    Notizen: The observed electronic line broadening of the X→B (6s Rydberg) absorption spectrum of CH3I vapor in moderate to high pressures (55–140 atm, reduced densities ∼0.08–0.14) of Ar and CH4 is analyzed via molecular dynamics simulations. Good fits to the absorption line shapes are found in this pressure/density range for a given set of ground and excited state solute–solvent potential parameters in the static limit, i.e., the absorption linewidths and shapes are dominated by inhomogeneous broadening on the time scale of the decay of the dipole correlation function. The pressure dependence of these absorption line shape changes is explained in terms of the shape of the solute–solvent ground-excited state difference potential. Consistent with the static limit description at these moderate to high pressures, the corresponding transition energy correlation function, a quantity of central importance in stochastic and Brownian oscillator line shape theories, decays on a much longer time scale than the inverse absorption widths. At moderate to high pressures, simulations find relatively long-lived solvent clusters surrounding the CH3I solute. The slow decay of the energy correlation functions, and hence the validity of the static approximation, is attributed to these cluster dynamical time scales. At bath pressures lower than observed here, MD simulations reveal that the static limit is no longer valid and satellite bands, due to an underdamped solvent response, are found. The evolution of a Gaussian optical line shape at higher densities is discussed with respect to the difference potential shape, the number density, and the central limit theorem. © 1995 American Institute of Physics.

    Notizen: Zygotic embryos of leek (Allium ampeloprasum L.) were isolated from mature seeds of different cultivars, selfings and full-sib families. The embryos were cultured on callus induction and shoot regeneration medium and employed to study several parameters: percentage of embryos forming calluses, percentage of embryos forming compact calluses, callus weight, percentage of regenerating calluses, numbers of shoot primordia and numbers of regenerated shoots. Differences between cultivars and selfings were found for most parameters studied. For all cultivars all parameters, except callus weight, decreased after one generation of selfing. Compact callus types enhanced primordia formation and shoot regeneration. Genetic characteristics of callus development and plant regeneration were studied in a 4 × 4 diallel cross. Analysis of variance revealed significant differences between full-sib families. The diallel analysis showed that additive gene effects were significant for all parameters. The predominance of additive gene effects indicated high narrow-sense heritability. Breeding for an increased number of regenerated shoots was successful.

    Notizen: Summary  The association between postoperative nerve root signs and outcome was investigated in 54 out of one hundred patients operated on by lumbar microdiscectomy in a prospective cohort study with one year follow up. The patients were classified as failures or successes at the 12 month follow up according to a clinical overall score. All the 14 failures were investigated with MRI at the one year follow up, and 40 patients classified as successes were picked at random for MRI. Three patients with signs of recurrent disc herniation on MRI were excluded from the study. The MRI scans were independently read by two neuroradiologists who were ignorant of treatment outcome. No association between nerve root thickening, nerve root enhancement or nerve root displacement and the clinical outcome was found when patients with recurrent disc herniation were excluded.

The MRI scans were rated according to the presence or absence of scar formation within the spinal canal and the degree of scar enhancement by two independent neuroradiologists ignorant of treatment outcome. No evidence of scar formation was found in 4 patients, a small amount in 11, intermediate in 38 and extensive scar tissue in 1 patient. Slightly enhancing scar tissue was found in 18 patients, intermediate in 27 patients and strong enhancement in 5 patients. No association between the amount or enhancement of peridural scar formation and clinical outcome was found. Both the total clinical score and the difference between pre- and post-operative clinical score were used in the calculations. Testing the different subsets in the clinical overall score, as well as the patient satisfaction VAS score, did not reveal any association.

