    Notizen: Abstract  Nitric oxide (NO) has been introduced recently for studying alveolar-capillary gas transfer. Due to extremely fast reaction kinetics for the association of NO with haemoglobin, pulmonary NO uptake is expected to depend only on diffusion, whereas in the case of carbon monoxide (CO) or oxygen-labelled carbon dioxide (C18O2) the alveolar-capillary transfer is, in addition, known to depend on a blood uptake component. To provide further data for NO, CO and C18O2, we determined the pulmonary diffusing capacities (D L) for the indicator gases mentioned, performing single-breath manoeuvres on ten rabbits. The inspired gas mixtures contained 0.05% NO and/or 0.2% CO or 1% C18O2 in nitrogen. Applying respiratory mass spectrometry to the expirates we obtained the following mean ± SD values: D L,NO/D L,CO = 3.55 ± 0.4, D L,C18O2/D L,NO = 6.0 ± 0.6, D L,C18O2/D L,CO = 21.4 ± 2.5. Graham’s law predicts D L ratios of 1.9 for NO/CO, 12 for C18O2/NO, and 23 for C18O2/CO. Thus we equally underestimated the predicted D L ratios for C18O2/NO and CO/NO by a factor of approximately 0.5. From this, and by excluding significant interactions between the indicator gases and lung tissues, we conclude that the closest approximation of the diffusive component of D L is indeed obtained by using NO.

    Notizen: Abstract  In this study we investigated the contribution of diffusion limitation to the exercise-induced hypoxaemia in interstitial lung disease (ILD). We applied isotopic analysis to the composition of the stable isotopic oxygen molecules 16O2 and 16O18O in expiratory gas mixtures obtained from six ILD patients and six healthy subjects at rest and during ergometer work (60 W). The changes in the 16O18O/16O2 ratios were interpreted by using the overall fractionation factor of respiration (α O) which would be increased towards 1.03 on increasing diffusion limitation. In addition, the O2 partial pressures of alveolar gas and arterial blood (P AO2, P aO2) were determined. In the patients, α O was significantly reduced from 1.0066 ± 0.0004 (mean ± SD) at rest to 1.0035 ± 0.0004 during exercise and in the healthy subjects from 1.0072 ± 0.0008 to 1.0044 ± 0.0004. Furthermore, the exercise-induced reduction of P aO2 (from 77 to 69 mmHg) was due to a drop of alveolar PO2 found in each patient, whereas in each healthy subject P aO2 was increased on exercise. On the basis of a resistance model we conclude that the patients’ data were inconsistent with increasing diffusion limitation but showed an increasing impairment of O2 transport by ventilation.

    Notizen: Abstract  To evaluate whether nitric oxide (NO) is an appropriate test gas for assessing pulmonary gas exchange, we determined the rates of disappearance from the alveolar space (λ) of NO and singly and doubly 18O-labelled carbon dioxide (C16O18O, C18O2) by performing single-breath manoeuvres on seven artificially ventilated rabbits. By exploiting unique features of both isotopic species and by analysing pulmonary gas transport and λ values with a commonly used model, we found that diffusion forms 98±6% (mean ± SD) of the overall resistance to alveolar-capillary NO transfer. This means that measurements of pulmonary NO uptake reveal the entire diffusive properties of the alveolar-capillary membrane, because the extremely fast binding of NO to haemoglobin negates the ”reactive” component within red blood cells of pulmonary capillaries.

    Notizen: Abstract Heat shock protein 70 (HSP70) gene expression was studied in a seasonal hibernator, the diurnal ground squirrel, Spermophilus lateralis. RNA transcripts of 2.7 and 2.9 kb hybridizing to an HSP70 cDNA were expressed in both brain and peripheral tissues of pre-hibernation euthermic animals; higher levels of expression were observed during the day than during nighttime samples. A decline in the expression of both transcripts occurred in all tissues examined during hibernation that remained low throughout the hibernation season, including the interbout euthermic periods and regardless of time of day. Quantitative comparisons showed pre-hibernation nighttime HSP70 expression to be as low as that observed during hibernation, despite the drastic increase in metabolic state and nearly 30°C difference in body temperature. In contrast to HSP70, some mRNAs, such as β-actin and HSP60, remained relatively constant, while others, such as glyceraldehyde 3-phosphate dehydrogenase, increased in specific tissues during the hibernation season. These results indicate that the expression of a highly conserved gene involved in protection from cellular stress, HSP70, can vary with an animal's arousal state.

    Notizen: Within molecular dynamics simulations of protein-solvent systems the exact evaluation of long-range Coulomb interactions is computationally demanding and becomes prohibitive for large systems. Conventional truncation methods circumvent that computational problem, but are hampered by serious artifacts concerning structure and dynamics of the simulated systems. To avoid these artifacts we have developed an efficient and yet sufficiently accurate approximation scheme which combines the structure-adapted multipole method (SAMM) [C. Niedermeier and P. Tavan, J. Chem. Phys., 101, 734 (1994)] with a multiple-time-step method. The computational effort for MD simulations required within our fast multiple-time-step structure-adapted multipole method (FAMUSAMM) scales linearly with the number of particles. For a system with 36,000 atoms we achieve a computational speed-up by a factor of 60 as compared with the exact evaluation of the Coulomb forces. Extended test simulations show that the applied approximations do not seriously affect structural or dynamical properties of the simulated systems.   © 1997 John Wiley & Sons, Inc.   J Comput Chem 18: 1729-1749, 1997
