    Notes: Abstract  Micturition in the decerebrate cat is characterized by a coordinated bladder contraction and a simultaneous decrease in external urethral sphincter (EUS) efferent activity. Without the suppression of EUS activity, voiding is significantly impaired, resulting in a state sometimes referred to as bladder-sphincter dyssynergia. The aim of the present study was to determine whether glycinergic inhibition contributes to the suppression of EUS activity during micturition evoked by bladder distension or electrical stimulation of the pontine micturition center (PMC) in decerebrate cats. Using subconvulsive intravenous doses of strychnine (0.1–0.24 mg/kg), we examined changes in bladder and EUS electroneurographic (ENG) activity during micturition. Following subconvulsive doses of strychnine, tonic EUS ENG activity increased during bladder filling in five of six animals. In the presence of strychnine, it was possible to evoke reflex bladder contractions of similar duration and peak pressure to those observed before strychnine administration. However, there was an absence of suppression of EUS ENG activity during the bladder contractions in all the animals. To determine whether the changes in sphincter activity could be due to strychnine acting at glycine receptors on EUS motoneurons, sacral spinal tissue was processed for a structural protein (gephyrin) associated with the glycine receptor. Motoneurons in Onuf′s nucleus in S1 were identified using choline acetyltransferase immunohistochemistry and subsequently processed with a gephyrin monoclonal antibody. Abundant gephyrin labeling was evident throughout Onuf′s nucleus. Since Onuf′s nucleus is made up of EUS and other motoneuron populations, a sample of antidromically identified urethral and anal sphincter motoneurons were intracellularly labeled with tetramethylrhodamine dextran (TMR-D) and then processed with the gephyrin antibody. Using dual-beam confocal microscopy, gephyrin immunoreactivity was observed on the soma and proximal processes of individual EUS motoneurons in both male and female animals. It was concluded that a strychnine-sensitive mechanism contributes to the suppression of sphincter activity normally observed during voiding. Although glycinergic inhibition may affect several components of the circuitry responsible for micturition, it appears that the suppression of EUS motoneurons during micturition may be partly due to a direct glycinergic inhibition of the EUS motoneurons.

    Notes: New crystals of the signal-transducing protein PII have been obtained in the presence of a number of different effector ligands. Various crystal forms are observed depending on the nature of the ligand(s). Co-crystallization with 2-ketoglutarate, glutamate and pyrophosphate produces hexagonal crystals similar to the wild type, ATP yields cubic crystals and ATP in conjunction with 2-ketoglutarate or glutamate yields orthorhombic crystal forms. All of the above crystals have been characterized by X-ray diffraction analysis. The hexagonal crystals belong to space group P63, cubic crystals to either I23 or I213 and orthorhombic crystals to I222. A molecular-replacement solution for the PII/ATP/2-ketoglutarate crystals has been obtained giving us an initial model for a trimer in the orthorhombic crystal form.

    Notes: The structure of the bacterial signal transduction protein PII has been refined to an R factor of 13.2% using 3σ data between 10 and 1.9 Å. The crystals exhibited twinning by merohedry and X-ray intensities were corrected using the method of Fisher & Sweet [Fisher & Sweet (1980). Acta Cryst. A36, 755–760] prior to refinement. Our earlier 2.7 Å structure [Cheah, Carr, Suffolk, Vasudevan, Dixon & Ollis (1994). Structure, 2, 981–990] served as a starting model. PII is a trimeric molecule, each subunit has a mass of 12.4 kDa and contains 112 amino-acid residues. The refined model includes all 1065 protein atoms per subunit plus 312 water molecules. The high-resolution refinement confirms the correctness of our 2.7 Å model, although it leads to a redefinition of the extent of various secondary-structural elements. The monomeric structure of PII exhibits an interlocking double βαβ fold. This is a stable fold found in a number of proteins with diverse functions. The association of the protein into a trimer leads to a new structure which we describe in detail. The effects of crystal packing forces are discussed and potential interaction sites with other proteins and effector molecules are identified.

