    Notes: Abstract Objective: This study examined the use of dapsone N-hydroxylation and cortisol 6β-hydroxylation, well accepted in vivo probes of cytochrome P4503A4 (CYP3A4) activity, on defining the effect of three HIV protease inhibitors on CYP3A4 activity. Methods: Subjects from University Hospital Infectious Disease Clinic about to be started on indinavir, and subjects from two clinical studies, one using ritonavir and the other using amprenavir, were recruited to participate in the study. Subjects received dapsone 100 mg p.o. followed by an 8-h urine collection for dapsone, dapsone N-hydroxylamine, cortisol, and 6β-hydroxycortisol concentrations before HIV protease inhibitor administration, and 3–4 weeks into receiving HIV protease inhibitors. Results: None of the HIV protease inhibitors demonstrated statistically significant alterations in dapsone recovery ratio and 6β-hydroxycortisol/cortisol ratio. In fact, with ritonavir, the dapsone recovery ratio tended to increase rather than decrease, suggesting induction. These negative results were found despite evidence of CYP3A4 inhibition by these three HIV protease inhibitors via published drug-drug interactions with drugs that are substrates for CYP3A4. Conclusions: These in vivo assays used to probe CYP3A4 activity are suboptimal, most likely because of the presence of extrahepatic sites of metabolism for both dapsone and cortisol, and multiple CYP isozymes involved in dapsone N-hydroxylation.

    Notes: Abstract. Objective: The effect of age on the pharmacokinetics and pharmacodynamics of prazosin (α1 adrenoceptor blocker) was studied in 20 healthy volunteers. Patients: Ten elderly (61–81 y) and ten young (23–28 y) subjects were studied. All subjects received 1 mg of prazosin orally in a fasting state. Serial blood samples were collected for calculation of oral pharmacokinetics, and blood pressure and pulse rate were measured during blood collection. Subjects remained supine and fasting for the first three hours post drug administration, after which they were allowed to ambulate and eat. Results: The oral pharmacokinetics of prazosin were not different in the two age groups. The serum t1/2 in the elderly was 210 min while in the young group was 139 min. The AUC0−∞ in the two groups was not different. The Cmax was identical in the two groups, and the time to Cmax was 84 min in the elderly and 114 min in the young subjects. Protein binding was 93.4% in the elderly and 93.5% in the young subjects and the serum α1 acid glycoprotein concentration was not different in the two groups of subjects. Even though the pharmacokinetics of prazosin were unchanged by age, the haemodynamic effects of the drug were greater in the elderly. The fall in systolic blood pressure and mean blood pressure was significantly greater in the elderly group at multiple time points after drug administration while the change in diastolic blood pressure was equivalent in the two age groups. Despite a greater decrease in mean blood pressure in the elderly, the compensatory increase in heart rate was similar in the two age groups suggesting a difference in the baroreceptor reflex in the two age groups. Conclusion: The results of this study demonstrate that age does not alter the pharmacokinetics of oral prazosin, but the pharmacodynamic response at equivalent plasma prazosin concentration is greater in the elderly.

    Notes: The very high gains, achieved when using mode-locked pulses to synchronously pump an optical parametric oscillator based on periodically poled lithium niobate, permit oscillation even with strong idler absorption. Oscillation has been achieved at idler wavelengths out to 6.3 μm. Idler power was directly measured, out to 5.41 μm. Beyond this, idler generation was inferred from the observed signal. We present an analysis of operation in the presence of strong idler absorption. © 1998 American Institute of Physics.

    Notes: We report the use of a 5-mm-long planar Nd:yttrium aluminum garnet waveguide grown by liquid-phase epitaxy as an amplifier at 1.064 μm. In a double-pass configuration pumped by a 1.2 W diode laser, small signal gains of 686 (28 dB) have been observed and efficient power extraction demonstrated with output powers of up to 290 mW being obtained. © 1997 American Institute of Physics.

    Notes: We report a singly resonant optical parametric oscillator in periodically poled lithium niobate, synchronously pumped by the second harmonic of an amplified 10 μs pulse train from a continuous wave mode-locked Nd:YLF laser. Pulses of ∼2 ps duration have been generated over the tuning range from 883 to 1285 nm with a typical threshold of 200 mW average power within the 10 μs envelope. The M2 beam quality factor for the generated signal was ∼1.1, indicating absence of any significant photorefractive damage. © 1996 American Institute of Physics.

    Notes: It is shown that the photorefractive grating produced by a pair of plane waves in periodically poled lithium niobate includes an additional set of spatial harmonics related to the periodic domain structure. This results in new schemes for photorefractive wave coupling. Using the modified phase matching conditions and the concept of optical oscillation we accurately describe the position of the diffraction peaks and explain the main characteristics of self-organized photoinduced scattering reported recently. © 1996 American Institute of Physics.

    Notes: We report a quasi-phase-matched optical parametric oscillator (OPO) pumped by the second harmonic of a single frequency Q-switched Nd:YAG laser. Both the frequency doubling to 532 nm and the parametric oscillation are performed in periodically poled lithium niobate crystals with a nonlinearity of ∼15 pm/V. The OPO has been operated in "singly resonant'' and "doubly resonant'' configurations. The threshold in the singly resonant case was ∼0.14 J/cm2, more than one order of magnitude below the damage limit. OPO tuning from 945 to 1225 nm was achieved by changing both the period of domain reversal (from 6.8 to 6.85 μm) and the temperature of the crystal. © 1995 American Institute of Physics.

    Notes: A new technique for fabricating active planar waveguide devices is reported. This process, based on the thermal bonding of precision finished crystal or glass components, allows waveguides to be assembled from very dissimilar materials and could be applied to a wide range of solid state laser or other optical media. The waveguide propagation losses, inferred from the laser performance, are found to be 0.7 dB/cm for Nd:Y3Al5O12 bonded to Y3Al5O12, Nd:Y3Al5O12 bonded to glass, and 0.4 dB/cm for Nd:Gd3Ga5O12 bonded to Y3Al5O12 devices. © 1997 American Institute of Physics.

    Notes: A new species of self-organized light-induced scattering, appearing only in bulk periodically poled lithium niobate, is reported. Clearly defined diffracted beams develop on exposure to 532 nm light from a frequency-doubled, Q-switched, and mode-locked Nd:YAG laser. The structure thus created also diffracts light at 1064 nm. The effect is strongly dependent on temperature, the polarization state of the light, and the period of domain reversal. It exhibits a sharply defined nonlinear threshold at which the far-field pattern of the diffracted light evolves rapidly. © 1995 American Institute of Physics.

    Notes: Abstract A Pockels cell Q-switch has been operated in a manner analogous to a saturable absorber by initially keeping the cell partly open for the first 1000 or so double passes and then opening it completely at a pre-set resonator power. This then allows simple and reliable selection of a single longitudinal mode. A detailed discussion is given of the design considerations which lead to the correct choice of mode-selecting device. With three different devices, a TEM00 mode Nd:CaWO4 laser has produced 400 kW pulses in a single longitudinal mode with a shot-to-shot frequency stability of ∼ 0.01 cm−1.
