    Notes: Abstract. Objective: Expression of CINCs was regulated differentially in lipopolysaccharide-stimulated rat macrophages. We examined whether the expression of CINCs in lipopolysaccharide-stimulated rat macrophages is similarly inhibited by anti-inflammatory drugs.¶Materials and Methods: Rat peritoneal macrophages were stimulated by lipopolysaccharide in the presence of anti-inflammatory steroids (dexamethasone, prednisolone and hydrocortisone) or non-steroidal anti-inflammatory drugs (indomethacin and piroxicam). The production and mRNA expression of three types of CINCs were measured using enzyme-linked immunosorbent assay and northern hybridization.¶Results: Anti-inflammatory steroids; dexamethasone, prednisolone and hydrocortisone, dose-dependently inhibited the production of CINC-1, -2 and -3, whose inhibitory patterns were similar to each other. Furthermore mRNA expression of each CINC was inhibited by dexamethasone in a dose-dependent manner. In contrast, non-steroidal anti-inflammatory drugs, indomethacin and piroxicam were without effect. Expression of each CINC was regulated differently; the production of CINC-1 reached a maximum at 12 h and then slightly decreased after lipopolysaccharide stimulation, whereas that of CINC-2 and CINC-3 increased up to 24 h. Dexamethasone inhibited the CINCs production and mRNA expression at 9 h after lipopolysaccharide stimulation.¶Conclusions: These results indicate no difference among CINC-1, -2 and -3 in the inhibition of production and mRNA expression of CINCs by anti-inflammatory steroids, although lipopolysaccharide differentially induces expression of each CINC expression in culture of rat macrophages.

    Notes: Background Th2 and Th1 cells have been suggested to express CCR3/CCR4 and CCR5/CXCR3, respectively.Objective We examined CCR3, CCR4, CCR5 and CXCR3 expression and cytokine production in peripheral blood CD4+ T cells from patients with atopic dermatitis (AD), which has been postulated to be a Th2-type cell-mediated disease, and then analysed the possible correlation between these values and the levels of several clinical parameters.Methods Intracellular cytokine production and chemokine receptor expression in peripheral blood CD4+ T cells from 40 AD patients and 20 sex- and age-matched healthy control subjects were studied by flow cytometry.Results The frequencies of IL-4- and IL-13-producing CD4+ T cells from patients with AD were significantly higher than those from healthy control subjects (IL-4:3.9 ± 2.1% vs. 1.6 ± 0.7%, P = 0.0005, IL-13:4.0 ± 2.1% vs. 1.8 ± 0.8%, P = 0.0023), whereas the frequencies of IL-2- and IFN-γ-producing CD4+ T cells were significantly decreased in AD patients (IL-2:38.1 ± 10.3% vs. 51.3 ± 6.3%, P = 0.0003, IFN-γ: 9.9 ± 3.5% vs. 26.4 ± 4.6%, P 〈 0.0001). The percentage of CCR4+ cells in CD4+ CD45RO+ T cells in AD patients was significantly higher than that in healthy control subjects (24.4 ± 8.0% vs. 10.9 ± 2.3%, P 〈 0.0001) and was correlated positively with the total serum IgE, serum lactic dehydrogenase (LDH) level, eosinophil number, eruption score, and IL-4 and IL-13 secretion in CD4+ T cells, and inversely with IL-2 and IFN-γ secretion in CD4+ T cells. In contrast, CCR3 was not detected on circulating CD4+ T cells even in AD patients. On the other hand, the percentage of CCR5+ or CXCR3+ cells in CD4+ CD45RO+ T cells in AD patients was significantly decreased (CCR5:23.2 ± 7.0% vs. 28.4 ± 5.4%, P = 0.023, CXCR3:29.9 ± 11.4% vs. 38.5 ± 6.7%, P = 0.028) and was positively correlated with eruption score (P 〈 0.05). Multiple regression analyses showed that the percentage of CCR4 expression highly correlated with serum IgE, LDH, eosinophil number and eruption in AD patients.Conclusion CCR4+ cells might be involved in the aetiopathogenesis of AD.

    Notes: Hatchery-reared juvenile black sea bream Acanthopagrus schlegeli (Bleeker) are characterized by high lipid storage and low levels of highly unsaturated fatty acids in their bodies. In the present study, we assessed the effects of dietary fortification of the diet with eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA) on the improvement of physiological activity. Black sea bream 50 days old with an average weight of 0.02 g were reared for 50 days on either a control diet (commercial diet) or a commercial diet fortified with 3% EPA and DHA. The fortification with EPA and DHA reduced lipid storage in adipocytes in the intraperitoneal cavity, but decreased muscle lipid level. Consequently, the total lipid level decreased in the bodies of the fish. The proportions of EPA and DHA in muscle, liver and adipocytes were markedly increased by diet fortification, and the EPA and DHA proportions did not differ from those of wild fish. The cell diameter of adipocytes was reduced by EPA and DHA fortification. Liver function and resistance to air-dipping were improved by fortification. The results of a starvation test revealed the efficient mobilization of lipid reserves in response to energy demands prior to protein exhaustion in the EPA/DHA-enriched group. The results implied that increasing the incorporation of dietary EPA and/or DHA should be considered in the larval stage of black sea bream culture.

