolizers, six heterozygous extensive metabolizers and six poor metabolizers) were given rabeprazole 10 mg once daily, 20 mg once daily or 10 mg twice daily, or water only (baseline data), for 7 days each. On day 7 of each regimen, 24-h intragastric pH-metry was performed. Results : No significant differences were observed in median pH values and pH 〉 4 holding time ratios between rabeprazole 10 mg once daily and 20 mg once daily. However, with rabeprazole 10 mg twice daily, these parameters were significantly higher than those with 20 mg once daily. The potency of acid suppression by rabeprazole was influenced by the CYP2C19 genotype status. The differences were somewhat significant but not large. The incidence of nocturnal acid breakthrough was lowest with rabeprazole 10 mg twice daily. Conclusions : Rabeprazole 10 mg twice daily, not 20 mg once daily, should be administered to achieve more potent and long-lasting acid suppression.
