    Notes: Abstract  Formation of the otocyst from the otic placode appears to differ from invagination of other cup-shaped organ primordia. It is known that the cellular cytoskeleton plays a limited role in otic placode invagination, whilst the extracellular matrix underlying the otic primordium intervenes in the folding process. In this study we have analysed the role of the basal lamina heparan sulphate proteoglycan in otic primordium invagination. At 10 H.H. stage, heparan sulphate proteoglycan immunomarking begins to appear on the otic placode basal lamina, increasing noticeably at 13 H.H. stage, coinciding with maximum folding of the otic epithelium, and is still present at later stages. Enzyme degradation of heparan sulphate proteoglycan in the otic primordium basal lamina, by means of microinjection with heparinase III prior to folding, significantly disrupts invagination of the otic placode, which remains practically flat, with a significant reduction in the depth of the otic pit and an increase in the diameter of the otic opening. The immunocytochemistry analysis revealed a notable depletion of basal lamina heparan sulphate proteoglycan in the otic primordia microinjected with heparinase, with no statistically significant differences observed in the volume or rate of cell proliferation in the otic epithelium relative to the control, which suggests that heparan sulphate proteoglycan disruption does not interfere with the epithelial growth. In addition, a study of apoptosis distribution by the TUNEL method confirmed that treatment with heparinase does not cause interference with cell survival in the otic epithelium. Our findings support the theory that otic primordium invagination may be regulated, at least in part, by the basal lamina components, which might contribute towards anchoring the otic epithelium to adjacent structures.

    Notes: The interaction potential energy surface (PES) of H4 is of great importance for quantum chemistry as a test case for molecule–molecule interactions. It is also required for a detailed understanding of certain astrophysical processes, namely collisional excitation and dissociation of H2 in molecular clouds, at densities too low to be accessible experimentally. The 6101 ab initio H4 energies reported in 1991 by Boothroyd et al. demonstrated large inaccuracies in analytic H4 surfaces available at that time. Some undesirable features remained in the more accurate H4 surfaces fitted to these energies by Keogh and by Aguado et al., due in part to the relatively sparse coverage of the six-dimensional H4 conformation space afforded by the 6101 ab initio energies. To improve the coverage, 42 079 new ab initio H4 energies were calculated, using Buenker's multiple reference (single and) double excitation configuration interaction program. Here the lowest excited states were computed as well as the ground state, and energies for the original 6101 conformations were recomputed. The ab initio energies have an estimated rms "random" error of ∼0.5 millihartree and a systematic error of ∼1 millihartree (0.6 kcal/mol). A new analytical H4 PES was fitted to these 48 180 ab initio energies (and to an additional 13 367 points generated at large separations), yielding a significant improvement over previous H4 surfaces. This new PES has an rms error of 1.43 millihartree relative to these 48 180 ab initio energies (the fitting procedure used a reduced weight for high energies, yielding a weighted rms error of 1.15 millihartree for these 48 180 ab initio energies). For the 39 064 ab initio energies that lie below twice the H2 dissociation energy, the new PES has an rms error of 0.95 millihartree. These rms errors are comparable to the estimated error in the ab initio energies themselves. The new PES also fits the van der Waals well to an accuracy of about 5%. For relatively compact conformations (energies higher than the H2 dissociation energy), the conical intersection between the ground state and the first excited state is the largest source of error in the analytic surface. The position of this conical intersection forms a somewhat complicated three-dimensional hypersurface in the six-dimensional conformation space of H4. A large portion of the position of the conical intersection has been mapped out, but trying to include the conical intersection explicitly in an analytic surface is beyond the scope of the present paper. © 2002 American Institute of Physics.

