    Notes: Two molecular marker approaches [amplified fragment length polymorphism (AFLP) and simple sequence repeat (SSR)] were employed to study genomic relationship among 96 rice cultivars. These included most of the best reputed Italian accessions. AFLP produced 461 fragments, 248 (53%) of which were polymorphic, SSR produced four to 11 alleles in the 12 genomic loci investigated. Genomic similarity was estimated independently for the two molecular marker techniques. Both AFLP and SSR dendrograms agree in splitting the cultivars into two main clusters: a small one, comprising four exotic accessions, and a larger one which could be split into four subgroups. These were also analysed on the basis of historical and pedigree information. This is the first report on the application of DNA polymorphism analysis to reveal genomic relationship among cultivated Italian rice germplasm. Results will be useful for breeding programmes.

    Notes: Morphological traits traditionally adopted to discriminate between Populus alba L. and P. tremula L. have frequently led to misclassifi-cation of their spontaneous hybrid P. × canescens Sm. Moreover, they may not be of any help in cases of spontaneous backcross phenomena. These limitations can be overcome by molecular markers, which are not environmentally influenced nor subjectively assessed. In this study, the effectiveness of amplified fragment length polymorphism (AFLP) and simple sequence repeat (SSR) markers in species and hybrid discrimination was evaluated by analysing a set of reference samples of P. alba, P. tremula and P. × canescens. Species-specific and species-indicative AFLPs, as well as diagnostic SSR alleles, were recorded in both P. alba and P. tremula reference samples. The results allowed a clear distinction between the two poplar species and their hybrid. Using these diagnostic markers, a natural population of P. alba trees sampled along the Ticino river basin in northern Italy was analysed, and P. × canescens individuals, intermingled with P. alba trees, were detected.

    Notes: The sequence-specific amplification polymorphism (S-SAP) method, derived from the amplified fragment length polymorphism (AFLP) technique, produces amplified fragments containing retrotransposon long terminal repeat (LTR) sequence at one end and a host restriction site at the other. The development and application of this procedure to the LTR of the Vine-1 element from grapevine is reported. Two primers derived from one of the LTR sequences flanking the retrotransposon were used in combination with MseI degenerated primers on 15 grapevine accessions. S-SAP results were compared with AFLP data. The heterozygosity and gene diversity values were higher for S-SAP than for the AFLP procedure. Results show that S-SAP amplification is effective in identifying polymorphisms and defining genetic distances among cultivars, and could be used for fingerprinting and for ‘Traminer’ clone identification. To the contrary Vine-1 retrotransposon-based S-SAP was not able to distinguish ‘Pinot’ clones.

    Notes: The polarization dependence of the optical reflectivity for sapphyrin layers deposited by the Langmuir–Blodgett technique onto a gold substrate has been measured. The experimental results show that characteristic reflectance anisotropy spectroscopy (RAS) spectra are related to layers of different thicknesses. In order to interpret the measured spectral features, the anisotropy of the optical properties of the sapphyrin molecule has been evaluated by using a semi-empirical quantum chemistry approach. The results show clearly that the main RAS structures are related to the electronic properties of the sapphyrin molecules. In particular, two different regimes are observed. Below one (true) monolayer, the optical signal remains very low and structureless, in agreement with the sapphyrin molecules being stacked with their planes almost perpendicular to the substrate plane. Above one monolayer, instead, a strong RAS signal related to the Soret band develops, suggesting that sapphyrin molecules lay more parallel to the substrate surface, in an ordered fashion. © 2000 American Institute of Physics.
