    Notes: Summary.  Haemophilia A is an X-linked inherited bleeding disorder. Linkage diagnosis using polymorphic markers in the factor VIII gene is used to archive the carrier detection and prenatal diagnosis. The objective of this study was to establish the allele frequency and heterozygosity rate (HR) of two new intragenic markers (Intron 1 and 24) and other markers (Intron 13 and 22) using fluorescent PCR. Five hundred unrelated healthy women were screened and haemophilic family was studied for carrier detection and prenatal diagnosis. We observed five different alleles of Intron 1, 10 of Intron 24, nine of Intron 13 and six of Intron 22. The observed HR for Intron 1, 24, 13 and 22 were 34.0, 35.2, 53.0 and 42.6%, while the expected HR were 33.6, 36.3, 50.1 and 44.3%, respectively. Heterozygosity rate with the combined use of all four intragenic markers was 76.6% (383/500). In prenatal diagnosis of a haemophilic family, a pregnant woman was heterozygous with three intragenic (Intron 1, 13 and 22) and one extragenic St14 VNTR (DXS52) markers. She was considered to be a carrier, and she carried a male foetus by AMXY PCR and chromosome analysis of amniocytes. Foetus did not have mutant haplotype as his uncle, suggesting a normal male status. Our study demonstrates the utility of two new intragenic markers in FVIII gene for carrier detection and prenatal diagnosis of haemophilic families.

    Notes: Freeze-thawing studies at different concentrations, using an actomyosin solution (extracted from Alaska pollock), revealed that an 8% (w/v) solution of oligosaccharides mixture (HBOS) was most effective in cryoprotection. During frozen storage (-18°C), HBOS showed cryoprotective effects similar to sucrose and a sucrose+sorbitol mixture (1:1). Surimi gel prepared with HBOS showed higher hardness and more dense microstructure than others, although water holding capacity was slightly lower than the gel with sucrose+sorbitol. HBOS containing gel showed lower whiteness than sucrose but no difference with sucrose+sorbitol. HBOS appeared to have good potential as a non-sweet cryoprotectant of fish protein.

    Notes: Background:  The genetic background of atopic dermatitis (AD) is not clearly understood. Interleukin (IL)-10 is a powerful Th-2 cell cytokine produced by lymphoid cells that exerts its function by inhibiting macrophage/monocyte and T-cell lymphocyte replication and secretion of inflammatory cytokines [IL-1, tumour necrosis factor-α (TNFA), IL-6, IL-8 and IL-12].Objective:  In an effort to discover additional polymorphism(s) in genes whose variant(s) have been implicated in total immunoglobulin E (IgE) level in AD patients, we scrutinized the single nucleotide polymorphisms (SNPs) in the IL10 gene as a potent candidate for contributing to the level of IgE in serum.Methods:  We recruited 334 AD patients and assayed their serum total IgE levels using the LIPA-200 system. Four SNPs in the IL10 gene were genotyped using the single-base extension (SBE) method. Logistic regression analyses were performed with single polymorphisms and haplotypes (ht) to determine their association with the level of serum total IgE.Results:  Genetic association analysis of total serum IgE in AD patients revealed that one of the IL10 ht, IL10-ht2, was associated with decreased serum total IgE in gene dose-dependent manner (P = 0.02–0.001).Conclusions:  It was predicted that the inhibition of innate immunity by increased IL-10 production in IL10-ht2-bearing individuals might be associated with decreased total serum IgE levels among AD patients. The greater effects of IL10 ht on decreased total serum IgE levels suggest that the effect of IL-10 polymorphism might be the result of a combined genotype (ht) rather than single polymorphisms.

    Notes: Abstract In this study, porosity and graphitizability of coal tar pitch with the treatment pressure were investigated. 4-directional carbon/carbon composites (4D C/C) were made from the matrix precursor of coal tar pitch through the process of impregnation and carbonization. Then the effects of applied pressure during the densification on the composites were observed. The matrix pitch which had 600 bar applied during the carbonization process had one and a half times less pore area ratio than that treated at 1 bar. When the pitch was heat treated up to 2300°C after the high pressure carbonization, the degree of graphitization was improved on a small scale and the crystal size tended to reduce. As the applied pressures to 4D C/C composites increased from 1 to 600 bar, the densification ratio was greatly improved. In the pore size distribution of the 4D C/C composites, the macropore portion was decreased while the mesopore portion increased, when high pressures were applied.

    Notes: We present contact-angle hysteresis and surface energy of differently treated indium–tin–oxide (ITO) thin films obtained from contact angles for liquids with different polar character. We find that the hysteresis and the polar and dispersion component of the surface energy depend strongly on the surface treatments. Oxygen-plasma treatments induce the highest polarity and the highest total surface energy, and we suggest that this improves the interface formation with polymers, and therefore, the performance of light-emitting diodes. We discuss the results in relation to the ITO surface roughness and chemical heterogeneity modified by the different treatments. © 1999 American Institute of Physics.

    Notes: A theoretical and computational study of the ion temperature gradient (ITG) and ηi instabilities in tokamak plasmas has been carried out. In a toroidal geometry the modes have a radially extended structure and their eigenfrequencies are constant over many rational surfaces that are coupled through toroidicity. These nonlocal properties of the ITG modes impose a strong constraint on the drift mode fluctuations and the associated transport, showing self-organized criticality. As any significant deviation away from marginal stability causes rapid temperature relaxation and intermittent bursts, the modes hover near marginality and exhibit strong kinetic characteristics. As a result of this, the temperature relaxation is self-similar and nonlocal, leading to radially increasing heat diffusivity. The nonlocal transport leads to Bohm-like diffusion scaling. Heat input regulates the deviation of the temperature gradient away from marginality. We present a critical gradient transport model that describes such a self-organized relaxed state. Some of the important aspects in tokamak transport like Bohm diffusion, near marginal stability, radially increasing fluctuation energy and heat diffusivity, intermittency of the wave excitation, and resilient tendency of the plasma profile can be described by this model, and these prominent features are found to belong to one physical category that originates from the radially extended nonlocal drift modes. The obtained transport properties and scalings are globally consistent with experimental observations of low confinement mode (L-mode) discharges. The nonlocal modes can be disintegrated into smaller radial islands by a poloidal shear flow, suggesting that the transport changes from Bohm-like to near gyro-Bohm. © 1996 American Institute of Physics.

    Notes: The linearized flux-surface-averaged resistive magnetohydrodynamic equations, which include the electron diamagnetic drift and hot particle effects, are derived and studied numerically. The effect of electron diamagnetic drift on the trapped hot particle-driven resistive interchange mode is then investigated. It is shown that the effect can have a strong stabilization effect on the mode, similar to the conventional resistive interchange mode case. © 1997 American Institute of Physics.