    Notes: Objectives 1. To determine the prevalence of endometrial hyperplasia in postmenopausal women taking standard proprietary regimens of sequential oestrogen/progestogen; 2. to determine the effects of nine months treatment with an oral continuous combined regimen of 2 mg 17ß-oestradiol and 1 mg norethisterone acetate (Kliofem [Kliogest outside the UK]; Novo Nordisk, Denmark) on endometrial histology in postmenopausal women.Design An open, prospective study in postmenopausal women.Setting Fifty-four menopause clinics in the UK.Participants 2028 postmenopausal women: 1312 (Group A) taking sequential oestrogen-progestogen hormone replacement therapy (HRT), and 716 (Group B) not taking HRT, were recruited. In Group A, 388 women took preparations containing 10 days of progestogen, 921 had 12 days, and 3 had 13 days per cycle.Methods Endometrial aspiration biopsies were taken towards the end of a three-month run-in period (Group A) or at study entry (Group B), before administration of the continuous combined HRT regimen. Biopsies were repeated at the end of the nine month treatment period.Main outcome measure Endometrial histology.Results Initial endometrial biopsy data were available for 1106 women in Group A, who by the time of endometrial investigation had been taking HRT for a median duration of 2.56 years (5th to 95th centiles: 0.77 to 8.49 years). Data were available for 661 untreated women, who had no bleeding and had not taken HRT within the last year (Group B). Complex hyperplasia was found in 59 women (5.3%), and atypical hyperplasia in a further eight (0.7%) in Group A. In Group B there were no cases with complex hyperplasia, but one woman showed atypical hyperplasia (0.2%). At the end of the nine months of continuous combined therapy there was no case of hyperplasia among 1196 biopsies (upper 95% confidence limit of risk 0.31%) in women completing the study. Within this Group all of the women with complex hyperplasia arising during previous sequential HRT and who completed the study (n= 38) reverted to normal endometrial patterns. There was no case of endometrial carcinoma during the study.Conclusions Despite taking standard regimens of sequential HRT containing at least 10 days of progestogen, there was a 5.3% prevalence of complex endometrial hyperplasia, and a 0.7% prevalence of atypical hyperplasia. However, continuous combined HRT (Kliofem) containing daily progestogen is not associated with an increased risk of hyperplasia and will convert the endometrium to normal in those with complex hyperplasia arising during previous sequential HRT.

    Notes: Background:  Intratumor hypoxia has been shown to promote more aggressive and metastatic cancer phenotypes that are associated with treatment resistance and poor prognosis. Cellular proliferation and its control are known to be important components of tumor progression. Hypoxia induces cell-cycle arrest in cultured cell lines, possibly via up-regulation of the cyclin-dependent kinase inhibitor p27. The effect of hypoxia on cell-cycle regulation in excised human tumors has not been investigated.Methods:  We performed immunohistochemistry for p27 and Ki-67 on 10 formalin-fixed paraffin-embedded metastatic melanomas, selected on the basis of histological evidence of zonal/geographic necrosis, adjacent to areas with viable perivascular tumor cells.Results:  In the majority of cases, there was a significant increase in p27 staining in cells adjacent to necrotic areas compared to perivascular zones. An inverse staining pattern between Ki-67 and p27 was identified in these tumors. Tumors with no zonal increase in p27 staining demonstrated a diffuse pattern of staining for Ki-67 within tumor nests.Conclusions:  While increased cellular proliferation is a characteristic of cancer, subsets of human melanomas may retain the ability to regulate their rate of proliferation in response to changes in the tumor microenvironment. The hypoxia-mediated cell-cycle arrest (decreased Ki-67) in these tumors may be mediated by p27 up-regulation.

    Notes: Objective:  To detect a possible allergen in a florist with occupational dermatitis. Patient and methods: a 27-year-old atopic female working in a greenhouse since 10 years presented with a 3 months history of severe eczema involving hands, face, eyelids and neck requiring a stop of the work. Tests were performed with European standard series (Trolab®), additional series, plant series, (Chemotechnique Diagnostics®) and flowers. Readings were performed at D2 and D3 according ICDRG criteria. Results: positive patch tests were parabens +/+ nickel sulphate ++/++ cobalt chloride −/++ palladium chloride ++/++ thiomersal ++/++, Iris petal ++/+. Then various colours of Iris were tested and pale blue, yellow and white petals were positive ++/++ but purple blue petal was negative. The proximal yellow part of purple blue petal and orange part of yellow petal were also positive ++/++. The leaf and pistil of Iris were negative. One control with a white Iris petal was negative. Then the patient presented flare up of her dermatitis in the presence of Iris, at home, without handling them, that could make suspect an airborne contact dermatitis. Even after examination by a botanist, the precise identificaton of these Iris was not possible. Positive patch tests to unspecified Iris (petal and leaf) have already been reported in a florist who was allergic to safflower (Van der Willigen A.H. and al, Contact Dermatitis, 1987, 17,184). Conclusion: this case could evoke the responsibility of anthocyans, which play a role in the colours, but essential oils or other components are also possible responsibles.