    Notizen: Summary A new simple, efficient and rapid in vitro method for mass clonal propagation of leek (Allium ampeloprasum L.) plants, using small (5 mm) flower stalk (peduncle) explants, was established. Adventitious shoots were produced from single subepidermal cells. A wide variation in the percentage of regenerating explants and number of regenerated shoots per explant between individual plants within one cultivar was observed. The concentration of the growth regulators 6-benzylaminopurine and α-naphthalene-acetic acid influenced the percentage of regenerating explants and the average number of regenerated shoots per explant. A combination of 10 mg.l−1 6-benzylaminopurine and 10 mg.l−1 α-naphthalene-acetic acid, resulted in a maximum percentage of regenerating explants and a high average number of regenerated shoots per explant. The percentage of regenerating explants and the average number of regenerated shoots per explant decreased with increasing flower stalk length (age). The basal explants gave both the highest percentage of regenerating explants and average number of regenerated shoots per explant. An average of 300 shoots per flower stalk was obtained for all plants, making this new in vitro method a powerful tool in hybrid leek breeding.

    Notizen: Abstract A two-step protocol for the induction of shoots from Alstroemeria leaf explants has been developed. Leaf explants with stem node tissue attached were incubated on shoot induction medium for 10 days, and then transferred to regeneration medium. Shoots from the area adjacent to the region between the leaf base and node tissue regenerated within 3 weeks after transfer to the regeneration medium, without a callus phase. The best induction was obtained with Murashige and Skoog medium containing 10 µm thidiazuron and 0.5 µm indole butyric acid. The regeneration medium contained 2.2 µm 6-benzylaminopurine. After several subcultures of the leaf explants with induced shoots, normal plantlets with rhizome were formed. In Alstroemeria, the percentage of responding leaf explants is more important than the number of shoots regenerated per leaf explant, because rhizome formation is the most important factor for micropropagation. The effect of other compounds in the induction medium, including glucose, sucrose, silver nitrate, and ancymidol, on regeneration was also investigated.

    Notizen: Summary In leek (Allium ampeloprasum L.) a cyclic system of somatic embryogenesis was developed. Somatic embryos used for cyclic embryogenesis were able to develop the same type of embryogenic callus as zygotic embryos in the primary cycle. For the first time a comparison of the efficiencies of both expiants was made. Ten families were investigated for somatic embryogenesis. There was a genetic relationship with respect to somatic embryo production between the reciprocal crosses. From each family one genotype was selected for investigating cyclic somatic embryogenesis. Different levels of somatic embryo production were found between the expiants of zygotic and somatic embryos. The two best genotypes, 92.001-03 and 92.002-33 produced twice as many somatic embryos as the overall average. On average, 56% of the somatic embryos finally developed into greenhouse plantlets.

    Notizen: Summary. The plant regeneration ability of callus obtained from zygotic embryos of the monocot Alstroemeria spp. was studied. The best explants for somatic embryogenesis were immature zygotic embryos in half-ovules when the endosperm was still soft and white. For 2 genotypes embryogenic callus was induced on callus induction medium with a success rate of 54%. The best callus induction period was 10 weeks. The morphology of embryogenic callus was nodular. Somatic embryos were formed after transfer of the callus to regeneration medium. These somatic embryos revealed later on the typical features of zygotic Alstroemeria embryos. The total duration of the plant regeneration protocol, from inoculation till rooted plantlets ready for transfer to the greenhouse, was 28 weeks.

    Notizen: Abstract Central nervous system involvement was evaluated in 36 patients with systemic lupus erythematosus (SLE) using cerebral computed tomography (CT), electroencephalography (EEG), and a neuropsychological test battery. The purpose was to investigate whether brain dysfunction as assessed by comprehensive neuropsychological investigation is associated with findings of routine investigation methods such as CT and EEG which are available in most hospitals. Abnormal EEG was found in 19%, and CT revealed cerebral atrophy in 47% of SLE patients. Few neuropsychological functions were affected by the presence of abnormal EEG, cerebral atrophy, or infarcts. Significant associations were found only between cortical atrophy and impairment of tactile spatial problem-solving and motor dexterity, and between cortical infarcts and motor dexterity in the dominant hand. The value of conventional EEG in assessing cerebral SLE is negligible, except for identifying epileptic activity and focal pathology. Cerebral CT has little relevance in predicting brain dysfunction as established by neuropsychological assessment in SLE, except for detecting cortical atrophy and infarcts.
