Notes: A small organic molecule (CUR-162590) that selectively enhances survival of midbrain dopaminergic neurons was identified by screening small molecule compound libraries. In embryonic midbrain cultures, CUR-162590 increased dopamine uptake and the number of dopaminergic neurons without altering the number of total neurons or astroglia or the uptake of GABA or serotonin. CUR-162590 reduced apoptosis of cultured dopaminergic neurons and protected against death induced by toxins such as MPP+. Several synthetic analogs of CUR-162590 also had similar bioactivities. CUR-162590 thus represents a new class of neurotrophic small molecules that may have utility in the treatment of Parkinson's disease, which is marked by degeneration of midbrain dopaminergic neurons.

Notes: Pruritus associated with intrathecal opioid administration is particularly common in pregnancy. Nonsteroidal anti-inflammatory drugs (NSAIDs) can reduce the severity of this pruritus but have undesirable side effects. The recent development of drugs that can specifically inhibit the cyclooxygenase 2 isoenzyme have become an attractive alternative. This study was designed to evaluate the efficacy of such a drug (celecoxib) in reducing intrathecal opioid-induced pruritus in a randomised double-blinded study of 60 women undergoing Caesarean section. All of them received spinal anaesthesia with 3 ml of 0.5% hyperbaric bupivacaine and 0.3 mg preservative-free morphine. After delivery of the baby, they received either oral celecoxib 200 mg or placebo. Visual analogue scores for pain and pruritus were measured at 30 min, 2, 4, 8, and 24 h. There was no difference in the severity and onset of pain and pruritus between the two groups. Timing of administration, inadequate dosing and possible altered pharmacokinetics in pregnancy may explain the lack of efficacy.

Notes: An oxidation study of plasma-enhanced chemical vapor deposited (PECVD) and rf sputtered hydrogenated amorphous silicon carbide (a-Si1−xCx:H) films was carried out using the infrared (IR) and electron spin resonance (ESR) techniques. a-Si1−xCx:H films with x=0.3, 0.5, and 0.8 were prepared with the PECVD method and significant oxide growth can only be obtained in a-Si0.2C0.8:H film. IR results showed that Si–CH3 bonds provide the necessary porous structure for oxide growth. The oxide quality was found to be poor when compared to oxide obtained from pure silicon. Annealing in nitrogen enhances the porosity of the PECVD films and results in faster oxide growth. For sputtered films, oxide growth can only be observed in film containing Si–CH3 bonds. We are not able to separate the contributions of the Si–H, Si–C, Si–CH3, and the Si dangling bonds to the oxide growth for the PECVD and rf sputtered films. © 2001 American Institute of Physics.

Notes: Smoking is a risk factor for complications during and after surgery, but most smokers are unable to quit before elective surgery. We tested the efficacy of bupropion in improving smoking cessation rates in this setting by enrolling 47 patients from the elective surgery waiting list in a double-blind randomised controlled trial. Patients receiving bupropion had a lower daily cigarette consumption at the time of hospital admission, median (IQR) cigarettes per day: 6 (2–7) vs. 15 (9–20), p = 0.046. They also had a reduction in end-expired carbon monoxide (p = 0.004), a known contaminant of cigarette smoke, and increased arterial oxygen saturation on pulse oximetry (p = 0.011). They were more likely to have stopped smoking at the 3-week visit (p = 0.036), but not at the 6-week visit (p = 0.25) or at the time of hospital admission for surgery (p 〉 0.99). This study found that smokers waiting for elective surgery are more likely to reduce or stop smoking when treated with bupropion.

Notes: Abstract  We report two cases of lateral ventricle dilatation due to membranous occlusion of the foramen of Monro following ventriculoperitoneal shunt insertion. Both cases were treated successfully by endoscopic foraminoplasty of the obstructed foramen of Monro and III ventriculostomy. One child had meningomyelocele and hydrocephalus. She had CSF infection after repair of the back lesion. Isolated left lateral ventricle occurred after insertion of a right ventriculoperitoneal shunt for hydrocephalus when the girl was 2 months old. A right ventriculoperitoneal shunt was then inserted. Chronic shunt infection with abdominal pseudocyst was found 8 years later. The shunts were exteriorized. Membranous obstruction of the foramen of Monro was found endoscopically. Fenestration of the membranous obstruction along with a III ventriculostomy was performed. After the endoscopic procedure, the exteriorized ventriculoperitoneal shunt was removed 2 weeks later. The patient was still symptom free without shunting 14 months after the operation. The other child had hydrocephalus after a premature birth and hemorrhage. Repeated ventriculoperitoneal shunt infections contributed to membranous obstruction of bilateral foramen of Monro. After the shunt infection was treated this patient’s shunting procedure was simplified by endoscopic foraminoplasty of the left and right foramen of Monro along with a III ventriculostomy. He was symptom free with a new ventriculoperitoneal shunt 9 months after the operation.

Notes: Abstract  One hundred and twenty four microsatellites were isolated from analysis of 5000 Vitis expressed sequence tags (ESTs). A diversity of dinucleotide and trinucleotide simple sequence repeat (SSR) motifs were present. Primers were designed for 16 of these SSRs and they were tested on seven accessions. Ten of the sixteen primer pairs resulted in PCR products of the expected size. All ten functional primers were polymorphic across the accessions studied. Polymorphisms were evident at the level of cultivars, Vitis species, and between related genera. SSRs that were from the 3′ untranslated region (3′UTR) were most polymorphic at the cultivar level, the 5′ untranslated region (5′ UTR) SSRs were most polymorphic between cultivars and species, and those SSRs within coding sequence were most polymorphic between species and genera. These results show that EST-derived SSRs in Vitis are useful as they are polymorphic and highly transferable. With EST SSRs being applicable to studies at several taxonomic levels, the large number of SSRs (approximately 1000) that will be available from an expanded EST database of 45 000 will have many potential applications in mapping and identity research.

Notes: Abstract: Using neutron reflectivity, we directly and self-consistently obtained all the parameters of the critical adsorption profile of three critical mixtures (n-hexane+perfluorohexane, deuterated methanol+cyclohexane and methanol+deuterated cyclohexane) assuming only a Liu-Fisher form for the profile. The characteristic exponent and the amplitude ratio which can be deduced from the neutron spectra alone are in good agreement with one another, with ellipsometric data and with theoretical values. Owing to auxiliary measurements of coexistence curves and correlation lengths, we could also check the amplitude of the intermediate and distal parts of the profile with good agreements for the methanol cyclohexane mixtures only. The adsorbed amount is much greater in the n-hexane+perfluorohexane mixture where both the power-law amplitude and the characteristic length of the profile are affected.

Notes: Abstract  Poly(N-isopropylacrylamide) (PNIPAM) precipitates out of water around 32 °C. This critical temperature is raised when hydrophilic acrylamide sequences are present on the polymer chain. We have used neutron scattering to study the structural properties of a statistical copolymer containing acrylamide and N-isopropylacrylamide segments at different temperatures and its interactions with an anionic surfactant, sodium dodecyl sulfate (SDS). At low temperatures, the copolymer behaves as a swollen polymer coil. With an increase in temperature, intermolecular attractions are observed, and close to the critical temperature of the copolymer, microphase separation is observed. Here, the structure consists of dense nodules of hydrophobic sequences stabilized by hydrophilic sequences. In the presence of a small amount of SDS, additional colloidal stability is observed: the nodule size is decreased. At high SDS concentration, the copolymer is completely solubilized at all temperatures studied and the structure of the polymer–surfactant complex resembles the “necklace” structure obtained for the homopolymer PNIPAM–SDS system.
