    Notes: Abstract This paper presents data on modelling of DNA damage induced by electrons, protons and alpha-particles to provide an insight into factors which determine the biological effectiveness of radiations of high and low linear energy transfer (LET). These data include the yield of single- and double-strand breaks (ssb, dsb) and base damage in a cellular environment. We obtain a ratio of 4–15 for ssb:dsb for solid and cellular DNA and a preliminary ratio of about 2 for base damage to strand breakage. Data are also given on specific characteristics of damage at the DNA level in the form of clustered damage of varying complexity, that challenge the repair processes and if not processed adequately could lead to the observed biological effects. It is shown that nearly 30% of dsb are of complex form for low-LET radiation, solely by virtue of additional breaks, rising to about 70% for high-LET radiation. Inclusion of base damage increases the complex proportion to about 60% and 90% for low- and high-LET radiation, respectively. The data show a twofold increase in frequencies of complex dsb from low-LET radiation when base damage is taken into account. It is shown that most ssb induced by high-LET radiation have associated base damages, and also a substantial proportion is induced by low-energy electrons.

    Notes: This paper describes a system for the study of the relation between structure and applied strain in thermoplastic polyurethane elastomers using the Australian National Beamline Facility at the Photon Factory, KEK, Tsukuba, Japan. The system uses the sagittal focusing monochromator at beamline 20B to provide a high-intensity focused beam which then falls on the specimen mounted in a miniature tensometer mounted in the unique vacuum diffractometer (BIGDIFF). Imaging plates were used to record simultaneously SAXS and WAXS patterns from the specimen at a particular strain. The change in SAXS and WAXS patterns with loading and unloading was recorded using a ten-plate imaging-plate changer.

    Notes: Two systems have been developed for the simultaneous recording of the SAXS and the WAXS patterns from carbon fibre and elastomer samples which are placed under stress. The systems have been designed to fit inside the versatile vacuum diffractometer (BIGDIFF) at the Photon Factory. In one system, use is made of the ability to move the imaging-plate cassette. In the other, use has been made of an imaging-plate changer which can deliver up to 13 plates into position with a duty cycle of about 60 s. In this case each imaging plate can record SAXS/WAXS patterns in the range 0.5–20° due to the passage of the beam through the specimen which is mounted in a specially designed tensometer. Because BIGDIFF is a vacuum diffractometer and parasitic scattering is small, exposure times as short as 2 s can give acceptable SAXS/WAXS patterns. The systems have been used for the study of both the change of structure with strain, and the relaxation processes which occur as a result of the sample being strained at a fixed rate by a predetermined amount.

    Notes: Abstract This report describes the development of a fluorescence microscope based on a standard inverted optical microscope which incorporates a pulsed picosecond dye laser excitation source and a detector consisting of a gated image intensifier coupled to a CCD camera. Fluorescence images have been obtained using gate durations of 0.5 ns from this apparatus, representing a reduction in gate duration of an order of magnitude compared with similar instruments reported by others recently. Subnanosecond gated fluorescence images of V79-4 Chinese hamster lung fibroblasts stained with a phthalocyanine photosensitizer used in photodynamic therapy are presented. The results of these measurements are discussed in terms of the intracellular distribution of the sensitizer. Other potential applications and limitations of this technique are also outlined.