    Notes: To assess large-whale stocks following the cessation of land-based South Georgia whaling in 1965, we report three independent sighting databases: a cruise in 1997, observations from Bird Island (NW of South Georgia) between 1979 and 1998, and mariner sightings between 1992 and 1997. All species were rare, with sightings of southern right whales being the most common event. Two right whales photographed off South Georgia matched animals known from Peninsula Valdés, Argentina, a population known to be growing at 7%per annum. In contrast, blue and fin whales appeared to be less abundant. A single blue whale mother-calf pair was observed off the Shag Rocks in February 1997. Extirpation of animals from this particular feeding ground is the most likely reason for ongoing low numbers of all species. Other factors may include competition for krill by traditional predators such as penguins and seals and more recently by humans, an unusually high rate of natural mortality, habitat change such as alteration in sea ice coverage, and/or the impact of ongoing whaling. The history of this critical area of large-whale habitat and this report demonstrate the need for improved, consistent longterm monitoring of population trends for these depleted stocks.

    Notes: Experiments are described to show some of the potential of the synchrotron radiation Laue method for the study of structural change within single crystals. In the metastable tetragonal crystals of P4N4Cl8 the eight-membered P4N4 ring is in a boat conformation, with symmetry {\bar 4}. On heating to ca 340 K the crystals transform, slowly to a second tetragonal form in which the ring conformation is a chair, its symmetry {\bar 1}. Both structures are known [Hazekamp, Migchelsen & Vos (1962). Acta Cryst. 15, 539–543; Wagner & Vos (1968). Acta Cryst. B24, 707–713]. In the transformation the molecular packing, unit-cell dimensions and crystal quality remain almost unchanged. To study this transformation, series of Laue diffraction patterns were recorded at 2–3 min intervals over a period of 30–40 min, while the temperature was raised to 373 K. For two series, reflection intensities were measured and they allowed determination and refinement of the fraction of boat and chair molecules present in a mixed boat/chair model of the structure. No significant change in the crystal occurs below ca 340 K; at or above 340 K, 40–50% of the molecules are converted from boat to chair conformations within 5 min, but the remainder of the conversion is much slower, even when the temperature is raised towards 370 K.

    Description / Table of Contents: Zusammenfassung Ungewöhnlich große idiomorphe Trapezoeder, die aus einzelnen Kristallen von Analcim oder aus Aggregaten von Pumpellyit bestehen, kommen in drei stratigraphisch benachbarten phonolitischen Lagen von Aghda vor. Die Trapezoeder enthalten konzentrisch verteilte Einschlüsse von Plagioklas, Pyroxen, Titanomagnetit und Apatit, die im Analcim frisch, im Pumpellyit jedoch intensiv umgewandelt sind. Die Analcim-Kristalle sind von bemerkenswerter Homogenität in ihrer Zusammensetzung und werden auf pseudomorphe Verdrängung von primärem Leucit durch Ionenaustausch zurück-geführt. Im unteren Phonolit ist der Analcim jedoch instabil geworden, und wurde durch Pumpellyit ersetzt, der ein weites Spektrum von Zusammensetzungen erkennen läßt und auf variierende Zufuhr von Fe, Mg, Ca, and Al vom Vorläuferanalcim und auf Umwandlung von Einschlüssen hinweist. Das Vorkommen von Analcim und Pumpellyit bei Aghda ist ein Hinweis auf Zeolithfazies-Bedingungen mit niedrigen fCO 2 und H2O und hohem Pfluid.
    Notes: Summary Spectacular large (3 cm) euhedral trapezohedra composed of single crystals of analcime or aggregates of pumpellyite occur in three stratigraphically adjacent phonolitic lavas from Aghda. The trapezohedra contain concentrically arranged inclusions of plagioclase, pyroxene, titanomagnetite and apatite which are fresh in the analcime but extensively altered in the pumpellyite. The analcime crystals are remarkably homogeneous in composition and are interpreted as having formed by ion-exchange pseudomorphous replacement of primary leucite. In the lower phonolite, however, the analcime became unstable and was replaced by pumpellyite which has a wide compositional range, reflecting variable input of Fe, Mg, Ca and Al from the precursor analcime and alteration of inclusions. The occurrence of analcime and pumpellyite at Aghda is indicative of zeolite facies conditions with low fCO 2and H2O activity and probably high Pfluid.

