    Notes: Pantoprazole is a benzimidazole derivative which selectively inhibits the proton pump H+, K+-ATPase necessary for the final step in gastric acid secretion.〈section xml:id="abs1-2"〉〈title type="main"〉Aim:To investigate the tolerability and the prophylactic effect of pantoprazole 40 mg once daily on relapse in patients whose reflux oesophagitis had been healed.〈section xml:id="abs1-3"〉〈title type="main"〉Methods:The safety of pantoprazole 40 mg once daily was assessed in an open 1-year trial on 222 patients whose reflux oesophagitis had been healed with omeprazole or pantoprazole. Relapse was defined as endoscopically-confirmed reflux oesophagitis (at least Grade I), with endoscopies being performed for patients experiencing 3 consecutive days of disease-specific symptoms.〈section xml:id="abs1-4"〉〈title type="main"〉Results:Kaplan–Meier survival analysis at 6 and 12 months gave estimated treatment failure rates of 2% and 6% from confirmed relapses (per-protocol), and of 9% and 30% for a worst-case group (all withdrawals counted as failures). The only population shift in laboratory variables was a doubling of the median serum gastrin level over the first 6 months; thereafter it stabilized. Fifty-four (24%) patients experienced adverse events; 15 of these withdrew. Serious adverse events were reported for 12 patients.〈section xml:id="abs1-5"〉〈title type="main"〉Conclusions:Pantoprazole appears to be highly effective and to have a good safety profile for long-term prophylaxis of reflux oesophagitis.

    Notes: Abstract A favourable combination of genetic features in the genus Oenothera offers access to fundamental biological aspects that are not readily approached with other materials. We have developed protocols for cell and tissue culture as well as for transformation, in order to establish the basis for a comprehensive cell and molecular biology of Euoenothera species, their genome/plastome hybrids and plastome mutants. Regeneration of plants from excised seedling parts (roots, hypocotyl, cotyledons, shoot tips) and leaf explants was optimal on NT medium containing 1 mg ⋅ l–1 6-benzylaminopurine and 3 mg ⋅ l–1 α-naphtalene acetic acid. This medium also proved to be efficient in the propagation of various wild-type genotypes, interspecific hybrids and plastome mutants. Using Ti-based approaches we also succeeded in generating transgenic Oenothera plants with relatively high efficiency.

    Notes: Abstract A protocol is presented for regenerating plants from leaf protoplasts of Oenothera. The method uses (1) embedding of isolated protoplasts at high cell densities in thin alginate layers, (2) initial culture in B5 medium containing 3 mg l–1 α-naphthaleneacetic acid (NAA) and 1 mg l-1 6-benzylaminopurine (BAP), (3) reduction of the osmotic pressure of the culture medium at early stages of culture and (4) plating of microcolonies recovered from the alginate onto solid B5 medium with 3 mg l–1 NAA and 1 mg l–1 BAP. The shortest time required from protoplast isolation to the appearance of shoot initials was 7 weeks. The efficiency of the procedure for protoplast to cell line formation is high (about 80%).

    Notes: Zum Thema Langzeitmessungen haben sich im oberen Verdauungstrakt für die pH-Metrie und für die Manometrie besonders bewährt. Sie dienen dem Studium physiologischer und pathophysiologischer Phänomene sowie der Überprüfung pharmakologischer Wirkungen, insbesondere säureblockierender Substanzen. Um mögliche Fehlinterpretationen von Untersuchungsergebnissen zu vermeiden ist es von großer Bedeutung, sich eindeutig über die Technik und die Methode der pH- und Manometrie zu informieren. Wie, wann und wo genau die Meßwerte erhoben und ob die Ergebnisse klinikintern und -extern reproduzierbar sind, auch der Einfluß und die Dokumentation exogener (z.B. Nahrungsaufnahme) und endogener Faktoren ist bei diesen Untersuchungsmethoden und den daraus folgenden Behandlungskonsequenzen außerordentlich wichtig. So werden in dieser Übersicht zunächst die technischen und methodischen Voraussetzungen eingehend dargelegt, des weiteren die Indikationen und Ergebnisse der pH- und Manometrie vorwiegend des oberen Gastrointestinaltrakts erläutert.