    Notes: Summary Background  Wrinkling and sagging of the skin during photoageing is physiologically associated with diminished elasticity, which can be attributed to increased fibroblast-derived elastase activity. This degrades the dermal elastic fibres needed to maintain the three-dimensional structure of the skin. We previously reported that ovariectomy accelerates ultraviolet (UV)B-induced wrinkle formation in rat hind limb skin by altering the three-dimensional structure of elastic fibres.Objectives  In this study, we used hairless mice to assess the effects of ovariectomy with or without chronic UVA or UVB radiation on sagging and wrinkling of skin, on the elasticity of skin, as well as on matrix metalloproteinase activities in the skin.Methods  Ovariectomies or sham operations were performed on 6-week-old female ICR/HR hairless mice.Results  Even in the ovariectomy group without UV irradiation, the skin elasticity was significantly decreased during the 3–13 weeks after ovariectomy, which was accompanied by a significant increase in elastase activity in the skin. After UVA or UVB irradiation, skin elasticity was significantly decreased to a greater extent in the ovariectomy group than in the sham operation group, and this was accompanied by a reciprocal increase in elastase activity but not in the activities of collagenases I or IV in the skin. Consistent with the decreased skin elasticity, UVA irradiation for 12 weeks elicited more marked sagging in the ovariectomy group than in the sham operation group. UVB irradiation for 12 weeks also induced more marked wrinkle formation in the ovariectomy group than in the sham operation group.Conclusions  These results suggest that ovariectomy alone is sufficient to accelerate skin ageing and to increase UV sensitivity, which results in the further deterioration of the skin and photoageing, and may account for the accelerated skin ageing seen in postmenopausal women.

    Notes: Summary Background A system has been developed whereby the morphology of the skin surface can be evaluated directly in three dimensions. This system employs a non-invasive device that utilizes white light of halogen origin, and which allows the computation of wrinkle depth and width, and other parameters of skin surface morphology. Using innovative engineering, an optical system has been devised so that light is transmitted via a slit and can be used to measure not only replicas of the skin but also the skin surface directly. The measurement area is 6·4 × 6·4 mm, and the theoretical resolution with a × 50 magnification lens is within 12·5 µm. Objectives To use this system to study age-related changes in the morphology of wrinkles at the eye corner areas of women of varying ages. Methods One hundred and one healthy women (age range 20–80 years) residing in the Tokyo area were the subjects used in this study. Results Wrinkles demonstrated a rapid increase in depth in women aged 40 years or older, and plateaued at the age of 60 years. Surface morphology parameters yielded results similar to those of age-related changes in wrinkles. Conclusions This new analytical system provides a rapid and convenient non-invasive method to evaluate skin surface morphology in three dimensions, especially with respect to wrinkle formation. The results obtained using this system provide a deeper insight into the mechanistic relationship between wrinkles and skin elasticity.

    Notes: Summary Background The incidence of atopic dermatitis (AD) is increasing worldwide. No large-scale study has previously compared the therapeutic management of this condition in different countries. Objectives The purpose of this study was to determine the treatment preferences of dermatologists in Japan, the U.S.A. and the U.K., and investigate their relationship with certain factors pertaining to the physician and his practice. Methods A questionnaire was sent to all registered members of dermatological societies in Japan, the U.S.A. and the U.K. Responses were collated and statistical analysis performed using χ2, Mantel–Haenszel and Breslow heterogeneity tests. Results Three thousand six hundred and eighty-eight completed surveys were returned. U.S.A. and U.K. physicians were significantly more aggressive in prescribing systemic medications, such as steroids, antibiotics and immunosuppressants, compared with those in Japan. Japanese dermatologists also utilized topical steroids significantly less. The use of alternative remedies was highest in Japan. All three countries had a relatively high degree of optimism for topical immunosuppressants, but less so for other emerging therapies. Conclusions Both similarities and differences in the therapy of AD exist in the three countries studied. Factors related to the physician, patient population and culture may influence these observations.

    Notes: A 50-year-old woman with von Recklinghausen’s disease, but not Carney’s complex, presented with a 1-year history of a hard subcutaneous mass on her right hip and right inguinal lymphadenopathy. Histological and immunohistochemical studies of the tumour revealed schwannian and melanocytic characteristics. Local recurrence without distant metastases was observed 5 years later. Although the diagnosis of malignant schwannoma with melanocytic differentiation, rather than neurotropic melanoma, was made for the primary tumour, based on the clinicohistopathological and ultrastructural findings, the overall clinical course in this case did not seem incompatible with malignant melanocytic schwannoma.

    Notes: We have obtained optical second harmonic (SH) intensity images of a silver grating surface. A stripe pattern was seen in the SH intensity image when the incident light polarization is perpendicular to the lines of the grooves of the grating. We have found that the strongest SH radiation comes from the bottoms of the grooves. On the other hand, the strong linear reflection comes from the slope of the grooves. We suggest that the enhancement of the electric field at the bottoms of the grooves has led to the observed strong SH radiation. © 2002 American Institute of Physics.