    Description / Table of Contents: Zusammenfassung Die spät-permische Schoschonit Provinz des südlichen Sidney Beckens besteht aus Laven und Intrusionen, die in einem 140 km langen Gürtel längs der Küste auftreten. Geophysikalische und sedimentologische Daten weisen darauf hin, daß diese Provinz ursprünglich eine Länge von mindestens 340 km hatte. Sowohl petrographische wie geochemische Daten weisen auf schoschonitische Affinität dieser spät-permischen Gesteine hin. Obwohl die SiO2 Gehalte von 48.4 bis 60.6% schwanken, haben alle Einheiten mit Ausnahme einer andesitischen Lava 〈 55%. Auf Subduktion weisen u.a. Anreicherungen an Al2O3 und LILE (Rb, K and LREE), die Verarmung an HFSE (Nb, Ta, Zr, Hf and Ti) niedrige Nb/U und Sr/Nd von ungefähr 30–35 hin. Die87Sr/86Sr Initiale, berechnet für 250 Ma, liegen zwischen 0.70294 und 0.70440 und dieɛ Nd-Werte zwischen +5,11 und +2,14 und fallen fast ausschließlich in den Mantel-Bereich. Niedrige MgO und mg-Zahlen (Maximal 7,37% bzw. 64,5) zeigen, daß die Schoschonite nicht primäre Magmen, die mit Mantelperidotiten im Gleichgewicht sind, darstellen. Isotopendaten und Elementgehalte weisen nicht auf ein Modell hin, bei dem Krusten-Kontamination bei der Magmen-Genese eine wichtige Rolle spielte, aber eine Nichtkontamination mit Material, das niedrige87Sr/86Sr Werte und hoheɛ Nd Werte hat, ist nicht auszuschließen. Eine zeitliche Beziehung zwischen den magmatischen Gesteinen des südlichen Sydney-Beckens, dem Dampier Ridge, Neuengland und Tasmanien weist auf ein weit verbreitetetes magmatisches Ereignis hin, das zumindest teilweise schoschonitischen Charakter hatte und das auf Subduktion längs der Ostküste von Gondwanaland zurückführbar ist. Die geochemische Ähnlichkeit von Proben des Dampier Ridge und des Sydney Beckens weist nicht auf räumliche Variationen in der Zusammensetzung über einen subduktionsbezogenen magmatischen Gürtel hin, sondern läßt eher, zusammen mit den Variationen der Isotopendaten, heterogene Quellen für diese Gesteine vermuten.
    Notes: Summary The Late Permian shoshonitic province of the southern Sydney Basin consists of lavas and intrusions confined to a 140 km long coastal belt, but geophysical and sedimentological data indicate that the province once had a length of at least 340 km. Both petrographic and geochemical data indicate shoshonitic affinities for these Late Permian rocks and, although the compositional range is from 48.4 to 60.6% SiO2, all units except an andesite lava have 〈55% SiO2. Subduction-related attributes of the province include enrichment in Al2O3 and LILE (Rb, K and LREE), depletion in HFSE (Nb, Ta, Zr, Hf and Ti), low Nb/U, and Sr/Nd of ∼ 30–35. Initial (at 250 Ma)87Sr/86Sr ranges from 0.70294 to 0.70440 whereasɛ Nd values range from +5.11 to +2.14 and plot almost exclusively in the mantle array. Low MgO and mg-numbers (maximum 7.37% and 64.5, respectively) demonstrate that none of the shoshonites represent primary magmas in equilibrium with mantle peridotite. Isotopic data and elemental contents are not supportive of a model in which crustal contamination is the dominant process in magma petrogenesis, but do not exclude contamination with Sr-rich material having a low87Sr/86Sr and highɛ Nd values. A temporal correlation between igneous rocks from the southern Sydney Basin, Dampier Ridge, New England and Tasmania indicates a widespread magmatic event which was, at least in part, shoshonitic in character and which developed in response to subduction along the east coast of Gondwanaland. The geochemical similarity between the Dampier Ridge and Sydney Basin samples does not support a model for spatial compositional variation across a subduction-related magmatic belt but, coupled with the variations in isotopic ratios, implies heterogeneity in the source.

    Notes: Summary The pore structures of zirconium oxide particles prepared by two different methods (PICA and Oil Emulsion processes) are compared. Nitrogen sorptometry and size exclusion chromatography are used to characterize the two different types of particles. Significant and unexpected differences were found in the accessible pore volumes and the bed packing densities of the two materials. The PICA material was found to have a higher totally included and totally excluded volume than would be normally expected. The chromatographic method provides results which are more useful for the application of these materials to separation sciences.