    Notes: Abstract Plastids contain a NAD(P)H-plastoquinone-oxidoreductase (NDH complex) which is homologous to the eubacterial and mitochondrial NADH-ubiquinone-oxidoreductase (complex I), but the metabolic function of the enzyme is unknown. The enzyme consists of at least eleven subunits (A-K), which are all encoded on the plastid chromosome. We have mutagenized ndhC and ndhJ by insertion, and ndhK and ndhA-I by deletion and insertion, of a cassette which carried a spectinomycin resistance gene as a marker. The transformation was carried out by the polyethylene glycol-mediated plastid transformation method. Southern analysis revealed that even after repeated regeneration cycles each of the four different types of transformants had retained 1–5% of wild-type gene copies. This suggests that complete deletion of ndh genes is not compatible with viability. The transformants displayed two characteristic phenotypes: (i) they lack the rapid rise in chlorophyll fluorescence in the dark after illumination with actinic light for 5 min; in the wild-type this dark-rise reflects a transient reduction of the plastoquinone pool by reduction equivalents generated in the stroma; and (ii) transformants with defects in the ndhC-K-J operon accumulate starch, indicating inefficient oxidation of glucose via glycolysis and the oxidative pentose phosphate pathway. Both observations support the theory of chlororespiration, which postulates that the NDH complex acts as a valve to remove excess reduction equivalents in the chloroplast.

    Notes: Die präoperative endoskopische Diagnostik kolorektaler Tumoren ist in den zurückliegenden Jahren grundlegend weiterentwickelt worden: Mit der weiten Verbreitung der Videoendoskopietechnik steht vielerorts ein diagnostisches Verfahren mit im Vergleich zur herkömmlichen Glasfaserendoskopie wesentlich höherer Detailauflösung zur Verfügung, das die Feindiagnostik tumoröser Schleimhautveränderungen auch im unteren Gastrointestinaltrakt verbessert. Neben anderen bildgebenden Verfahren wie konventionellem Ultraschall, Computertomographie, Magnetresonanztomographie kommt der Endosonographie zunehmend eine überlegene Bedeutung in der präoperativen Diagnostik, insbesondere bei der Stadienerhebung, zu. Besondere Bedeutung erreichen die nichtoperativen Methoden zum Staging und die zunehmend besseren Kenntnisse über die postoperative Prognose kolorektaler Tumoren durch die Etablierung endoskopischer Therapieverfahren: Mit ihnen werden die Tumormasse verkleinert und Stenosen beseitigt; verschiedene Thermokoagulationsverfahren (allen voran die Laserablation) sind anerkannt, erste Erfahrungsmitteilungen liegen über die Endoprothetik und die photodynamische Therapie im Kolorektum vor.

    Notes: Das kolorektale Karzinom ist das am dritthäufigsten diagnostizierte Karzinom. Die Häufigkeit nimmt in der westlichen Welt noch zu. Für ein asymptomatisches Individuum, das keiner Risikogruppe für die Erkrankung an diesem Tumor angehört, beträgt die Wahrscheinlichkeit während seines Lebens an einem kolorektalen Karzinom zu erkranken 6%. Die Lebenserwartung der an einem kolorektalen Karzinom erkrankten Patienten sinkt um 6–7 Jahre [3]. Durch Verbesserungen und Erfahrungen in Früherkennung und Nachsorge ist es möglich, das Karzinomrisiko zu senken bzw. rechtzeitig zu erkennen.

    Notes: Summary Regeneration of plants from protoplasts is regarded a difficult and lengthy procedure which requires well developed skills on the side of the experimenter. Therefore, where alternative procedures for genetic engineering of plants are available, protoplast-based techniques are frequently avoided. Here, we demonstrate, that by our newly developed thin-alginate-layer technique it is possible to regenerate shoots from leaf protoplasts ofNicotiana tabacum L. at very high efficiency and very rapidly, with the first shoots appearing within less than two weeks. Root formation is induced on a third medium with first roots being found after only 10 more days of culture.
