    Notes: Abstract The methods for the determination of protein requirements are reviewed and the difficulties in achieving the recommendations of the dietary management of phenylketonuria proposed by a Medical Research Council Working Party on Phenylketonuria using currently available low phenylalanine (Phe) protein substitutes and low protein foods are examined. These recommendations are that all infants whose blood Phe concentrations exceed 600 μmol/l in the presence of a normal or low plasma tyrosine and an otherwise normal plasma amino acid profile while receiving a normal protein intake (2–3 g/kg/day), should start a low Phe diet immediately. Infants whose blood Phe concentrations remain persistently between 400 and 600 μmol/l for more than a few days should also start treatment. The diet should contain a protein substitute which is Phe free (or at least very low in Phe) and otherwise nutritionally complete with a composition sufficient to provide 100–120 mg/kg per day of tyrosine and a total amino acid intake of at least 3 g/kg per day in children under 2 years of age. In children over 2 years the intake of amino acids should be maintained at a level of 2 g/kg per day. The protein substitute should be spread as evenly as possible through the 24 h. Blood Phe concentrations should be maintained between 120 and 360 μmol/l. In children aged over 10 years it is suggested that the protein substitute should supply the protein reference nutrient intake + 50%. An upper blood Phe limit of 480 μmol/l rather than 360 μmol/l may be acceptable in school age children. Adults and adolescents should continue treatment with the aim to maintain blood Phe concentrations no higher than 700 μmol/l. During the period before conception and during pregnancy women should aim to have plasma Phe concentrations between 60–250 μmol/l.

    Notes: Mycobacterium avium is an intracellular pathogen that is associated with disseminated infection in acquired immunodeficiency syndrome (AIDS) patients. Human monocyte-derived macrophages were infected with M. avium strain 101 and a quinolone (Bay y 3118) was used at 8 μg ml−1, a concentration that kills growing bacteria but fails to eliminate static organisms. Infected monolayers were treated with Bay y 3118 for 4 days and viable bacteria obtained from the lysis of macrophages were used to infect other macrophages without passage in media. The procedure was repeated five times, after which seven different subpopulations that failed to grow within macrophages were identified. While the DNA fingerprinting confirmed that all came from the same strain, three protein profiles were observed. Static subpopulations were not killed by cytokine-stimulated macrophages, in contrast to the replicating subpopulation. Three of the static subpopulation strains were shown to be auxotrophic for glutamic acid or methionine. All seven non-duplicating subpopulation strains grew well in complete 7H10 agar. The importance of a static subpopulation of M. avium within macrophages is presently unknown. It is possible, however, that the non-growing bacteria would persist within macrophages.

    Notes: The use of water-cooling channels in silicon x-ray monochromators for the Cornell High Energy Synchrotron Source (CHESS) high power wiggler beamlines has been studied by finite element analysis. The efficiency from channels of different dimensions, ranging from 25 mm to 2 mm width and 5 mm depth, has been calculated. The new crystals are designed to replace the indirect cooled monochromators currently used at CHESS wiggler stations. At typical operation parameters of 150 mA electron current at 5.3 GeV and a gap of 40 mm, the 24-pole wiggler at CHESS provides an x-ray beam with a total power of 2.7 kW at the monochromator. Procedures have been developed for fabrication of internally cooled crystals using a silver-glass dye attach paste. Tests of a new crystal with a conventional x-ray source revealed very small amounts of residual strain. Experiments with synchrotron radiation are scheduled in the near future. © 1996 American Institute of Physics.

    Notes: Abstract Various studies on indoor and outdoor particulate matter in the urban environment in the vicinity of busy arterial roads in the centre of the subtropical city of Brisbane have indicated that the revised United States Environmental Protection Agency National Ambient Air Quality Standards (US EPA NAAQS) for Particulate matter PM2.5 could be exceeded not only outdoors but also indoors. The aim of this work was to investigate outdoor exposure to submicrometer particles and their relationship with indoor exposure in a hypothetical office building located in the vicinity of a busy arterial road. The outdoor exposure values and trends were measured in terms of particle number in the submicrometer size range and were then recalculated to represent mass concentration trends. The results of this study indicate that exposure to PM0.7 particles in ambient air close to a busy road often exceeds the levels of the annual and 24-hour US EPA NAAQS PM2.5 standards. It is likely that exposure to PM2.5 is even higher, and may significantly exceed these standards.

    Notes: Summary Minimum capillary lengths on commercial instruments are fixed and cannot be decreased further. To effectively reduce the capillary length used for separation the sample can be injected from the end of the capillary nearest the detector. This procedure is known as a ‘short-end’ injection and can reduces analysis times by at least two-thirds compared to conventional injections. The time reduction benefits are shown in rapid separations of basic drugs, drug-related impurities and chiral compounds. Short-end injections, in combination with both increased electrolyte strength and reduced voltage are an effective approach to reducing the detrimental impact of high sample solution ionic strength. They can also lead to improved resolution by increasing stacking effects and reducing peak tailing. Peak area and migration time precision obtained are shown to be equivalent to those obtained for conventional injection procedures. It is concluded that short-end injections should be considered for routine operation as they are a useful means of reducing analysis time, increasing sensitivity, decreasing buffer depletion effects. They also allow use of higher electrolyte strengths which can improve resolution and reduce peak tailing, and can overcome significant problems which occur when analysing samples containing high salt contents.

    Notes: Summary A capillary electrophoresis method is described for the novel application of quantifying levels of the simple organic acid counter-ions of a variety of basic drugs. These counter-ions are organic acids such as succinic and maleic. The method uses indirect UV detection and an electroosmotic flow modifier. Acceptable precision and detector linearity were obtained using internal standards. Method validation was completed and acceptable data was generated. The method is now in routine use for this type of testing.

    Notes: Abstract The ability of living organisms to survive on the smaller bodies in our solar system is examined. The three most significant sterilizing effects include ionizing radiation, prolonged extreme vacuum, and relentless thermal inactivation. Each could be effectively lethal, and even more so in combination, if organisms at some time resided in the surfaces of airless small bodies located near or in the inner solar system. Deep within volatile-rich bodies, certain environments theoretically might provide protection of dormant organisms against these sterilizing factors. Sterility of surface materials to tens or hundreds of centimeters of depth appears inevitable, and to greater depths for bodies which have resided for long periods sunward of about 2 A.U.