    Notes: Using a serial bilayer model we show that heterogeneous ferroelectric systems may exhibit substantial Maxwell–Wagner piezoelectric relaxation. The relaxation has its origin in the coupling of the dielectric and piezoelectric properties. The model predicts both retardation (positive phase angle and counterclockwise charge–pressure hysteresis) and relaxation (negative phase angle and clockwise charge–pressure hysteresis) of the longitudinal direct piezoelectric coefficient. The theoretical results are confirmed experimentally in ferroelectric ceramic–ceramic bilayers and in a single-phase ferroelectric Aurivillius compound ceramic in which neighboring grains with strongly anisotropic properties may behave as basic Maxwell–Wagner units. © 2001 American Institute of Physics.

    Notes: Transport in the collisional regime is analyzed here for tokamaks and compact toroid of any shape. Toroidal axisymmetry is assumed. The treatment is performed with an improved system of coordinates, here described. Pfirsch–Schlüter current is determined in a general case. The average velocity around a magnetic surface is found as a function of closed geometrical integrals and separated factors that include the electric and magnetic fields. The contribution of the induced electric field Ecursive-phi is analyzed and the similarities with the Ware pinch are shown. The general equations have been applied to families of nested curves with elliptic shapes. © 2000 American Institute of Physics.

    Notes: This paper reports experimental results showing that coherent helical structures are formed in the reversed field pinch (RFP) self-organizing plasma core as a result of transitions to states where the n-spectrum of the m=1 modes is dominated by a single (1,nmax) geometrical helicity. These states are dubbed quasi-single helicity (QSH) states. Their magnetic and thermal properties measured in the reverse field experiment (RFX) [G. Rostagni, Fusion Eng. Des. 25, 301 (1995)] device are described. The present theoretical understanding of QSH states is discussed and some recent theoretical results are presented. The role of aspect ratio is discussed. These results represent a significant step to open a path beyond the standard paradigm that a bath of magnetohydrodynamic (MHD) modes is intrinsic to the RFP. © 2000 American Institute of Physics.

    Notes: Caenorhabditis elegans has previously been proposed as an alternative host for models of infectious disease caused by human pathogens. When exposed to some human pathogenic bacteria, the life span of nematodes is significantly reduced. We have shown that mutations in the age-1, and/or age-2 genes of C. elegans, that normally enhance life expectancy, can also increase resistance to killing by the bacterial pathogens Pseudomonas aeruginosa, Salmonella enterica var. Typhimurium, Burkholderia cepacia or Yersinia pseudotuberculosis. We also found that the rate at which wild-type C. elegans was killed by the bacterial pathogens tested increased as nematodes aged. In the case of P. aeruginosa infection, the difference in life span of wild type and age-1 mutants of C. elegans was not due to differences in the level of bacterial colonisation of the gut.

    Notes: Caenorhabditis elegans has previously been used as an alternative to mammalian models of infection with bacterial pathogens. We have developed a liquid-based assay to measure the effect of bacteria on the feeding ability of C. elegans. Using this assay we have shown that Pseudomonas aeruginosa strain PA14, Burkholderia pseudomallei and Yersinia pestis were able to inhibit feeding of C. elegans strain N2. An increase in sensitivity of the assay was achieved by using C. elegans mutant phm-2, in place of the wild-type strain. Using this assay,P. aeruginosa PA01 inhibited the feeding of C. elegans mutant phm-2. Such liquid-based feeding assays are ideally suited to the high-throughput screening of mutants of bacterial pathogens.
