    Notes: Abstract The structure and crystallographic orientation of mineral phase in the oblique prismatic layer of Mytilus edulis shell were studied by SEM, XRD and TEM with selected area electron diffraction (SAED). A crystallographic orientation regulation, i.e. the adjacent 1–5 calcite prisms with the same three-dimensional orientation in the oblique prismatic layer, was found for the first time. It is observed that the calcite prisms in the oblique prismatic layer were grown with their (104) parallel to the shell surface.

    Notes: Abstract. ATP-sensitive K+ (KATP) channels have been characterized in pituitary GH3 cells with the aid of the patch-clamp technique. In the cell-attached configuration, the presence of diazoxide (100 μm) revealed the presence of glibenclamide-sensitive KATP channel exhibiting a unitary conductance of 74 pS. Metabolic inhibition induced by 2,4-dinitrophenol (1 mm) or sodium cyanide (300 μm) increased KATP channel activity, while nicorandil (100 μm) had no effect on it. In the inside-out configuration, Mg-ATP applied intracellularly suppressed the activity of KATP channels in a concentration-dependent manner with an IC50 value of 30 μm. The activation of phospholipase A2 caused by mellitin (1 μm) was found to enhance KATP channel activity and further application of aristolochic acid (30 μm) reduced the mellitin-induced increase in channel activity. The challenging of cells with 4,4′-dithiodipyridine (100 μm) also induced KATP channel activity. Diazoxide, mellitin and 4,4′-dithiodipyridine activated the KATP channels that exhibited similar channel-opening kinetics. In addition, under current-clamp conditions, the application of diazoxide (100 μm) hyperpolarized the membrane potential and reduced the firing rate of spontaneous action potentials. The present study clearly indicates that KATP channels similar to those seen in pancreatic β cells are functionally expressed in GH3 cells. In addition to the presence of Ca2+-activated K+ channels, KATP channels found in these cells could thus play an important role in controlling hormonal release by regulating the membrane potential.

    Notes: Abstract  The product of the Wilms' tumor gene WT1 is a transcription factor overexpressed not only in leukemic blast cells of almost all patients with acute myeloid leukemia, acute lymphoid leukemia, and chronic myeloid leukemia, but also in various types of solid tumor cells. Thus, it is suggested that the WT1 gene plays an important role in both leukemogenesis and tumorigenesis. Here we tested the potential of WT1 to serve as a target for immunotherapy against leukemia and solid tumors. Four 9-mer WT1 peptides that contain HLA-A2.1-binding anchor motifs were synthesized. Two of them, Db126 and WH187, were determined to bind to HLA-A2.1 molecules in a binding assay using transporter associated with antigen processing-deficient T2 cells. Peripheral blood mononuclear cells from an HLA-A2.1-positive healthy donor were repeatedly sensitized in vitro with T2 cells pulsed with each of these two WT1 peptides, and CD8+ cytotoxic T lymphocytes (CTLs) that specifically lyse WT1 peptide-pulsed T2 cells in an HLA-A2.1-restricted fashion were induced. The CTLs also exerted specific lysis against WT1-expressing, HLA-A2.1-positive leukemia cells, but not against WT1-expressing, HLA-A2.1-negative leukemia cells, or WT1-nonexpressing, HLA-A2.1-positive B-lymphoblastoid cells. These data provide the first evidence of human CTL responses specific for the WT1 peptides, and provide a rationale for developing WT1 peptide-based adoptive T-cell therapy and vaccination against leukemia and solid tumors.

    Notes: Abstract Vallecular cyst, a rare but generally benign lesion in the larynx, may cause stridor and even life-threatening airway obstruction in early infancy. We retrospectively studied 14 cases of newborn infants with vallecular cyst. There was no gender predilection and most cases were full-term and appropriate for gestational age. The clinical presentations included stridor, chest wall retraction, feeding difficulties and failure to thrive. Laryngomalacia was the most common associated anomaly. Flexible laryngoscopy was sufficient for diagnosing the vallecular cyst and larygmalacia. Maintenance of airway patency, nutritional support, and de-roofing of the cyst were the mainstays of management. Conclusion Vallecular cyst should be included in the differential diagnosis of stridor in newborn infants. Respiratory and feeding difficulties in these patients can be dramatically improved after appropriate surgical removal of the cyst.

    Notes: Fractionation of the extract of Aspergillus niger. IFB-E003, an endophyte in Cyndon dactylon, gave four known compounds naphtho-γ-pyrones rubrofusarin B, fonsecinone A, asperpyrone B and aurasperone A, which were further investigated biologically. Rubrofusarin B was shown to be cytotoxic to the colon cancer cell line SW1116 (IC50: 4.5 μg ml−1), and aurasperone A inhibitory on XO (xanthine oxidase) (IC50: 10.9 μmol l−1). Moreover, the four naphtho-γ-pyrones exhibited growth inhibitions against the five test microbes with MICs ranging in between 1.9 and 31.2 μg ml−1. The present recognition of rubrofusarin B and aurasperone A as strong co-inhibitors on XO, colon cancer cell and some microbial pathogens is of significance for the imperative discovery of new relevant therapeutic agents.

    Notes: The role of energetic electrons in periodic self-oscillations of a discharge plasma has been studied by measuring the spatiotemporal evolution of plasma potential, electron density, and electron velocity distribution function. It is found that the self-oscillation involves the instabilities of sheaths, propagation of a double layer and competition between the ionization, thermalization, and diffusion. The energetic electrons are the key factor which links these processes to form the oscillation cycle. The time interval of each phase in the cycle is estimated according to the physical process and the calculations are in agreement with experimental measurements. The study of the probe perturbation effect on the oscillations indicates that the length of the oscillation period is related to the amount of energetic electrons; the more energetic electrons, the shorter the period. © 2001 American Institute of Physics.

    Notes: The magneto-optical and optical properties of GdFe single layer films, which are covered with a thin Si3N4 layer, were studied in the visible wavelength region. A Kerr rotation peak and a reflectivity drop were observed near 4.1 eV in GdFe alloyed films and attributed to the Gd element. Compared with the single thick GdFe film, the Kerr effect of SiN/GdFe bilayers was enhanced, due to the optical interference between Si3N4 and GdFe. The Kerr rotation of GdFe films showed a nonlinear function of the compositions in the whole measured wavelength range. Magneto-optical measurements directly evidence the spin–flip in the GdFe films as the Gd content increased from 20.7 to 24.2 at. %, which showed advantages over conventional magnetometry. © 2001 American Institute of Physics.

    Notes: Summary.  With 1.3 billion people, China has the largest population in the world, and therefore has the largest population of persons with haemophilia (PWH). As there is no national registry for haemophilia, it is difficult to ascertain how many PWH have actually been diagnosed. Between January 1983 and June 2002, 1312 patients with coagulation disorders were referred to our hospital, and 1190 patients were evaluable. Among them, 1069 (89.8%) patients had haemophilia, 68 had vWD, 20 had factor XI deficiency, 10 had acquired factor VIII inhibitor and 23 had other coagulation disorders. Of the 1069 PWH, 14.7% were unclassified, 38.4% severe, 35.7% moderate and 11.1% mild. If the unclassified cases were excluded, 45.1% were severe, 41.9% moderate and 13.0% mild. Twenty-nine of the 68 vWD patients had vWF:Ag 〈5%, and subcategorized as type 3 vWD. Because vWF multimer analysis was not performed in our centre, the remaining vWD patients were not subdivided.
